Overview

EN 16772:2016 - Water quality: Guidance on methods for sampling invertebrates in the hyporheic zone of rivers (CEN) - provides practical, field-focused guidance for collecting and assessing invertebrate fauna living within the saturated sediments beneath and alongside wadable river channels (the hyporheic zone). The standard describes sampling equipment and field procedures, advises on developing a sampling strategy, and helps users select appropriate survey techniques for specific study objectives. It complements, but does not replace, standards for benthic macroinvertebrate sampling.

Key topics and requirements

  • Scope and definitions: Clear definitions for hyporheic terms (hyporheic zone, hyporheos, benthos, meiofauna, macroinvertebrate) and the standard’s focus on wadable rivers.
  • Survey objectives: Emphasises defining aims before method selection; suitability of methods depends on whether the study targets diversity, abundance, biomass, or impact assessments.
  • Sampling strategy: Guidance on designing spatial and temporal sampling (reach selection, substrate types, instream vs riparian locations) and on quantitative vs semi‑quantitative approaches.
  • Detailed sampling methods: Descriptions and field use of techniques and equipment, including (as covered in the standard):
    • Karaman‑Chappuis pit
    • Bou‑Rouch pump
    • Vacuum pump
    • Standpipe trap
    • Williams corer
    • Colonization devices
    • Freeze coring Each method includes notes on species typically sampled and relevant environmental variables.
  • Sample processing: Field and laboratory handling recommendations to ensure data quality.
  • Safety and adaptability: Emphasises health & safety compliance and that methods may need modification for local conditions.

Practical applications

  • River monitoring and ecological status assessments under water‑quality programmes
  • Catchment management, river restoration and impact studies (pollution, habitat alteration, fish spawning nursery evaluation)
  • Scientific research into hyporheic biodiversity, biogeochemical processes and surface–groundwater interactions
  • Consultants and environmental contractors designing targeted invertebrate sampling campaigns

Who should use it

  • Environmental agencies and regulators
  • Hydrogeologists and stream ecologists
  • Water quality scientists and biodiversity researchers
  • Environmental consultants and field survey teams

Related standards and policy links

  • Complements standards for benthic macroinvertebrate sampling (see Bibliography)
  • Relevant EU legislation referenced in the standard: Habitats Directive, Water Framework Directive, Groundwater Directive, Nitrates Directive

Sampling site location may be influenced by pre-existing monitoring networks or previous investigations. The following should be considered: — sampling method; — number and location of sampling sites; — number of replicates per site required to characterize site heterogeneity (e.g. upwelling and downwelling, substrate characteristics); — sampling frequency; — sampling depth; — seasonal variability; — abiotic data requirements; — spatial and temporal scale of investigation. Scale is important when examining the hyporheic zone, as various processes occur at different spatial scales. For example, at a stream-bed (patch) scale the size, shape, sorting and stability of unconsolidated material are the primary determinants of porosity and permeability. These factors have a major influence on community composition over relatively short distances and methods have been developed to address this [11]. At a broader scale, lateral connectivity (e.g. between the riparian zone and out to the wider floodplain) is a key consideration. Hyporheic flow paths occur at multiple scales, from the stream bed to the catchment. SIST EN 16772:2016
