Water quality - Guidance for the identification and enumeration of benthic diatom samples from rivers and lakes

This European Standard specifies methods for the identification and enumeration of relative proportions of diatom taxa on prepared slides and of data interpretation relevant to assessments of water quality in rivers and lakes. It is suitable for use with indices and assessment methods based on the relative abundance of taxa. The methods for identification and enumeration may also be applied to the study of benthic diatoms in other habitats provided that data interpretation methods appropriate to these habitats are used.

Overview

EN 14407:2014 - Water quality: Guidance for the identification and enumeration of benthic diatom samples from rivers and lakes (CEN) provides standardized methods for identifying and counting benthic diatom taxa on prepared slides and for interpreting those data in the context of water quality assessment. Applicable to rivers and lakes, the standard supports indices and assessment methods based on relative taxon abundance and can be adapted to other benthic habitats with appropriate interpretation. EN 14407:2014 supersedes EN 14407:2004 and aligns with monitoring needs driven by policies such as the Water Framework Directive.

Key topics and technical requirements

  • Principle: Clean diatom frustules/valves are mounted in a high-refractive-index medium and identified/enumerated using high-power light microscopy; results are interpreted with indices or reference-condition comparisons.
  • Apparatus: Recommended equipment includes a mechanical-stage light microscope with 100× oil-immersion objective (phase contrast or DIC recommended), eyepiece graticule (calibrated to a stage micrometer), immersion oil, floras/identification guides and imaging/photomicroscopy tools for documentation.
  • Taxonomy and nomenclature: Define and document the taxonomic level (usually species) and nomenclature sources in advance to avoid confusion between parallel naming systems.
  • Units for enumeration: Decide whether counts use valves or frustules (two valves = one frustule); document the convention for comparability.
  • Sample size and counting strategy: Typical count sizes are 300–500 units, but this should be set according to study objectives. Enumeration approaches include field-by-field counts or linear traverses using a graticule; consistent rules for partial/overlapping fields are required.
  • Broken/unidentifiable specimens: Establish a priori rules (e.g., include only if ~¾ present or central area visible) to handle fragmented material.
  • Quality assurance, archiving and data interpretation: Guidance on QA procedures, archiving slides/samples and applying indices or reference-condition frameworks for ecological status assessment.

Applications and who uses it

  • Environmental laboratories and biomonitoring teams performing diatom-based water quality assessments
  • Regulatory agencies and consultants implementing the Water Framework Directive or lake/river monitoring programs
  • Researchers studying eutrophication, acidification, habitat condition or long-term ecological change
Practical benefits include improved comparability of diatom data, reproducible enumeration methods, and robust input for indices that inform ecological status and management decisions.

Related standards and references

  • Supersedes EN 14407:2004 (CEN).
  • Definitions reference ISO-style instruments (e.g., stage micrometer per ISO 10934-1 terminology).
  • Designed to be used alongside national floras, regional checklists and index-specific taxonomic conventions.
