Abstract

ISO 11701:2009 specifies a method for the quantitative determination of phospholipids content by high performance liquid chromatography (HPLC) using a diol column and a light-scattering detector. The method is applicable to crude, oil-containing lecithins, and to oil-free lecithins and lecithin fractions from vegetable fats and oils. The method is not applicable to animal and ruminant lecithins and enzymatically hydrolysed lecithins as the peak separation of lysophosphatidylethanolamine (LPE), lysophosphatidylinositol (LPI) and lysophosphatidic acid (LPA) is insufficient.

1 Scope
This International Standard specifies a method for the quantitative determination of phospholipids content by
high performance liquid chromatography (HPLC) using a diol column and a light-scattering detector.
The method is applicable to crude, oil-containing lecithins, and to oil-free lecithins and lecithin fractions from
vegetable fats and oils.

NOTE The content is expressed in grams per 100 g, numerically equal to a percentage mass fraction.
4 Principle
The individual phospholipids are separated by HPLC using a diol column and a light-scattering detector. For
the purpose of quantification, a certified reference mixture is used.
5 Reagents
WARNING — Comply with any local regulations which specify the handling of hazardous substances.
Technical, organizational and personal safety measures shall be followed.
During the analysis, unless otherwise stated, use only reagents of recognized analytical grade.
5.1 Water, HPLC grade.
5.2 n-Hexane, HPLC grade.
5.3 2-Propanol, HPLC grade.
5.4 Acetic acid, w , 99,8 % mass fraction.

5.5 Triethylamine.
5.6 Solvent mixture: A mixture of 80 ml n-hexane (5.2) and 20 ml 2-propanol (5.3) (volume fraction
ϕ = 80 ml/100 ml for n-hexane and ϕ = 20 ml/100 ml for 2-propanol) is used to dissolve the standards and
sample.

5.7 Reference substance (external standard) ILPS-LE01 , mixed soy phospholipid reference standard,
is a certified reference mixture with defined contents of N-acyl-PE, PA, PE, PC, PI, and LPC.
5.8 Mobile phase for the HPLC.
5.8.1 Eluent A. Mix 814,2 ml of n-hexane (5.2), 170,0 ml of 2-propanol (5.3), 15 ml of acetic acid (5.4), and
0,8 ml of triethylamine (5.5) (volume fraction ϕ = 81,42 ml/100 ml for n-hexane, ϕ = 17,00 ml/100 ml for 2-
propanol, ϕ = 1,50 ml/100 ml for acetic acid, and ϕ = 0,08 ml/100 ml for triethylamine).
In order to obtain a reproducible eluent composition, it is recommended that the solvents be weighed out
taking into account their densities. For a batch size of 2,5 l: 1 341,4 g of n-hexane, 331,5 g of 2-propanol,
39,4 g of acetic acid, and 1,45 g (2,0 ml) of triethylamine.
5.8.2 Eluent B. Mix 844,2 ml of 2-propanol (5.3), 140 ml of water (5.1), 15,0 ml of acetic acid (5.4) and
0,8 ml of triethylamine (5.5) (volume fraction ϕ = 84,42 ml/100ml for 2-propanol, ϕ = 14,00 ml/100 ml for water,
ϕ = 1,50 ml/100 ml for acetic acid and ϕ = 0,08 ml/100 ml for triethylamine).
In order to obtain a reproducible eluent composition, it is recommended that the solvents be weighed out
taking into account their densities. For a batch size of 2,5 l: 1 646,2 g of 2-propanol, 350,0 g of water, 39,4 g
of acetic acid and 1,45 g (2,0 ml) of triethylamine.
6 Apparatus
6.1 Analytical balance, capable of being read to the nearest 0,000 1 g.
6.2 HPLC basic equipment with gradient system and light-scattering detector.
6.3 HPLC column oven, adjustable to 55 °C.
6.4 Degasser or similar equipment for degassing the eluent.

7 Sampling
A representative sample should have been sent to the laboratory. It should not have been damaged or
changed during transport or storage.
Sampling is not part of the method specified in this International Standard. A recommended sampling method

8 Preparation of test sample
See ISO 661.
The sample is heated to a maximum of 60 °C to soften it (overheating shall be avoided) and then
homogenized by vigorous stirring.

1 Domaine d'application
La présente Norme internationale spécifie une méthode de dosage quantitatif des phospholipides par
chromatographie liquide à haute performance (CLHP) en utilisant une colonne de diol et un détecteur à
diffusion de la lumière.
La méthode est applicable aux lécithines brutes contenant de l'huile, aux lécithines exemptes d'huile et aux
fractions de lécithine provenant de corps gras d'origine végétale.
La méthode n'est applicable ni aux lécithines d'origine animale et provenant de ruminants, ni aux lécithines
hydrolysées par des enzymes, la séparation des pics de la lysophosphatidyléthanolamine (LPE), du
lysophosphatidylinositol (LPI) et de l'acide lysophosphatidique (LPA) étant insuffisante.

4 Principe
Les phospholipides individuels sont séparés par CLHP en utilisant une colonne de diol et un détecteur à
diffusion de la lumière. Pour la quantification, un mélange de référence certifié est utilisé.
5 Réactifs
AVERTISSEMENT — Se conformer aux réglementations locales spécifiant comment manipuler les
substances dangereuses. Des mesures de sécurité d'ordre technique, organisationnel et personnel
doivent être suivies.
Au cours de l'analyse, sauf indication contraire, utiliser uniquement des réactifs de qualité analytique
reconnue.
5.1 Eau, de qualité CLHP.
5.2 n-Hexane, de qualité CLHP.

5.4 Acide acétique, w , fraction massique 99,8 %.

5.6 Mélange solvant: Un mélange de 80 ml de n-hexane (5.2) et de 20 ml de 2-propanol (5.3) (fraction
volumique ϕ = 80 ml/100 ml de n-hexane et ϕ = 20 ml/100 ml de 2-propanol) est utilisé pour dissoudre les
étalons et l'échantillon.

5.7 Substance de référence (étalon externe) ILPS-LE01 , étalon de référence constitué d'un mélange
de phospholipides de soja, qui est un mélange de référence certifié ayant une teneur définie en N-acyl-PE,
PA, PE, PC, PI et LPC.
5.8 Phase mobile de CLHP.
5.8.1 Éluant A: Mélanger 814,2 ml de n-hexane (5.2), 170,0 ml de 2-propanol (5.3), 15 ml d'acide
acétique (5.4) et 0,8 ml de triéthylamine (5.5) (fraction volumique ϕ = 81,42 ml/100 ml de n-hexane,
ϕ = 17,00 ml/100 ml de 2-propanol, ϕ = 1,50 ml/100 ml d'acide acétique et ϕ = 0,08 ml/100 ml de
triéthylamine).
Pour obtenir une composition d'éluant reproductible, il est recommandé de peser les solvants en tenant
compte de leur masse volumique. Pour une taille de lot de 2,5 l: 1 341,4 g de n-hexane, 331,5 g de
2-propanol, 39,4 g d'acide acétique et 1,45 g (2,0 ml) de triéthylamine.
5.8.2 Éluant B: Mélanger 844,2 ml de 2-propanol (5.3), 140 ml d'eau (5.1), 15,0 ml d'acide acétique (5.4) et
0,8 ml de triéthylamine (5.5) (fraction volumique ϕ = 84,42 ml/100 ml de 2-propanol, ϕ = 14,00 ml/100 ml
d'eau, ϕ = 1,50 ml/100 ml d'acide acétique et ϕ = 0,08 ml/100 ml de triéthylamine).
Pour obtenir une composition d'éluant reproductible, il est recommandé de peser les solvants en tenant
compte de leur masse volumique. Pour une taille de lot de 2,5 l: 1 646,2 g de 2-propanol, 350,0 g d'eau,
39,4 g d'acide acétique et 1,45 g (2,0 ml) de triéthylamine.

8 Préparation de l'échantillon pour essai

Pour ramollir l'échantillon, le faire chauffer jusqu'à 60 °C maximum (éviter de le surchauffer), puis
l'homogénéiser en l'agitant vigoureusement.

1 Область применения
Настоящий международный стандарт устанавливает метод количественного определения содержания
фосфолипидов методом жидкостной хроматографии высокого разрешения (HPLC) с использованием
диоловой колонки и детектора рассеянного света.
Данный метод применим к неочищенным маслосодержащим лецитинам, не содержащим масла
лецитинам и лецитиновым фракциям из растительных жиров и масел.
Метод не применим к животным и жвачным лецитинам, а также к ферментативно гидролизованным
лецитинам, поскольку разделение пиков лизофосфатидилэтаноламина (LPE), лизофосфатидилинозитола
(LPI) и лизофосфатидной кислоты (LPA) неудовлетворительно.

4 Принцип
Отдельные фосфолипиды разделяют с помощью HPLC, используя диоловую колонку и детектор
рассеянного света. Для количественного определения используют сертифицированную эталонную
смесь.
5 Реактивы
ПРЕДОСТЕРЕЖЕНИЕ — Необходимо учитывать местные регламенты, в которых указываются
правила обращения с опасными веществами. Необходимо соблюдать меры по охране
здоровья и технике безопасности.
При анализе, если не оговорено иначе, используют только реактивы признанного аналитического
качества.
5.1 Вода, качества HPLC.
5.2 н-Гексан, качества HPLC.
5.3 2-Пропанол, качества HPLC.
5.4 Уксусная кислота, w , 99,8 % массовая доля.

5.5 Триэтиламин.
5.6 Смесь растворителей: Смесь 80 мл н-гексана (5.2) и 20 мл 2-пропанола (5.3) (объемная доля
ϕ = 80 мл/100 мл для н-гексана и ϕ = 20 мл/100 мл для 2-пропанола) используют для растворения
стандартов и пробы.

5.7 Эталонное вещество (внешний стандарт) ILPS-LE01 , эталон смеси соевых фосфолипидов —
это сертифицированная эталонная смесь с заданным содержанием N-ацил-PE, PA, PE, PC, PI и LPC.
5.8 Подвижная фаза для HPLC.
5.8.1 Элюент A. Смешивают 814,2 мл н-гексана (5.2), 170,0 мл 2-пропанола (5.3), 15 мл уксусной
кислоты (5.4) и 0,8 мл триэтиламина (5.5) (объемная доля ϕ = 81,42 мл/100 мл для н-гексана,
ϕ = 17,00 мл/100 мл для 2-пропанола, ϕ = 1,50 мл/100 мл для уксусной кислоты и ϕ = 0,08 мл/100 мл
для триэтиламина).
Для получения воспроизводимого состава элюента рекомендуется взвешивать растворители с учетом
их плотности. Для объема партии 2,5 л: 1 341,4 г н-гексана, 331,5 г 2-пропанола, 39,4 г уксусной
кислоты и 1,45 г (2,0 мл) триэтиламина.
5.8.2 Элюент B. Смешивают 844,2 мл 2-пропанола (5.3), 140 мл воды (5.1), 15,0 мл уксусной кислоты
(5.4) и 0,8 мл триэтиламина (5.5) (объемная доля ϕ = 84,42 мл/100мл для 2-пропанола, ϕ = 14,00 мл/100 мл
для воды, ϕ = 1,50 мл/100 мл для уксусной кислоты и ϕ = 0,08 мл/100 мл для триэтиламина).
Для получения воспроизводимого состава элюента рекомендуется взвешивать растворители с учетом
их плотности. Для объема партии of 2,5 л: 1 646,2 г 2-пропанола, 350,0 г воды, 39,4 г уксусной кислоты
и 1,45 г (2,0 мл) триэтиламина.
6 Аппаратура
6.1 Аналитические весы, с возможностью считывания до 0,000 1 г.
6.2 Основное оборудование для HPLC с градиентной системой и детектором рассеянного света.
6.3 Термостат колонки для HPLC, регулируемый до 55 °C.

7 Отбор проб
В лабораторию следует поставлять представительную пробу. Ёе не следует подвергать порче или
изменению во время транспортировки или хранения.
Отбор проб не включен в метод, установленный в этом международном стандарте. Рекомендуемый

8 Подготовка пробы для испытания
См. ISO 661.
Для размягчения пробы нагревают её максимум до 60 °C (необходимо избегать перегрева) и затем
гомогенизируют путем энергичного перемешивания.
