Международная организация по стандартизации (ISO) является всемирной федерацией национальных
организаций по стандартизации (комитетов-членов ISO). Разработка международных стандартов
обычно осуществляется техническими комитетами ISO. Каждый комитет-член, заинтересованный в
деятельности, для которой был создан технический комитет, имеет право быть представленным в этом

Введение
Чтобы обеспечить получение репрезентативных проб для лаборатории, ответственной за испытания,
важно применить подходящий метод отбора проб. Особенности отбора проб зависят от цели
получения пробы, а также от природы пробы. Микроорганизмы являются живыми существами. Кроме
того, когда они попадают в воду, они образуют не настоящий раствор, а суспензию с присущей
степенью изменчивости.
Цели отбора проб могут быть разными. Эти цели описаны в серии стандартов ISO 5667 (ISO 5667-1,
ISO 5667-2 и ISO 5667-3), в т.ч.:
a) определение соответствия воды качеству, требуемому регламентом;
b) характеризация загрязнения, уровень загрязнения (средний) и его варианты:
1) что такое случайное изменение?
2) является ли это тенденцией?
3) существуют ли циклы?
c) идентификация источников загрязнения.
Относительно количества или частоты выборки она будет меняться в зависимости от цели отбора
проб.
Минимальное число проб будет меньше, если средняя концентрация значительно отличается от
технических условий (гораздо ниже или гораздо выше), минимальное число проб будет выше, если
средняя концентрация и технические требования близки по значению. Аналогично, в случае b), при
рассмотрении тенденции: чем менее очевидна тенденция, тем выше частота отбора проб (см. также
Приложение A).

ПРЕДУПРЕЖДЕНИЕ — Пользователи должны быть знакомы с обычной лабораторной практикой.
Настоящий стандарт не ставит цели рассмотреть все проблемы, связанные с безопасностью
(если таковые имеются) и возникающие при его применении. Пользователь несет
ответственность за установление соответствующей техники безопасности и охраны здоровья и
обеспечение соответствия условиям национальных регламентов.

1 Область применения
Настоящий международный стандарт обеспечивает руководство по планированию проведения отбора

микробиологических исследований.

3 Точка отбора проб
Место отбора проб должно обеспечивать репрезентативные характеристики и учитывать изменчивость
по вертикали, по горизонтали и по времени, а также должно быть точно идентифицировано в
соответствии с общими рекомендациями, приведенными в ISO 5667-1 и ISO 5667-2, принимая во
внимание дополнительные аспекты, специфические для микробиологии.

Следует избегать точек отбора проб, в которых условия нестабильны, при этом необходимо учитывать
гетерогенность водной системы. При изучении эффективности обеззараживания точку отбора проб
необходимо выбирать так, чтобы обеспечить завершение реакции.
ПРИМЕР Примерами того, как гетерогенность системы может повлиять на результаты, показаны ниже.
 Не будет равноценным отбор поверхностной и подповерхностной пробы, или подповерхностной пробы,
“загрязненной” при извлечении через поверхностную пленку. В некоторых случаях (например, озера,
плавательные бассейны), концентрация в поверхностной пленке может быть в 1 000 раз выше, чем под
поверхностью.
 Все точки водной системы будут не равноценны, поскольку могут иметься участки стоячей воды и участки, где
течение незначительно, особенно если рассматриваемая система питается из двух источников.
 Качество на выходе из хорошо перемешиваемого резервуара обычно такое же, как в самом водном объекте,
но может значительно отличаться от качества на входе.
4 Техника отбора проб
4.1 Персонал
Для всего персонала, занятого отбором проб должно быть описано в деталях официальное обучение,
отчеты об обучении и определение компетенции должно быть описано для всех тех, кто производит
отбор проб, эта информация должна быть подтверждена документально.
4.2 Контейнеры для проб
4.2.1 Общие положения
Для повседневных проб (например, отбор проб из водопровода, водоемов в зонах отдыха,
плавательных бассейнов), используют чистые, стерильные бутыли. Объем бутылей должны быть
адекватным для анализа по всем требуемым параметрам.
Для отбора проб путем погружения в чистую воду, используют бутыли, которые являются стерильными
внутри и снаружи и защищены, например, крафт-бумагой (чтобы оставались сухими после обработки в
автоклаве), алюминиевой фольгой или наружными пластиковыми пакетами.
Без обработки в автоклаве, можно использовать стерилизацию гамма-лучами или этиленоксидом.
Пакет можно открывать непосредственно перед отбором проб, он также может служить в качестве
перчатки, чтобы держать бутыль, обеспечивая максимальную асептику перед помещением в стойку-
штатив или другое оборудование, в котором можно стерилизовать бутыли.
Альтернативно, с наружной стороны бутыли для проб можно обеззараживать непосредственно перед
погружением с помощью специального дезинфицирующего средства, например, изопропанола (4.3.1.1)
и дать остыть перед применением. Это не подходит для анализа бактерий, образующих споры.
В большинстве случаев достаточно использовать, бутыли вместимостью 500 мл, поскольку
испытывают не более 5 видов микроорганизмов, причем в каждом исследовании инокулируют
максимум 100 мл материала.
В некоторых случаях, потребуются бóльшие объемы, например:
 для анализа бутилированной воды (250 мл на измерение каждого параметра);

 для Legionella spp. или Salmonella spp. (до 1 л);
 для вирусов, Giardia цист, Cryptosporidium оцист, амёб в чистой воде, исследуют от 10 до
нескольких сотен литров или больше. Обычно, этап концентрирования выполняют на месте,
используя патронный фильтр, который затем транспортируют в лабораторию.
Бутыли можно изготавливать из стекла или различных пластмасс (полипропилена, полистирола,
полиэтилена, поликарбоната). Стекло обычно предпочтительно для повторного использования,
полиэтилен используется один раз.
Прилипание к поверхностям может ухудшить обнаружение микроорганизмов, также необходимо
учитывать критическое тангенциальное поверхностное натяжение γ, если используется нестандартный

материал .
Пробки для стеклянных бутылей должны быть либо притертыми, либо пластиковыми, для пластиковых
бутылей и канистр используют пластиковые плотно надеваемые крышки, или пластиковые или
металлические навинчивающиеся крышки для других сосудов. Отверстия бутылей, закрытые
пластиковой или стеклянной крышкой, необходимо защитить сверху от загрязнения, например,
алюминиевой фольгой.
Если для анализа необходимы большие объемы, например, для вирусов, Salmonella spp., амёб,
Cryptosporidium oцист, Giardia цист, иногда требуется подвергнуть анализу десятки или сотни литров.
Чтобы избежать трудностей обращения, охлаждения и встряхивания таких объемов, рекомендуется
применять концентрирование in situ (путем флокуляции, центрифугирования или фильтрования).
Можно использовать шланговый насос со стерильными трубками.
ПРИМЕЧАНИЕ 1 Металлические крышки, особенно из алюминия, при автоклавировании могут выделять
токсичные вещества. Это можно предотвратить путем использования термостойкой непротекающей прокладки.
ПРИМЕЧАНИЕ 2 Определенные материалы могут также давать токсичные побочные продукты при
стерилизации под нагревом, даже в сушильной печи, или приводить к изменению рН.
ПРИМЕЧАНИЕ 3 Хлопковая вата некоторых производителей, используемая для изготовления тампонов для
стеклянной посуды, может стать токсичной при слишком продолжительном нагревании при слишком высоких
температурах.
ПРИМЕЧАНИЕ 4 Плотно надеваемые пластиковые крышки на бутыль или канистру имеют несколько
преимуществ, так как они не протекают, как навинчиваемые крышки, и при открытой крышке, что облегчает
наполнение и забор проб из емкости пипеткой, такая крышка остается присоединенной к бутыли, поэтому бутыли
и крышки держатся вместе, что защищает крышку от дополнительного загрязнения.
4.2.2 Стерилизация бутылей
При повторном использовании стеклянные бутыли с крышками моют нетоксичным, не содержащим
фосфора моющим веществом с последующим тщательным споласкиванием деионизованной или
дистиллированной водой.
Бутыли обрабатывают в автоклаве при температуре (121 ± 3) °C в течение не менее 15 мин. Бутыли
закрывают неплотно, чтобы пар мог вытеснить воздух при подъеме температуры и чтобы
предотвратить растрескивание пластиковых бутылей при охлаждении. После стерилизации
завинчивающиеся крышки плотно закрывают. Стеклянные пробки обрабатывают в автоклаве отдельно
от бутылей или используют бумагу или алюминиевый разделитель, чтобы предотвратить заедание при
охлаждении.

При необходимости бутыли стерилизуют в сушильной печи в течение не менее 1 ч при температуре
(170 ± 10) °C. Притертые пробки в горлышка прокладывают бумажной лентой или веревочкой, чтобы
пробка не застряла при охлаждении. Бутыли должны прослеживаться по дате стерилизации.
Процесс стерилизации контролируют с помощью химических или биологических индикаторов.
Если невозможно осуществить стерилизацию другими средствами, дезинфицируют бутыли путем
погружения их в открытом виде в кипящую воду не менее чем на 30 мин. Сразу после кипения
опустошают бутыли и закрывают их прокипевшими крышками и оборачивают чистой бумагой.
ПРИМЕЧАНИЕ 1 Полиэтиленовые бутыли можно стерилизовать под воздействием газа этиленоксида, но ввиду
его токсичности, процедуру можно выполнить только на специальной установке с учетом времени, разрешенного
для десорбции газообразного этиленоксида. Поэтому такой метод не используется в качестве повседневного в
лабораторной практике.

достаточной энергии (от 1 × 10 Gy до 2 × 10 Gy) является очень эффективной техникой стерилизации, доступной
только для специальных установок. остаточной антибактериальной активности не будет, но некоторые материалы
могут измениться за счет полимеризации после повторного облучения.
4.2.3 Дезактивация дезинфицирующего средства
Чтобы оценить микробиологическое качество воды, обеззараженной окислителем (например, хлором,
хлорамином, бромом или озоном), прекращают действие окислителя, как только пробу отобрали.
Добавляют восстановитель, такой как тиосульфат натрия в бутыли для проб.
Теоретическая масса тиосульфата натрия (пентагидрата), требующегося для дезактивации 1 мг хлора
составляет 7,1 мг. Таким образом, 0,1 мл раствора пентагидрата тиосульфата натрия (4.3.1.2)
добавляют на каждые 100 мл вместимости бутыли. Это дезактивирует не менее 2 мг/л и вплоть до
5 мг/л остатка свободного хлора, в зависимости от динамики дезактивации, что будет достаточным для
большинства проб.
В определенных обстоятельствах, например, как меры по дезинфекции тазов в плавательных
бассейнах (например, уничтожение Legionella в водопроводных системах подачи питьевой воды),
может быть обнаружена более высокая концентрация и пропорционально потребуется более высокая
доза тиосульфата натрия.
Тиосульфат натрия не разрушается при автоклавировании или сухим теплом. Обеспечивают, чтобы
pH раствора тиосульфата натрия был примерно нейтральным (низкий pH может привести к
разложению).
Тиосульфат натрия не действует на пробу и может также использоваться для не хлорированной воды.
ПРИМЕЧАНИЕ Имеется информация, что Legionella чувствительна к натрию и что предпочтительно
использовать тиосульфат калия, однако, отрицательного эффекта натрия при концентрации, используемой для
дезактивации хлора в обычной концентрации, обнаружено не было.
Для других дезинфицирующих средств необходимо принимать другие соответствующие меры
дезактивации. Если дезактивация невозможна или не осуществима, об этом необходимо сообщить.
Для защиты бактерий от токсичного действия тяжелых металлов, таких как медь и цинк, рекомендуется
использовать хелатообразователи. Этилендинитрилотетрауксусную кислоту (EDTA) или нитрилоацетат
натрия, ее натриевую соль (NTA) (Na C H NO ) можно использовать как фильтр-стерилизующий раствор

при конечной концентрации примерно 50 мг на литр, но добавлять ее следует только при необходимости
(например, если вода была обработана серебром или медью). Серебро можно также дезактивировать
сульфидом натрия. Добавляют 1 мл раствора сульфида натрия (4.3.1.3) на 1 л пробы.

4.2.4 Контроль качества бутылей для проб
4.2.4.1 Определение стерильности
Лаборатория должна убедиться в стерильности бутылей для проб, независимо от того, стерилизовали
бутыли в лаборатории или приобрели в продаже, а также пластиковые это бутыли или стеклянные.
Приобретенные в продаже бутыли должны поставляться с сертификатом, подтверждающим
стерилизацию, что является условием приемки, однако тест на стерильность рекомендуется провести
для полученной партии. Это касается партии бутылей после этикетирования, добавления
дезактивирующих веществ, где необходимо, и хранения.
Стерильность бутылей обычно можно гарантировать путем контроля процесса стерилизации. В
противном случае стерильность контейнеров необходимо подтвердить тестированием.
ПРИМЕР Далее следуют примеры процедур испытания (обычно выполняемых на 1 из 100 бутылей):
a) Метод “катания бутыли”
Этот метод заключается в следующем: в испытуемую бутыль вводят 20 мл или 50 мл расплавленного
пищевого агара (агар для подсчета колоний в чашках) и покрывают стенки агаром, вращая бутыль в процессе
охлаждения (если необходимо, под струйкой воды). Инкубация при температуре (22 ± 2) °C в течение пяти
дней не должна выявить заметного роста.
b) Метод с использованием жидкого бульона
Метод заключается в следующем: в бутыль наливают от 20 мл до 50 мл тиогликолята или другого пищевого
бульона, вращая бутыль таким образом, чтобы смочить все стенки, и инкубируют при температуре (22 ± 2) °C
в течение 5 дней. Если бутыль стерильна, не должно появиться помутнения на ее стенках.
4.2.4.2 Тест на присутствие дезактивирующих веществ
Присутствие тиосульфата можно проверить иодометрическим методом:

Добавляют 10 мл дистиллированной воды в бутыль и титруют раствором йода (4.3.1.4), используя
крахмал или тиофен в качестве титрующего вещества до конечной точки титрования.
4.2.4.3 Тест на остаточную токсичность в бутылях для проб
Причиной остаточной токсичности в контейнерах для проб может быть метод промывания стекла,
результат выделения компонентов или добавок из пластиковых бутылей, а также собственно процесс
стерилизации. Повседневное применение стеклянных или полиэтиленовых бутылей не требует
регулярной проверки на токсичность, но в случае сомнений, выполняют тест по Гельдриху (Geldreich)
[8]
1975 (например).
4.3 Реактивы, аппаратура и материалы

4.3.2.5 Маркеры, карандаши, этикетки.
4.3.2.6 Гаечные ключи, клещи, отвертки, нож.
4.3.2.7 Коробка для льда и лед, или кубики льда, портативные холодильники или холодильные
камеры в автомобилях.
4.3.2.8 Термометр или регистрирующее устройство.
4.3.2.9 Балластированный держатель для бутылей или равноценное приспособление, с
веревкой или цепью (предпочтительно из нержавеющей стали, хотя бы нижняя часть).
4.3.2.10 Шест или длинные щипцы или пробоотборники, приспособленные для различных глубин.
4.3.2.11 Карты, перечень точек отбора проб, бланки для заполнения.
4.3.2.12 Средство транспортирования и документация, удостоверение личности или
разрешение на забор проб.
4.3.2.13 Водонепроницаемая (защитная) обувь.
4.3.2.14 Аппаратура для измерения pH, хлора, растворенного кислорода, проводимости.
4.3.2.15 Стерильные перчатки.
4.4 Процедура наполнения бутылей
4.4.1 Питьевая вода из водопровода
4.4.1.1 Общие положения
Отбор проб из водопровода может производиться с разными целями:
a) чтобы определить качество воды в водопроводной системе (ответственность несет организация,
ведающая этой системой);
b) чтобы определить качество воды, подаваемой по водопроводу потребителю — в состоянии подачи
в водопровод — (которое может измениться в обслуживающей сети внутри здания);

c) чтобы определить качество потребляемой воды, т.е. в состоянии, в котором она вытекает из
водопроводного крана (возможно уже загрязненном).
Пробы для оценки качества в магистральном водопроводе [пункт a)] лучше всего отбирать через
специальные краны (также находящиеся в системе распределения), которые расположены вблизи
магистрального водопровода, чистые, без подсоединений, дезинфицируемые с помощью

Можно использовать обычные краны для определения качества воды в магистральном водопроводе
[снова пункт a)], если они дезинфицируются фламбированием, а в случае неясных результатов,
рассматривать обслуживающую сеть как потенциальный источник загрязнения.
Ситуация, описанная в случае b), предполагает метод выбора оценки качества питьевой воды,
включая влияние обслуживающей сети внутри здания. В этом случае, краны, дезинфицируемые
фламбированием, имеются не всегда, и необходимо рассмотреть другие методы обеззараживания

Отбор проб из резервуаров-хранилищ питьевой воды обычно выполняют из крана на выходном
отверстии. Подповерхностные пробы иногда забирают из самого резервуара, в случае чего требуются
бутыли, стерильные снаружи и внутри (см. 4.2.1).
Обеспечивают асептический отбор проб чистыми руками или в стерильных перчатках, защищая пробу
от сквозняков и разбрызгивания.

При наполнении бутыли не допускается контакта внутренней стороны крышки бутыли с чем-либо (с
пальцами, землей, карманом, зубами).
В бутыли оставляют воздушное пространство, чтобы можно было встряхивать содержимое перед
анализом.
Бутыль сразу после забора проб закрывают. Полученную пробу воды нельзя использовать для
измерения температуры или других параметров, определяемых на месте.

4.4.1.2 Вода на очистных сооружениях и в резервуарах-хранилищах
На очистных сооружениях и в резервуарах-хранилищах рекомендуется иметь специальные краны для
отбора проб на каждом выходном отверстии и в других точках отбора проб. Необходимо, чтобы эти
краны можно было стерилизовать фламбированием, поддерживать в чистом состоянии, четко
этикетировать и использовать исключительно для отбора проб. Для дополнительной информации см.
ISO 5667-5 и ISO 5667-13.
Тщательное фламбирование металлического крана с помощью паяльной лампы обеспечивает
дезинфекцию выходного отверстия крана, если температура при фламбировании достигает 80 °C или
больше. Это нельзя применять, если в кране, в нагреваемой его части, остается вода.
ПРИМЕЧАНИЕ Фламбирование с помощью зажигалки является поверхностным (этого недостаточно).
4.4.1.3 Вода в магистральном водопроводе
Чтобы определить качество воды в магистральном водопроводе, отбирают пробы в этом трубопроводе
или вблизи него (обычно сразу после счетчика). Следят за тем, чтобы из окружающего пространства
крана загрязнение не попало в пробу. Не допускается отбирать пробы из текущих кранов, а также
рекомендуется, по возможности, избегать смесителей. Снимают все насадки с крана или другие
соединения или вставки (потребуются гаечные ключи и плоскогубцы). Соскребают всю налипшую грязь
(окалину, известковый налет, смазку или другой налипший материал) и полностью открывают и
закрывают кран, чтобы смыть грязь из крана. Предпочтительно дезинфицировать кран
фламбированием (после фламбирования при открытии крана должен возникнуть шипящий звук).
Далее открывают кран наполовину напора и сливают воду, пока не будет достигнута постоянная
температура воды (чтобы не набрать воду из водопровода, расположенного уже внутри здания). Затем
помещают открытую бутыль в струю воды и наполняют ее в асептических условиях.
Только если фламбирование невозможно, дезинфицируют кран другими адекватными методами.
Чтобы дезинфицировать носик пластикового крана после тщательного промывания, погружают его на
2–3 мин в стакан с раствором гипохлорита, этанола или изопропанола (4.3.1.1). Альтернативно можно
использовать ершик или промывную бутыль или аналогичное приспособление, чтобы
дезинфицировать кран снаружи, и по возможности, максимально с внутренней стороны.
Далее спускают воду достаточно долго, чтобы свести к минимуму влияние воды водопроводной сети
внутри здания. Необходим подробный план компоновки сети (объем резервуаров или установок
умягчения воды и время удерживания), чтобы определить время промывания перед отбором проб.
Стабилизацию температуры воды можно отслеживать, чтобы достигать одинакового эффекта для
одинаковых целей. Большинство обнаруживаемых в воде микроорганизмов попадают в нее в
результате разрыва биопленки и повторного суспендирования отложений из стыков или колен в случае
стока паводка или резервуаров, работающих под давлением. Чтобы свести к минимуму такие эффекты,
открывают кран на максимальный напор на 5–10 с, затем уменьшают напор вдвое на необходимо
время и помещают бутыль под струю, больше не закрывая и снова не открывая кран.

ISO (the International Organization for Standardization) is a worldwide federation of national standards bodies

Introduction
Appropriate sampling is essential to provide representative samples to the laboratory in charge of testing.
Sampling features depend on the objective of sampling, but also on the nature of the sample. Microorganisms
are living organisms. In addition, when they are introduced into water, they do not form a perfect solution, but
a suspension with an inherent degree of variability.
Sampling objectives may serve different purposes, which are described in the ISO 5667 series of standards
(ISO 5667-1, ISO 5667-2 and ISO 5667-3):
a) determination of the compliance of a water with a regulatory quality specification;
b) characterization of any contamination, its level (mean) and its variations:
1) what is its random variation?
2) is there a trend?
3) are there cycles?
c) identification of the sources of pollution.
Regarding the number or frequency of samples, it will vary according to the aim of the sampling.
The minimum number of samples will be low if the mean concentration differs greatly from the specification
(much lower or much higher), and the minimum number of samples will be higher if the mean concentration
and the specification are close to one another. Similarly, in case b), when looking for a trend: the less obvious
the trend, the higher the frequency of sampling (see also Annex A).

1 Scope
This International Standard provides guidance on planning water sampling regimes, on sampling procedures
for microbiological analysis and on transport, handling and storage of samples until analysis begins. It focuses
on sampling for microbiological investigations.
General information in respect to the sampling from distinct water bodies is given in the respective parts of
ISO 5667.

3 Sampling point
The sampling site shall provide representative characteristics and account for any vertical, horizontal and
temporal variations and shall be identified precisely following the general recommendations of ISO 5667-1 and
ISO 5667-2, taking into account additional aspects specific to microbiology.
Sampling points where conditions are unstable should be avoided, and the heterogeneity of the hydraulic
system shall be taken into consideration. In studies on the efficacy of disinfection, the sampling point shall be
chosen to ensure that the reaction is complete.
EXAMPLE Examples of how the heterogeneity of the system may influence the results are given below.
⎯ It is not equivalent to take a subsurface or a surface sample, or a subsurface sample “contaminated” during recovery
through the surface film. In some instances (e.g. lakes, swimming pools), the concentration in the surface film can be

⎯ All the points of a network are not equivalent, as there may be dead ends and sections where the flow is reduced,
particularly if the network is fed from two sources.
⎯ The quality at the outlet of a well-mixed tank is generally the same as in the body of water, but can be quite different
from the inlet.
4 Sampling technique
4.1 Personnel
Formal training, training records and determination of competence shall be described for all those who sample,
and this information shall be properly documented.
4.2 Sample containers
4.2.1 General
For routine samples (for example, sampling at taps, recreational waters, swimming pool waters), use clean,
sterile bottles. The volume of the bottles should be adequate for analysis of all requested parameters.
For sampling by immersion in clean waters, use bottles that are sterile both inside and out and protected, for
example, by kraft paper (to keep dry after autoclaving), aluminium foil or by plastic outer bags.
If not autoclavable, sterilization with gamma rays or by ethylene oxide may be used. The bag can then be
opened just before sampling and can also serve as a glove to hold the bottle to provide maximum asepsis
before being placed on a pole or other sterilizable sampling apparatus.
Alternatively, the outside of sample bottles may be disinfected immediately prior to immersion by a suitable
disinfectant such as isopropanol (4.3.1.1) and allowed to dry before use. This is not suitable for analysis of
spore-forming bacteria.
In most cases, 500 ml bottles are sufficient, as less than five categories of microorganisms are measured,
each involving inoculation of a maximum of 100 ml.
In some cases, larger volumes are necessary, e.g.:
⎯ for bottled water analysis (250 ml per parameter);
⎯ for Legionella spp. or Salmonella spp. (up to 1 l);
⎯ for viruses, Giardia cysts, Cryptosporidium oocysts, amoebae in clean waters, from 10 to several hundred
litres or more are examined. Usually, a concentration step is made on site using a cartridge filter which is
then transported to the laboratory.
Bottles can be made of glass or various plastics (polypropylene, polystyrene, polyethylene, polycarbonate).
Usually glass is preferred for re-use, and polyethylene is used as disposable.
Adhesion to surfaces can lower the detection of microorganisms, and the critical tangential surface tension γ
[13]
has to be considered if a non-standard material is used .
Closures can be a ground glass or plastic stopper for glass bottles, a plastic press-on lid for plastic bottles or
jars, or a plastic or metal screw cap for either. Bottle openings closed with plastic or glass stoppers should be
further protected from contamination by, e.g. aluminium foil.
When larger volumes are necessary for the assay of, for example, viruses, Salmonella spp., amoebae,
Cryptosporidium oocysts, Giardia cysts, it is sometimes necessary to analyse tens of litres or hundreds of

litres. To avoid the difficulties of handling, refrigerating and agitating such volumes, a concentration step in situ
(by flocculation, centrifugation or filtration) is recommended. Peristaltic pumps can be used with sterile tubing.
NOTE 1 Metal caps, especially aluminium, can produce toxicity when autoclaved. This can be prevented by
incorporating a heat-resistant leak-proof liner.
NOTE 2 Certain materials can also give toxic by-products when heat sterilized, even in a dry oven, or induce pH
changes.
NOTE 3 Some brands of cotton wool used to make plugs for glassware may become toxic if they are heated for too
long at too high temperatures.
NOTE 4 Press-on plastic lids attached to the bottle or jar have several advantages in that they are as leak-proof as
screw-caps, and the lids can stand open, which facilitates filling and pipetting. When open, the lid remains linked to the
bottle, so bottles and closures are kept together, and the lid is also protected from contamination.
4.2.2 Sterilization of bottles
If re-used, clean glass bottles and their closures with a non-toxic, phosphorus-free detergent, followed by a
thorough rinse with deionized or distilled water.
Autoclave bottles at (121 ± 3) °C for at least 15 min. Keep the closure of the bottles loose, to allow the steam
to replace all the air during the temperature rise, and to prevent plastic bottles from collapsing when cooling.
Tighten screw caps after sterilization. Autoclave glass stoppers separately from the bottle, or use a paper or
aluminium separator to prevent the stopper sticking on cooling.
If necessary, sterilize bottles in a dry oven for at least 1 h at (170 ± 10) °C. Separate ground glass stoppers
from the neck by a paper strip or a piece of string to avoid jamming during cooling. The bottles should be
traceable to the sterilization date.
Control the effectiveness of the sterilization process by chemical or biological indicators.
When sterilization is not possible with any other means, disinfect by immersing open bottles in boiling water
for at least 30 min. Immediately after boiling, empty the bottles and close them with boiled caps and wrapped
in clean paper.
NOTE 1 Polyethylene bottles can be sterilized by exposure to ethylene oxide gas, but, because of its toxicity, the
procedure is carried out in specialized facilities and time allowed for desorption of the ethylene oxide. It is therefore not
used as a routine laboratory procedure.

(1 × 10 Gy to 2 × 10 Gy) is a very efficient sterilization technique, available in specialized installations. There is no
residual antibacterial activity, but some materials may be altered by polymerization after repeated irradiation.
4.2.3 Inactivation of disinfectants
To assess the microbiological quality of water disinfected by an oxidant (e.g. chlorine, chloramine, bromine or
ozone), stop the action of the oxidant as soon as the sample is taken. Add a reducing agent such as sodium
thiosulfate to the sample bottles.
The theoretical mass of sodium thiosulfate (pentahydrate) necessary to inactivate 1 mg of chlorine is 7,1 mg.
Thus, 0,1 ml of sodium thiosulfate pentahydrate solution (4.3.1.2) is added for each 100 ml of bottle capacity.
This will inactivate at least 2 mg/l and up to 5 mg/l of free chlorine residual, depending on inactivation
dynamics, which is sufficient for the majority of samples.
In certain circumstances, such as foot baths in swimming pools, disinfection measures (e.g. Legionella
eradication in drinking water distribution systems), higher chlorine concentrations can be found and a
proportionately higher dosage of sodium thiosulfate will be necessary.

Sodium thiosulfate is not destroyed by autoclaving or dry heat. Ensure that the pH of the sodium thiosulfate
solution is around neutral (low pH can cause decomposition).
Sodium thiosulfate has no effect on the sample and can be used for non-chlorinated waters too.
NOTE It has been claimed that Legionella are sensitive to sodium and that potassium thiosulfate is preferable, but no
adverse effect of sodium has been detected at the concentration used to inactivate usual chlorine concentrations.
For other disinfectants, corresponding inactivation measures need to be taken. If inactivation is not possible or
feasible, it has to be reported.
Chelating agents have been recommended to protect bacteria from the toxic action of heavy metals such as
copper or zinc. Ethylene dinitrilotetraacetic acid (EDTA) or sodium nitrilotriacetate (NTA) (Na C H NO ) can

be used as a filter-sterilized solution at a final concentration of about 50 mg per litre but should only be added
when necessary (e.g. water treated with silver or copper). Silver can also be inactivated by sodium sulfide.
Add 1 ml of a sodium sulfide solution (4.3.1.3) to 1 l of sample.
4.2.4 Quality control of sample bottles
4.2.4.1 Testing of sterility
The laboratory shall ensure the sterility of the sample bottles, whether they are prepared in-house or
commercially, whether they are made of glass or of plastic. Commercially prepared bottles should be
delivered with a certificate of sterilization as a condition for acceptance, and sterility tests are also advisable
on the batch in use. This relates to the batch of bottles after labelling, addition of inactivation agents where
relevant, and storage.
The sterility of bottles can usually be guaranteed by control of the sterilization process. If not, the sterility of
the containers should be tested.
EXAMPLE The following are examples of testing procedures (usually performed a rate of 1 per 100 bottles):
a) “Roll bottle” method
This consists of introducing 20 ml or 50 ml of melted nutrient agar (plate count agar) into the test bottle and lining the
walls with agar by rotating the bottle while cooling (under a trickle of water if necessary). Incubation at (22 ± 2) °C for
five days should give no visible growth.
b) Liquid broth method
This consists of placing 20 ml to 50 ml of thioglycollate or other nutrient broth inside the bottle, rolling the bottle to wet
the walls and incubating at (22 ± 2) °C for five days. No turbidity should appear if sterile.
4.2.4.2 Testing for the presence of inactivating agents
The presence of thiosulfate may be checked by an iodometric method:

Add 10 ml distilled water to the bottle and titrate with iodine solution (4.3.1.4), using starch or thiophene as an
end point titration agent.
4.2.4.3 Testing for residual toxicity in sample bottles
Residual toxicity in sample containers may result from the washing procedure of glassware, from the release
of components or additives from plastic bottles and also from the sterilization process. Routine use of glass or
polyethylene bottles does not require a regular check for toxicity, but if in any doubt, test according to
[8]
Geldreich, 1975 (for example).

4.4 Filling procedure
4.4.1 Potable water from a tap
4.4.1.1 General
Sampling at a tap can have different purposes:
a) to determine the quality of the water in the distribution main (which is the responsibility of the distributor);
b) to know the quality of the water as it flows from the tap to be consumed — as it is delivered to the tap —
(which can be altered by the service network inside the building);
c) to know the quality of the water as it is consumed, i.e. as it flows out of the (possibly contaminated) tap.
Samples to assess the quality in the main [case a)] are best taken at special taps (also in the distribution
system) that are close to the main distribution, clean, without attachments and disinfectable by flaming or
suitable equivalent.
Normal taps may be used to assess the quality in the mains [still case a)], if they are disinfectable by flaming
but in case of unclear results, consider the service network as potential source of contamination.
The situation described in case b) is the method of choice to assess the quality of drinking water including the
influence of the service network inside the building. In this case, taps disinfectable by flaming are not always
available and other disinfection methods [application of hypochlorite solution, ethanol or isopropanol (4.3.1.1)]
need to be considered.
The situation described in case c) is the method to assess the quality of drinking water in special situations,
e.g. outbreaks.
Depending on the purpose, it is either necessary or incorrect to:
⎯ remove attached devices and inserts;
⎯ disinfect the tap;
⎯ flush (see Table 1).

Sampling from drinking water storage tanks is usually made from a tap on the outlet. Subsurface samples are
sometimes taken from the tank itself, in which case, bottles that are sterile both inside and outside (see 4.2.1)
are required.
Ensure samples are taken aseptically using clean hands or sterile gloves with protection of the sample from
air drifts and splashing.

During filling, the inside of the bottle closure shall not come in contact with anything (fingers, ground, pocket,
teeth).
Leave some air space in the bottle to allow for adequate shaking before analysis.
Close the bottle immediately. Do not use this water sample for the measurement of temperature or any other
on-site tested parameter.

4.4.1.2 Water in treatment works and storage tanks
In water treatment works and storage tanks, dedicated sample taps should be provided on each outlet main
and other sampling points. These should be capable of being sterilized by flaming, maintained in a clean state,
labelled clearly and used exclusively for sampling. For details, see ISO 5667-5 and ISO 5667-13.
Flaming a metal tap intensely with a blowlamp ensures disinfection of the mouth if the temperature there
reaches 80 °C or more. This is not the case if water remains in the heated portion.
NOTE Flaming with a lighter is only superficial (not sufficient).
4.4.1.3 Water in the distribution main
To determine the quality in the distribution main, sample in the distribution main or close to it (usually just after
the water meter). Ensure that no contamination from the outer surface of the tap reaches the sample. Do not
sample taps with leaking spindles and avoid mixer taps, if possible. Take out any faucet nozzle or other
attachment or insert (spanners and pliers shall be available). Scrape off any dirty (scale, slime, grease or
other extraneous matter) and fully open and close the tap repeatedly to rinse out the dirt from the tap.
Disinfect the tap preferentially by flaming (after flaming and opening the tap, a sizzling noise should occur).
Subsequently, open the tap to half-flow and flush until constant water temperature is reached (to overcome
the water inside the building). Then place the open bottle in the water flow and fill it under aseptic conditions.
Only if flaming is not possible, disinfect the tap by other adequate methods. To disinfect the mouth of a plastic
tap, after thorough cleaning, dip it for 2 min to 3 min in a beaker with hypochlorite solution, ethanol or
isopropanol (4.3.1.1). Alternatively, a swab or a wash bottle or similar device may be used to disinfect the
outside and as much of the inside as possible.
Subsequently, allow the water to flow long enough to minimize the influence of the network inside the building.
It is necessary to know the detailed layout of the network (volume of tanks or softeners and retention time) to
determine the flush time before sampling. Water temperature stabilization may be monitored to achieve the
same effect for the same purpose. Many water microorganisms result from disruption of biofilm and
resuspension of deposits from joints or knees in case of peak flows and pressure jars. To minimize these
effects, open the tap at maximum flow for 5 s to 10 s, then reduce to half flow for the time needed, and place
the bottle under the tap without closing and re-opening the tap.
4.4.1.4 Water as it is delivered to the consumers tap
To determine the quality as it is delivered to the consumer's tap, ensure that no contamination from the outer
surface of the tap reaches the sample. Scrape off any dirt (scale, slime, grease or other extraneous matter)
which could fall off, before filling the bottles. Do not sample taps with leaking spindles. Take out any faucet
nozzle or other attachment or insert (spanners and pliers shall be available). Disinfect the tap preferentially by
flaming or, if not possible, by other adequate methods (see above). Subsequently, allow the water to flow just
long enough to ensure that the sample has no residual thermal or disinfectant effect. Place the bottle under
the tap without closing and re-opening the tap.

4.4.1.5 Water as it is consumed
To determine the quality of the water as it is consumed (e.g. in outbreak situations), contamination of the
water by bacteria from the outside of the tap and from any attachment or device should be taken into
consideration. Therefore, attachments should be kept in place and the tap should not be disinfected prior to
sampling.
4.4.2 Water from springs and wells
The analysis of well water can have different purposes:
1) to know the quality of the ground water;
2) to know the quality of the well water;
3) to know the quality of the water as it is used.
Depending on the purpose, different sampling types need to be chosen, where it has to be differentiated
between boreholes or wells in which pumps are permanently installed and boreholes or wells without
permanently installed pumps. Table 2 and Table 3 give an overview on the kind of sampling method which
should be chosen depending on the purpose in wells with and without a permanently installed pumping device.

Boreholes or wells in which a pump is permanently installed, usually also have a metal tap or outlet.
Depending on the purpose of sampling, extensive pumping is necessary or not, and disinfection of the tap,
preferentially by flaming, is necessary or not, see Table 2.
Extensive pumping for purpose 1 means that the pumping should last until constant water temperature and
electric conductivity are reached or at least a three times renewal of the well volume is guaranteed. For
purpose 2 (to know the quality of the well water) only a short water exchange is necessary, just to overcome

Introduction
Un échantillonnage approprié est essentiel pour fournir des échantillons représentatifs au laboratoire
d'analyse. Les modalités de l'échantillonnage dépendront de l'objectif de celui-ci, mais aussi de la nature des
échantillons. Les micro-organismes sont des organismes vivants. De plus, lors de leur introduction dans l'eau,
ils ne forment pas une solution parfaite mais une suspension ayant son propre degré d'hétérogénéité.
Les objectifs d'échantillonnage peuvent être multiples et ceux-ci sont décrits dans la série de normes
ISO 5667 (l'ISO 5667-1, l'ISO 5667-2 et l'ISO 5667-3):
a) déterminer la conformité d'une eau à une spécification réglementaire de qualité;
b) caractériser toute contamination, son niveau (moyen) et ses variations:
1) quelle est sa variation aléatoire?
2) existe-t-il une tendance?
3) existe-t-il des cycles?
c) identifier les sources de pollution.
Le nombre ou la fréquence des échantillons variera en fonction de l'objectif du prélèvement.
Le nombre minimal d'échantillons sera faible si la concentration moyenne diffère considérablement de la
spécification (très inférieure ou très supérieure), et le nombre minimal d'échantillons sera plus élevé si la
concentration moyenne est proche de la spécification. Il en est de même dans le cas b) lorsqu'il s'agit d'une
tendance: la fréquence de prélèvement est d'autant plus élevée que la tendance n'apparaît pas clairement
(voir aussi l'Annexe A).

1 Domaine d'application
La présente Norme internationale fournit des conseils sur les régimes de planification du prélèvement d'eau,
sur les modes opératoires de prélèvement en vue de l'analyse microbiologique et le transport, et sur la
manipulation et la conservation des échantillons avant le début de l'analyse. Elle porte sur les prélèvements
pour recherches microbiologiques.
Des informations générales sur l'échantillonnage à partir de différentes masses d'eau sont données dans les
parties respectives de l'ISO 5667.

3 Point de prélèvement
Le site de prélèvement doit présenter des caractéristiques représentatives et rendre compte de toute variation
verticale, horizontale et temporelle. Il doit être identifié avec précision, selon les recommandations générales
fournies dans l'ISO 5667-1 et l'ISO 5667-2, en tenant compte des aspects supplémentaires spécifiques à la
microbiologie.
Il convient d'éviter les points de prélèvement pour lesquels les conditions ne sont pas stables. L'hétérogénéité
du système hydraulique doit être prise en considération. Dans le cadre d'études portant sur l'efficacité de la
désinfection, le point de prélèvement doit être choisi de sorte à garantir que la réaction soit complète.

EXEMPLE Des exemples de l'influence de l'hétérogénéité du système sur les résultats sont donnés ci-dessous.
⎯ Il n'est pas équivalent de prélever un échantillon de subsurface ou de surface, ou un échantillon de subsurface
«contaminé» lors de la remontée à travers le film superficiel. Dans certains cas (par exemple lacs, piscines), la
concentration dans le film superficiel peut être 1 000 fois supérieure à celle en subsurface.
⎯ Tous les points d'un réseau ne sont pas équivalents car il peut exister des culs-de-sac et des sections où le débit est
réduit, notamment si le réseau est alimenté par deux sources.
⎯ La qualité de l'eau à la sortie d'un réservoir bien mélangé est généralement la même qu'au sein de la masse d'eau
mais elle peut varier considérablement par rapport à l'entrée du réservoir.
4 Technique de prélèvement
4.1 Personnel
La formation formelle, les dossiers de formation et la détermination des compétences doivent être décrits pour
tout personnel en charge des prélèvements et cette information doit être correctement documentée.
4.2 Flacons de prélèvement
4.2.1 Généralités
Pour les prélèvements de routine (par exemple prélèvements à des robinets, dans les eaux pour activités de
loisir, dans les eaux de piscine) utiliser des flacons propres et stériles. Il convient que le volume des flacons
soit adapté à l'analyse de tous les paramètres demandés.
Pour le prélèvement par immersion dans des eaux propres, utiliser des flacons stériles à l'intérieur et à
l'extérieur, protégés par du papier kraft (à maintenir au sec après le passage à l'autoclave), du papier
d'aluminium ou d'un sachet extérieur en plastique.
Si ce sachet ne peut être passé à l'autoclave, la stérilisation devra se faire au moyen de rayons gamma ou à
l'oxyde d'éthylène. Le sachet peut ensuite être ouvert juste avant le prélèvement et peut également servir de
gant pour tenir le flacon et garantir des conditions d'asepsie maximales avant de le placer sur une perche ou
autre appareil de prélèvement stérilisable.
Il est également possible de désinfecter la paroi extérieure des flacons de prélèvement immédiatement avant
l'immersion au moyen d'un désinfectant approprié, tel que l'isopropanol (4.3.1.1) et de les laisser sécher avant
utilisation. Ceci n'est pas adapté à l'analyse de bactéries sporulées.
Dans la plupart des cas, des flacons de 500 ml sont suffisants dans la mesure où moins de cinq catégories de
micro-organismes sont mesurées, impliquant chacune l'ensemencement d'un volume maximal de 100 ml.
Dans certains cas, des volumes plus importants sont nécessaires, par exemple:
⎯ pour l'analyse d'eau embouteillée (250 ml par paramètre);
⎯ pour les Legionella spp. ou Salmonella spp. (jusqu'à 1 l);
⎯ pour les virus, les kystes de Giardia, les oocystes de Cryptosporidium, les amibes dans les eaux propres,
de 10 à plusieurs centaines de litres ou plus sont examinés. En général, une étape de concentration est
réalisée sur le site au moyen d'un filtre à cartouche qui est ensuite transporté au laboratoire.
Les flacons peuvent être en verre ou en différentes matières plastiques (polypropylène, polystyrène,
polyéthylène, polycarbonate). Généralement, le verre est préféré en cas de réutilisation, et le polyéthylène est
utilisé pour un usage unique.

L'adhérence aux surfaces peut diminuer la détection de micro-organismes, et la tension de surface

tangentielle critique γ doit être pris en considération si un matériau non standard est utilisé .
Le dispositif de fermeture peut être un bouchon en verre dépoli ou en plastique pour les flacons en verre, une
cape à pression en plastique pour les flacons ou bocaux en plastique, ou une capsule à vis métallique ou
plastique pour l'un ou l'autre. Il convient que les ouvertures des flacons fermés par des bouchons en plastique
ou en verre disposent aussi d'une autre protection contre la contamination, par exemple du papier
d'aluminium.
Quand de grands volumes sont nécessaires pour la recherche, par exemple de virus, de Salmonella spp.,
d'amibes, d'oocystes de Cryptosporidium, de kystes de Giardia, il est parfois nécessaire d'analyser des
dizaines de litres ou des centaines de litres. Pour pallier les difficultés de manipulation, de réfrigération et
d'agitation de tels volumes, il est recommandé de procéder à une étape de concentration in situ (par
floculation, centrifugation ou filtration). Des pompes péristaltiques peuvent être utilisées avec des tubulures
stériles.
NOTE 1 Les capsules en métal, notamment celles en aluminium, peuvent libérer de la toxicité lorsqu'elles sont
autoclavées. Ceci peut être évité si elles incorporent un revêtement étanche thermorésistant.
NOTE 2 Certains matériaux peuvent également entraîner la formation de sous-produits toxiques lorsqu'ils sont
stérilisés à chaud, même au four, ou induire des variations de pH.
NOTE 3 Certaines marques de coton utilisées pour boucher la verrerie peuvent devenir toxiques si elles sont
chauffées trop longtemps à des températures trop élevées.
NOTE 4 Les capes à pression en plastique fixées aux flacons ou bocaux présentent plusieurs avantages compte tenu
du fait qu'elles sont aussi étanches que les capsules à vis, et que les capes peuvent rester ouvertes, ce qui facilite le
remplissage et le pipetage. Lorsque la cape est ouverte, elle reste attachée au flacon de sorte que les flacons et les
dispositifs de fermeture sont maintenus ensemble et la cape est également protégée contre la contamination.
4.2.2 Stérilisation des flacons
En cas de réutilisation, nettoyer les flacons en verre et leur dispositif de fermeture à l'aide d'un détergent non
toxique, exempt de phosphore, puis les rincer soigneusement avec de l'eau déionisée ou distillée.
Passer les flacons à l'autoclave à (121 ± 3) °C pendant au moins 15 min. Laisser le dispositif de fermeture
des flacons desserré pour permettre à la vapeur de remplacer la totalité de l'air contenu pendant la montée en
température, et pour éviter que les flacons en plastique s'écrasent pendant le refroidissement. Serrer les
capsules à vis après la stérilisation. Passer à l'autoclave les bouchons en verre séparément du flacon ou
utiliser un séparateur en papier ou en aluminium pour éviter le coincement du bouchon lors du refroidissement.
Si besoin est, stériliser les flacons dans un four pendant au moins 1 h à (170 ± 10) °C. Placer une bande de
papier ou un bout de ficelle entre le bouchon en verre dépoli et le goulot du flacon pour éviter qu'ils se
coincent pendant le refroidissement. Pour les flacons, il convient qu'il y ait une traçabilité de la date de
stérilisation.
Contrôler l'efficacité du procédé de stérilisation à l'aide d'indicateurs chimiques ou biologiques.
Lorsqu'aucune autre méthode de stérilisation n'est possible, désinfecter les flacons en les immergeant dans
de l'eau bouillante pendant au moins 30 min. Immédiatement après l'ébullition, vider les flacons et les fermer
avec des capsules bouillies emballées dans du papier propre.
NOTE 1 Les flacons en polyéthylène peuvent être stérilisés par exposition au gaz oxyde d'éthylène mais, en raison de
sa toxicité, ce mode opératoire est réalisé dans des installations spécialisées et du temps est donné pour la désorption de
l'oxyde d'éthylène. En conséquence, il n'est pas utilisé comme mode opératoire de routine dans les laboratoires.

accélérés d'énergie suffisante (de 1 × 10 Gy à 2 × 10 Gy) sont des techniques de stérilisation très efficaces, disponibles
dans des installations spécialisées. Toute activité antibactérienne résiduelle est éliminée, en revanche certains matériaux
peuvent être altérés par polymérisation après une exposition répétée aux rayonnements.

4.2.3 Neutralisation des désinfectants
Pour évaluer la qualité microbiologique de l'eau désinfectée par un oxydant (par exemple chlore, chloramines,
brome ou ozone), l'action de l'oxydant doit être interrompue dès que l'échantillon est prélevé. Ajouter un agent
réducteur, tel que le thiosulfate de sodium, dans les flacons de prélèvement.
La masse théorique du thiosulfate de sodium (anhydre) nécessaire pour neutraliser 1 mg de chlore est de
7,1 mg. En conséquence, on ajoute 0,1 ml d'une solution de thiosulfate de sodium pentahydraté (4.3.1.2) pour
chaque 100 ml contenu dans le flacon. Ceci neutralisera au minimum 2 mg/l et jusqu'à 5 mg/l de chlore libre,
en fonction des dynamiques de neutralisation, ce qui est suffisant pour la majorité des échantillons.
Dans certaines circonstances, telles que pédiluves dans les piscines, mesures de désinfection (par exemple
éradication des Legionella dans les réseaux de distribution d'eau potable), les concentrations de chlore
peuvent être supérieures, auquel cas un dosage proportionnellement plus élevé de thiosulfate de sodium sera
nécessaire.
Le thiosulfate de sodium n'est pas détruit par le passage à l'autoclave ni par la chaleur sèche. Veiller à ce que
le pH de la solution de thiosulfate de sodium soit presque neutre (un pH bas peut entraîner une
décomposition).
Le thiosulfate de sodium n'a pas d'effet sur l'échantillon et peut également être utilisé pour les eaux non
chlorées.
NOTE Il a été déclaré que les Legionella étaient sensibles au sodium et qu'il était préférable d'utiliser du thiosulfate
de potassium. Toutefois, aucun effet néfaste du sodium n'a été détecté aux concentrations utilisées pour neutraliser des
concentrations habituelles de chlore.
Pour d'autres désinfectants, des mesures de neutralisation correspondantes doivent être prises. Si la
neutralisation n'est pas possible ou réalisable, ceci doit être consigné.
Des agents chélatants ont été recommandés pour protéger les bactéries contre l'action toxique de métaux
lourds tels que le cuivre ou le zinc. L'acide éthylène diamine tétracétique (EDTA) ou le nitrilotriacétate de
sodium (NTA) (Na C H NO ) peuvent être utilisés en solution stérilisée par filtration à une concentration finale

d'environ 50 mg/l mais il convient de ne l'ajouter qu'en cas de nécessité (par exemple eau traitée à l'argent ou
au cuivre). L'argent peut également être neutralisé par du sulfure de sodium. Ajouter 1 ml d'une solution de
sulfure de sodium (4.3.1.3) dans 1 l d'échantillon.
4.2.4 Contrôle de la qualité des flacons de prélèvement
4.2.4.1 Essais de stérilité
Le laboratoire doit s'assurer de la stérilité des flacons de prélèvement, qu'ils soient préparés en interne ou
dans le commerce, qu'ils soient en verre ou en plastique. Il convient que les flacons préparés dans le
commerce soient accompagnés d'un certificat de stérilisation, condition de leur acceptation. Il est également
recommandé de réaliser des essais de stérilité sur le lot en usage. Ceci correspond au lot de flacons après
étiquetage, ajout d'agents neutralisants le cas échéant et stockage.
La stérilité des flacons peut généralement être garantie par le contrôle du procédé de stérilisation. Dans le cas
contraire, il convient de soumettre les flacons à des essais de stérilité.
EXEMPLE Des exemples de modes opératoires d'essai de stérilité sont fournis ci-dessous (généralement réalisés
au taux de 1 pour 100 flacons):
a) Méthode du «flacon roulé»
Cette méthode consiste à introduire 20 ml ou 50 ml de gélose nutritive en fusion (gélose pour dénombrement en
boîte) dans le flacon d'essai et à recouvrir la paroi du flacon avec la gélose en le faisant tourner pendant la phase de
refroidissement (sous un mince filet d'eau, si nécessaire). Il convient que l'incubation à (22 ± 2) °C pendant 5 j ne
donne lieu à aucune croissance visible.

b) Méthode en bouillon liquide
Cette méthode consiste à placer entre 20 ml et 50 ml de bouillon au thioglycolate ou autre bouillon nutritif dans le
flacon en le faisant tourner de façon à mouiller les parois et à incuber à (22 ± 2) °C pendant 5 j. Il convient qu'aucune
turbidité n'apparaisse si le flacon est stérile.
4.2.4.2 Essai de présence d'agents neutralisants
La présence de thiosulfate peut être vérifiée par une méthode iodométrique:

Ajouter 10 ml d'eau distillée dans le flacon et titrer avec une solution d'iode (4.3.1.4), en utilisant de l'amidon
ou du thiofène comme agent de titrage pour la détermination du point d'équivalence.
4.2.4.3 Essai de toxicité résiduelle dans les flacons de prélèvement
La toxicité résiduelle dans les flacons de prélèvement peut provenir du mode opératoire de nettoyage de la
verrerie, du dégagement de composants ou d'additifs des flacons en plastique et également du procédé de
stérilisation. Pour une utilisation courante de flacons en verre ou en polyéthylène, il n'est pas requis de vérifier

régulièrement la toxicité mais, en cas de doute, procéder à l'essai conformément à Geldreich, 1975 (par
exemple).
4.3 Réactifs, appareillage et matériel
4.3.1 Réactifs

réfrigérants dans des véhicules.

4.3.2.9 Porte-flacon lesté ou équivalent, avec corde ou chaîne (de préférence en acier inoxydable, au
moins la partie inférieure).
4.3.2.10 Perche, longues pinces ou échantillonneurs adaptés aux différentes profondeurs.
4.3.2.11 Cartes, liste des points de prélèvement, formulaires de prélèvement.
4.3.2.12 Véhicules et papiers, carte d'identité ou autorisation.
4.3.2.13 Bottes étanches (de sécurité).
4.3.2.14 Appareillage de mesure du pH, du chlore, de l'oxygène dissous, de la conductivité.
4.3.2.15 Gants stériles.
4.4 Mode opératoire de remplissage
4.4.1 Prélèvement d'eau potable à un robinet
4.4.1.1 Généralités
Le prélèvement à un robinet peut avoir différents objectifs:
a) connaître la qualité de l'eau dans le réseau de distribution (qui relève de la responsabilité du distributeur);
b) connaître la qualité de l'eau telle qu'elle coule du robinet des consommateurs — eau telle qu'elle parvient
au robinet — (qui peut être altérée par le réseau de distribution à l'intérieur du bâtiment);
c) connaître la qualité de l'eau telle qu'elle est consommée, c'est-à-dire telle qu'elle coule du robinet
(potentiellement contaminé).
Les échantillons permettant d'évaluer la qualité de l'eau dans le réseau principal [cas a)] sont de préférence
prélevés au niveau de robinets spécifiques (également dans le système de distribution) qui sont situés au plus
près des conduites principales, sont propres, ne comportent aucun accessoire, etc. et peuvent être stérilisés
par flambage ou un procédé équivalent approprié.
Des robinets normaux peuvent être utilisés pour évaluer la qualité de l'eau dans les conduites principales
[toujours dans le cas a)] s'ils peuvent être désinfectés par flambage. Toutefois, si les résultats sont ambigus,
le réseau intérieur doit être considéré comme source potentielle de contamination.
Le cas b) correspond à la méthode de choix pour évaluer la qualité de l'eau potable, y compris l'influence du
réseau de distribution à l'intérieur du bâtiment. Dans ce cas, il se peut que les robinets ne puissent pas être
désinfectés par flambage et d'autres méthodes de désinfection [application de solution d’hypochlorite,
d’éthanol ou d'isopropanol (4.3.1.1)] doivent alors être envisagées.
Le cas c) est la méthode utilisée pour évaluer la qualité de l'eau potable dans des situations particulières, par
exemple en cas d'épidémie.
En fonction de l'objectif de l'analyse, il est nécessaire ou inapproprié de:
⎯ retirer les dispositifs accessoires ou les inserts;
⎯ désinfecter le robinet;
⎯ rincer (voir Tableau 1).

Rincer brièvement, uniquement pour annuler les effets de la désinfection du robinet.

Le prélèvement à partir de réservoirs de stockage d'eau potable est généralement réalisé à un robinet au
niveau de la sortie. Des échantillons de subsurface sont parfois prélevés dans le réservoir proprement dit,
auquel cas des flacons stériles à l'intérieur et à l'extérieur (voir 4.2.1) sont requis.
Veiller à ce que les échantillons soient prélevés aseptiquement avec des mains propres ou en utilisant des
gants stériles en protégeant l'échantillon contre les courants d'air et les éclaboussures.
Pendant le remplissage, le dispositif de fermeture du flacon ne doit entrer en contact avec aucun élément (doigts,
sol, poche, dents).
Laisser un volume d'air dans le flacon de sorte à permettre l'agitation appropriée avant l'analyse.
Fermer le flacon immédiatement. Ne pas utiliser cet échantillon d'eau pour la mesure de la température ou
autre paramètre mesuré sur site.

4.4.1.2 Eau dans les installations de traitement des eaux et réservoirs de stockage
Dans les usines de traitement des eaux et les réservoirs de stockage, il convient de prévoir des robinets de
prélèvement spécifiques sur chaque canalisation de sortie et autres points de prélèvement. Il convient que
ces robinets puissent être stérilisés par flambage, maintenus en état propre, étiquetés clairement et utilisés
exclusivement pour le prélèvement. Pour le détail, voir l'ISO 5667-5 et l'ISO 5667-13.
Le flambage intensif d'un robinet métallique avec un chalumeau garantit la désinfection de la sortie si la
température à cet endroit atteint 80 °C ou plus. Ceci n'est pas le cas si de l'eau reste dans la partie chauffée.
NOTE Le flambage au briquet n'est que superficiel (insuffisant).
4.4.1.3 Eau dans les conduites principales
Pour déterminer la qualité de l'eau dans les conduites principales, effectuer le prélèvement dans la conduite
principale ou à proximité de celle-ci (généralement juste après le compteur d'eau). Veiller à ce qu'aucune
contamination de la surface extérieure du robinet n'entre en contact avec l'échantillon. Ne pas prélever d'eau
à des robinets présentant des fuites à la manette et éviter si possible les robinets mélangeurs. Retirer tout
brise-jet ou autre accessoire ou insert (des clés et des pinces doivent être disponibles). Gratter toute salissure
[tartre, film biologique (mucus), graisse ou autre substance étrangère] et manœuvrer de façon répétée le
robinet en ouverture et fermeture totale afin de faire partir les
