This document specifies a method for the determination of ammonium nitrogen (NH4-N) in drinking water, groundwater, surface water, wastewater, bathing water and mineral water using the small-scale sealed tube method. The result can be expressed as NH4 or NH4-N or NH3 or NH3-N. NOTE 1 In the habitual language use of sewage treatment and on the displays of automated sealed-tube test photometers or spectrophotometers, NH4 without indication of the positive charge has become the common notation for the parameter ammonium. This notation is adopted in this document even though not being quite correct chemical nomenclature. This method is applicable to (NH4-N) concentration ranges from 0,01 mg/l to 1 800 mg/l of NH4-N. The measuring ranges of concentration can vary depending on the type of small-scale sealed tube method of different manufacturers. Concentrations even slightly higher than the upper limit indicated in the manufacturers manual relating to the small-scale sealed tube method used, cannot be reported as accurate results. It is up to the user to choose the small-scale sealed tube test with the appropriate application range or to adapt samples with concentrations exceeding the measuring range of a test by preliminary dilution. Introduction
Ammonium nitrogen is one of the possible forms of nitrogen present in water. It can come directly from
industrial or public waste, from fertilizers or can be generated by the microbial breakdown of urea
and proteins under anaerobic conditions. Ammonium nitrogen can also be found naturally in water in
aquifers with low flow velocities, which are confined and contain organic matter.

1 Scope

develop a colour to be read by a photometer or spectrophotometer
4 Principle
This method is based on the principle of Berthelot reaction and its modification to develop indophenol
blue colour. In strongly alkaline solution, ammonia ions react with dichloroisocyanurate and salicylate
ions or chlorophenol ions, forming blue indophenol. The presence of sodium nitroprusside as catalyst
promotes the development of the blue colour. Smallscale sealed tube is to be read on a photometer or
spectrophotometer.
The method serves to obtain the concentration of ammonium nitrogen present in the sample.
5 Interferences
Typical interferences are due to both cations and anions. Examples for maximum tolerable concentration
values are listed in Table 1. These values refer to both smallscale sealed tube methods applicable at
high concentrations and to those applicable at low concentrations of NH N.
Every manufacturer of small-scale sealed tubes shall provide information about interference levels
above which the ion interferes. The concentration of interfering substances can depend on the ratio of
sampled volume and predosed reagents, in the smallscale sealed tube.

Primary amines can react under analysis conditions and lead to overestimated NH N values. All
reducing substances interfere and lead to underestimated results. The presence of urea is tolerated up
to concentrations which are 10 000 times greater than those of the ammonia present in the sample to
be analysed.
Turbidity, colour and high salinity levels can interfere both in the instrumental measurement and in the
speed of colour development.
As specified in ISO 5667-3, preliminary filtration is necessary using filters with 0,45 μm pore width.
The filtration has to be performed on-site. Filtration shall be carried out under pressure and not under
vacuum, to avoid ammonia leakage. Before filtering, the filter shall be washed with water with the same
pH as the sample. Generally, it can be done with the sample itself by discarding the first millilitres of
sample, already filtered.
When measuring a reagent blank value, follow the normal procedure but use pure water (7.1) instead
of sample.
When measuring a blank value caused by a coloured sample, add the sample to a small-scale sealed
tube but do not add further reagents. Then follow the normal procedure.
In some cases, interferences can be eliminated using appropriate dilutions such that interfering ions
fall below the maximum tolerable concentration and NH N remains within the measurement range.
It is the laboratory's responsibility to identify interferences by, for example, spiking with reference
material containing known amounts of ammonium nitrogen to the sample containing the suspected ion
or to a solution of known concentration of the suspected ion.
In case of higher concentration of interfering substances, highly coloured wastewater or turbid
suspension which cannot be easily filtered or diluted as described above, proceed with a distillation as
described in ISO 5664. An alternative method for distillation is described in Annex A.
6 Sampling and sample preparation
For methods of sampling and storing samples before analysis, proceed according to ISO 5667-1,
ISO 56673 and ISO 566710.
Persons performing the analysis shall be aware of the risks associated with the method.
It shall be noted that, in the case of acidification of unknown samples, toxic gases such as HCN and H S
can form. Work in a fume cupboard.
Non-preserved samples shall be analysed as soon as possible, in accordance with ISO 5667-3. Longer
storage periods require stabilization by acidification with sulfuric acid to pH < 2; if the sample is
acidified, before proceeding with the determination, the pH shall be brought within the range indicated
in the procedure for the test being used. When stabilizing samples, make sure to use ammonium-free
reagents. Store samples in the refrigerator as described in ISO 56673.

Introduction
Ammonium nitrogen is one of the possible forms of nitrogen present in water. It can come directly from
industrial or public waste, from fertilizers or can be generated by the microbial breakdown of urea and
proteins under anaerobic conditions. Ammonium nitrogen can also be found naturally in water in
aquifers with low flow velocities, which are confined and contain organic matter.

1 Scope
This document specifies a method for the determination of ammonium nitrogen (NH -N) in drinking
water, groundwater, surface water, wastewater, bathing water and mineral water using the small-scale
sealed tube method. The result can be expressed as NH or NH -N or NH or NH -N.

NOTE 1 In the habitual language use of sewage treatment and on the displays of automated sealed-tube test
photometers or spectrophotometers, NH without indication of the positive charge has become the common
notation for the parameter ammonium. This notation is adopted in this document even though not being quite
correct chemical nomenclature.
This method is applicable to (NH -N) concentration ranges from 0,01 mg/l to 1 800 mg/l of NH -N. The

measuring ranges of concentration can vary depending on the type of small-scale sealed tube method of
different manufacturers. Concentrations even slightly higher than the upper limit indicated in the
manufacturers manual relating to the small-scale sealed tube method used, cannot be reported as
accurate results. It is up to the user to choose the small-scale sealed tube test with the appropriate
application range or to adapt samples with concentrations exceeding the measuring range of a test by
preliminary dilution.
NOTE 2 The results of a small-scale sealed tube are most precise in the middle of the application range of the test.
All manufacturers' methods are based on the Berthelot reaction and its modifications to develop
indophenol blue colour. Reagents mixtures can differ slightly based on manufacturers small-scale sealed
tube method, see Clause 9. This method is applicable to non-preserved samples by using small-scale
sealed tubes for the determination of drinking water, groundwater, surface water, wastewater and to
preserved samples. The method is applicable to samples with suspended materials if these materials are
removable by filtration.

develop a colour to be read by a photometer or spectrophotometer.
4 Principle
This method is based on the principle of Berthelot reaction and its modification to develop indophenol
blue colour. In strongly alkaline solution, ammonia ions react with dichloroisocyanurate and salicylate
ions or chlorophenol ions, forming blue indophenol. The presence of sodium nitroprusside as catalyst
promotes the development of the blue colour. Small-scale sealed tube is to be read on a photometer or
spectrophotometer.
The method serves to obtain the concentration of ammonium nitrogen present in the sample.
5 Interferences
Typical interferences are due to both cations and anions. Examples for maximum tolerable concentration
values are listed in Table 1. These values refer to both small-scale sealed tube methods applicable at high
concentrations and to those applicable at low concentrations of NH -N.
Every manufacturer of small-scale sealed tubes shall provide information about interference levels above
which the ion interferes. The concentration of interfering substances can depend on the ratio of sampled
volume and predosed reagents, in the small-scale sealed tube.

Primary amines can react under analysis conditions and lead to overestimated NH -N values. All reducing
substances interfere and lead to underestimated results. The presence of urea is tolerated up to
concentrations which are 10 000 times greater than those of the ammonia present in the sample to be
analysed.
Turbidity, colour and high salinity levels can interfere both in the instrumental measurement and in the
speed of colour development.
As specified in ISO 5667-3, preliminary filtration is necessary using filters with 0,45 μm pore width. The
filtration has to be performed on-site. Filtration shall be carried out under pressure and not under
vacuum, to avoid ammonia leakage. Before filtering, the filter shall be washed with water with the same
pH as the sample. Generally, it can be done with the sample itself by discarding the first millilitres of
sample, already filtered.
When measuring a reagent blank value, follow the normal procedure but use pure water (7.1) instead of
sample.
When measuring a blank value caused by a coloured sample, add the sample to a small-scale sealed tube
but do not add further reagents. Then follow the normal procedure.
In some cases, interferences can be eliminated using appropriate dilutions such that interfering ions fall
below the maximum tolerable concentration and NH -N remains within the measurement range.
It is the laboratory's responsibility to identify interferences by, for example, spiking with reference
material containing known amounts of ammonium nitrogen to the sample containing the suspected ion
or to a solution of known concentration of the suspected ion.
In case of higher concentration of interfering substances, highly coloured wastewater or turbid
suspension which cannot be easily filtered or diluted, as described above, proceed with a distillation as
described in ISO 5664. An alternative method for distillation is described in Annex A.
6 Sampling and sample preparation
For methods of sampling and storing samples before analysis see, proceed according to ISO 5667--1,
ISO 5667--3 and ISO 5667--10.
Persons performing the analysis shall be aware of the risks associated with the method.
It shall be noted that, in the case of acidification of unknown samples, toxic gases such as HCN and H S
maycan form. Work in a fume cupboard.
Non-preserved samples shall be analysed as soon as possible, following in accordance with ISO 5667-3.
Longer storage periods require stabilization by acidification with sulfuric acid to pH < < 2; if the sample

is acidified, before proceeding with the determination, the pH shall be brought within the range indicated
in the procedure for the test being used. When stabilizing samples, make sure to use ammonium-free
reagents. Store samples in the refrigerator as described in ISO 5667--3.
Due to the differences in used small-scale sealed tube between all the manufacturers it is recommended
to follow manufacturers' manuals.
7 Reagents
7.1 Water, the concentration of nitrogen in the used water shall be below 25 % of the lower
measurement range of the used sealed tube.
7.2 Ammonium chloride, NH Cl, of analytical grade, previously dried at 105 °C for 2 h, to be used for
preparing suitable solutions to verify the calibration in the NH -N concentration range. Commercially
available ammonium chloride solutions, with known concentration, can also be used as reference
material.
7.3 Reagents provided by the manufacturers for the small-scale sealed tube method and used to
determine NH -N with colour reactions (see Clause 9)

Introduction
L’azote-ammonium est l’une des formes possibles d’azote présent dans l’eau. Il peut provenir directement
de déchets industriels ou ménagers, ou d’engrais, ou peut être généré par la dégradation microbienne de
l’urée et des protéines dans des conditions anaérobies. L’azote-ammonium peut également être trouvé
naturellement dans l’eau des aquifères à faible vitesse d’écoulement, qui sont confinés et qui contiennent de
la matière organique.

1 Domaine d’application
Le présent document spécifie une méthode de détermination de l’azote-ammonium (NH -N) dans l’eau
potable, les eaux souterraines, les eaux de surface, les eaux usées, les eaux de baignade et les eaux minérales
en utilisant la méthode à petite échelle en tubes fermés. Le résultat peut être exprimé en NH ou NH -N ou

NOTE 1 Dans le langage habituel du traitement des eaux usées et sur les écrans des photomètres ou des
spectrophotomètres à essai en tubes fermés automatisés, NH sans indication de la charge positive est devenu la
notation courante pour le paramètre ammonium. Cette notation est adoptée dans le présent document, même si la
nomenclature chimique n’est pas tout à fait correcte.
Cette méthode s’applique aux plages de concentration de (NH -N) de 0,01 mg/l à 1 800 mg/l de NH -N. Les

plages de concentration de mesure peuvent varier en fonction du type de méthode en tubes fermés à petite
échelle des différents fabricants. Les concentrations, même légèrement supérieures à la limite supérieure
indiquée dans le manuel du fabricant concernant la méthode en tubes fermés à petite échelle utilisée, ne
peuvent pas être consignées comme des résultats exacts. Il appartient à l’utilisateur de choisir l’essai en
tubes fermés à petite échelle avec la plage d’application appropriée ou d’adapter les échantillons dont les
concentrations dépassent la plage de mesure d’un essai par dilution préliminaire.
NOTE 2 Les résultats en tubes fermés à petite échelle sont plus précis au milieu de la plage d’application de l’essai.
Toutes les méthodes des fabricants sont basées sur la réaction de Berthelot et ses modifications pour
développer la couleur bleu indophénol. Les mélanges de réactifs peuvent différer légèrement en fonction
de la méthode en tubes fermés à petite échelle du fabricant, voir l’Article 9. Cette méthode s’applique aux
échantillons non stabilisés en utilisant des tubes fermés à petite échelle pour la détermination de l’eau
potable, des eaux souterraines, des eaux de surface et des eaux usées, ainsi qu’aux échantillons stabilisés. La
méthode s’applique aux échantillons contenant des matériaux en suspension si ces matériaux peuvent être
retirés par filtration.

destiné à développer une couleur à lire par un photomètre ou un spectrophotomètre
4 Principe
Cette méthode est basée sur le principe de la réaction de Berthelot et sa modification pour développer
la couleur bleu indophénol. Dans une solution fortement alcaline, l’ammoniac réagit avec les ions de
dichloroisocyanurate et de salicylate ou de chlorophénol, formant de l’indophénol bleu. La présence de
nitroprussiate de sodium comme catalyseur favorise le développement de la couleur bleu. Le tube fermé à
petite échelle est lu sur un photomètre ou un spectrophotomètre.
La méthode permet d’obtenir la concentration en azote-ammonium présente dans l’échantillon.
5 Interférences
Les interférences types sont dues à la fois aux cations et aux anions. Des exemples de valeurs de concentration
maximale tolérable sont donnés dans le Tableau 1. Ces valeurs font référence à la fois aux méthodes en
tubes fermés à petite échelle applicables à des concentrations élevées et à celles applicables à de faibles
concentrations de NH -N.
Chaque fabricant de tubes fermés à petite échelle doit fournir des informations sur les niveaux d’interférence
au-delà desquels l’ion interfère. La concentration des substances interférentes peut dépendre du rapport
entre le volume prélevé et les réactifs pré-dosés, dans le tube fermé à petite échelle.

Les amines primaires peuvent réagir dans les conditions d’analyse et conduire à des valeurs de NH -N
surestimées. Toutes les substances réductrices interfèrent et conduisent à des résultats sous-estimés. La
présence d’urée est tolérée jusqu’à des concentrations 10 000 fois supérieures à celles de l’ammoniac présent
dans l’échantillon à analyser.
La turbidité, la couleur et des niveaux élevés de salinité peuvent interférer à la fois dans la mesure
instrumentale et dans la vitesse de développement des couleurs.
Comme spécifié dans l’ISO 5667-3, une filtration préliminaire est nécessaire en utilisant des filtres d’une
taille de pore de 0,45 μm. La filtration doit être réalisée sur site. La filtration doit être effectuée sous pression
et non sous vide, afin d’éviter les fuites d’ammoniac. Avant la filtration, le filtre doit être lavé avec de l’eau
ayant le même pH que l’échantillon. En général, cela peut être fait avec l’échantillon lui-même en éliminant
les premiers millilitres d’échantillon, déjà filtrés.
Lors de la mesure d’une valeur de blanc réactif, suivre le mode opératoire normal, mais utiliser de l’eau pure
(7.1) à la place de l’échantillon.
Lors du mesurage d’une valeur de blanc due à un échantillon coloré, ajouter l’échantillon dans un tube fermé
à petite échelle, mais ne pas ajouter d’autres réactifs. Suivez ensuite le mode opératoire normal.
Dans certains cas, les interférences peuvent être éliminées en utilisant des dilutions appropriées de sorte
que les ions interférents tombent en dessous de la concentration maximale tolérable et que le NH -N reste
dans la plage de mesure.
Il est de la responsabilité du laboratoire d’identifier les interférences, par exemple en enrichissant
l’échantillon contenant l’ion suspecté ou une solution de concentration connue de l’ion suspecté avec un
matériau de référence contenant des quantités connues d’azote-ammonium.
En cas de concentration plus élevée de substances interférentes, d’eaux usées fortement colorées ou de
suspension trouble qui ne peut pas être facilement filtrée ou diluée comme décrit ci-dessus, procéder à une
distillation comme décrit dans l’ISO 5664. L’Annexe A décrit une autre méthode de distillation.
6 Échantillonnage et préparation des échantillons
Pour les méthodes d’échantillonnage et de stockage des échantillons avant analyse, procéder conformément
à l’ISO 5667-1, l’ISO 5667-3 et l’ISO 5667-10.
Les personnes réalisant l’analyse doivent être conscientes des risques associés à la méthode.
Il faut noter que, dans le cas de l’acidification d’échantillons inconnus, des gaz toxiques tels que le HCN et le
H S peuvent se former. Travailler sous hotte aspirante.
Les échantillons non stabilisés doivent être analysés dès que possible, conformément à l’ISO 5667-3. Des
périodes de stockage plus longues nécessitent une stabilisation par acidification à l’acide sulfurique jusqu’à
un pH < 2; si l’échantillon est acidifié, avant de procéder à la détermination, le pH doit être ramené dans
la plage indiquée dans le mode opératoire d’essai utilisé. Lors de la stabilisation des échantillons, veiller
à utiliser des réactifs exempts d’ammonium. Stocker les échantillons au réfrigérateur comme décrit dans
l’ISO 5667-3.
En raison des différences dans les tubes fermés à petite échelle utilisés par les différents fabricants, il est
recommandé de suivre les manuels des fabricants.
7 Réactifs
7.1 Eau, la concentration d’azote dans l’eau utilisée doit être inférieure à 25 % de la plage de mesure
inférieure du tube fermé utilisé.
7.2 Chlorure d’ammonium, NH Cl, de qualité analytique, préalablement séché à 105 °C pendant 2 h, à
utiliser pour préparer des solutions appropriées afin de vérifier l’étalonnage dans la gamme de concentrations
de NH -N. Des solutions de chlorure d’ammonium disponibles sur le marché, dont la concentration est
connue, peuvent également être utilisées comme matériau de référence.

Lors du mesurage d’une valeur de blanc due à un échantillon coloré, ajouter l’échantillon dans un tube fermé
à petite échelle, mais ne pas ajouter d’autres réactifs. Suivez ensuite le mode opératoire normal.
Dans certains cas, les interférences peuvent être éliminées en utilisant des dilutions appropriées de sorte que
les ions interférents tombent en dessous de la concentration maximale tolérable et que le NH -N reste dans la
plage de mesure.
Il est de la responsabilité du laboratoire d’identifier les interférences, par exemple en enrichissant
l’échantillon contenant l’ion suspecté ou une solution de concentration connue de l’ion suspecté avec un
matériau de référence contenant des quantités connues d’azote-ammonium.

6 Échantillonnage et préparation des échantillons
Pour les méthodes d’échantillonnage et de stockage des échantillons avant analyse, procéder conformément
à l’ISO 5667--1, l’ISO 5667--3 et l’ISO 5667--10.

En raison des différences dans les tubes fermés à petite échelle utilisés par les différents fabricants, il est
recommandé de suivre les manuels des fabricants.
7 Réactifs
7.1 7.1 Eau, la concentration d’azote dans l’eau utilisée doit être inférieure à 25 % de la plage de
mesure inférieure du tube fermé utilisé.
7.2 7.2 Chlorure d’ammonium, NH Cl, de qualité analytique, préalablement séché à 105 °C
pendant 2 h, à utiliser pour préparer des solutions appropriées afin de vérifier l’étalonnage dans la gamme de
concentrations de NH4-N. Des solutions de chlorure d’ammonium disponibles sur le marché, dont la
concentration est connue, peuvent également être utilisées comme matériau de référence.
7.3 7.3 Réactifs fournis par les fabricants pour la méthode des tubes fermés à petite
échelle et utilisés pour déterminer le NH -N avec des réactions colorimétriques (voir
l’Article 9l’Article 9)).

8.2 8.2 Photomètre ou spectrophotomètre, pour la lecture des tubes fermés à petite échelle à
une longueur d’onde conforme aux réactions colorimétriques données en 9.29.2 ou 9.39.3.
NOTE En général, un instrument automatisé possède, en mémoire, les courbes d’étalonnage requises pour une
utilisation de routine et peut également en enregistrer de nouvelles.
