This part of ISO 10272 describes a horizontal method for the semi-quantitative determination of
Campylobacter spp. It is applicable to products intended for human consumption or for the feeding of animals, and to environmental samples in the area of food production and food handling. However, it is possible that this part of ISO 10272 is not appropriate in every detail for certain products, deviations from it being made necessary for technical reasons. It is possible that this part of ISO 10272 is not applicable at all to some other products.

Ta del ISO 10272 opisuje horizontalno metodo za semikvantitativno metodo določevanja bakterij Campylobacter spp. Velja za proizvode, namenjene za človeško porabo ali za krmljenje živali ter za okoljske vzorce v območjih proizvodnje hrane in ravnanja z hrano. Vendar je možno, da ta del ISO 10272 ni v vseh podrobnostih primeren za določene proizvode, zato so odstopanja zaradi tehničnih razlogov potrebna. Možno je, da ta del ISO 10272 sploh ne velja za nekatere druge proizvode.

This document specifies a method for the detection of celery (Apium graveolens) in emulsion-type sausages (e.g. Frankfurter, Wiener).
Real-time PCR (polymerase chain reaction) detection of celery is based on an 101 bp (base pair) sequence from the gene of the mannitol dehydrogenase (GenBank Acc. No. AF067082 ) of celery (Apium graveolens).
The method has been validated on emulsion-type sausages (Bavarian “Leberkäse”) spiked with celery. For this purpose meat batter containing mass fractions of 50 % pork meat, 25 % pork fat, 23 % crushed ice and 1,8 % of a mixture of sodium chloride, nitrite, nitrate, phosphates and ascorbates was prepared according to a standard procedure for emulsion-type sausage. The meat batter was spiked with either ground celery seeds or celery root powder to 1000 mg/kg. Lower spiking levels were obtained by diluting with celery-free meat batter. The batter was stuffed into casings and heated at 65 °C for 60 min [1].

Foodstuffs - Determination of vitamin B6 by microbiological assay

This European Standard specifies a method for the determination of total vitamin B6 in foodstuffs by microbiological assay (MBA). Vitamin B6 is determined as the mass fraction of pyridoxine, pyridoxal and pyridoxamine, including their phosphorylated or glycosylated derivatives. It is usually expressed as milligram vitamin B6 per 100 g of foodstuff. The method is applicable to samples that can be rendered homogeneous and do not contain high concentrations of antibiotics or other interfering substances. This method has been validated in an inter-laboratory test on fortified and non-fortified samples such as wholemeal flour, milk powder, mixed vegetables and pigs liver at levels from 0,5 mg/100 g to 1,9 mg/100 g. Foodstuffs - Microbiological screening for irradiated food using LAL/GNB procedures

This European Standard specifies a microbiological screening method comprising two procedures, which are carried out in parallel. It permits the identification of an unusual microbiological profile in poultry meat. The presence of a large excess population of dead micro-organisms can under certain circumstances be presumptive of irradiation treatment, which means, that the results of the procedure of the determination of endotoxin concentration in the test sample using the Limulus amoebocyte lysate (LAL) test and of the procedure of the enumeration of total Gram negative bacteria (GNB) in the test sample are not radiation specific. Therefore, it is recommended that a positive result be confirmed using a standardized reference method for the detection of irradiated food, e.g. EN 1784, EN 1785 or EN 1786 [1] to [3].
This screening method has been successfully tested by inter-laboratory trials [4], [5], [6] and the procedure is generally applicable to whole or parts of poultry, e.g. breast, legs, wings of fresh, chilled or frozen carcasses with or without skin.
The method can also provide information about the microbiological quality of a product prior to irradiation.

The DEFT/APC technique is not radiation specific, therefore, it is recommended to confirm positive results using a standardised method (e.g. EN 1788, prEN 13751) to specifically prove an irradiation treatment of the suspected food.
The method has been successfully tested in interlaboratory tests with herbs and spices [1] to [5].
