Water quality — Determination of selenium — Part 2: Method using hydride generation atomic absorption spectrometry (HG-AAS)

ISO/TS 17379-2:2013 specifies a method for the determination of selenium. The method is applicable to drinking water, surface water, ground water, and rain water. The dynamic range of ISO/TS 17379-2:2013 is approximately 0,5 µg/l to 20 µg/l. Samples containing selenium at higher concentrations than the application range can be analysed following appropriate dilution. The method is unlikely to detect organoselenium compounds. The sensitivity of this method is dependent on the selected operating conditions. It is important to use high purity reagents in all cases with minimum levels of selenium.

Введение
Данная часть ISO/TS 17379 предназначена для применения аналитиками, имеющими опыт работы с
микропримесями в очень низких концентрациях.
Неорганический селен обычно встречается в форме двух окислов: Se(VI) и Se(IV). Важно
преобразовать все формы селена в Se(IV) перед образованием гидридов. Селен (VI) гидрида не дает.
В природных источниках воды соединения селена обычно встречаются в очень малых количествах,
менее 1 мкг/л. Можно обнаружить более высокие концентрации, например, в промышленных сточных
водах. Серен встречается в природе в органических соединениях и может иметь валентность –II, 0, IV,
и VI.
Чтобы полностью разложить все соединения селена, необходима процедура вываривания.
Вываривание можно опустить только в том случае, если определено, что селен в пробе может
образовывать ковалентный гидрид без необходимости предварительного окисления на стадии
вываривания.
Пользователю следует знать, что определенные проблемы могут потребовать задания
дополнительных граничных условий.

1 Область применения
Настоящая часть ISO/TS 17379 устанавливает метод определения селена. Этот метод применим к
питьевой воде, поверхностным, грунтовым и дождевым водам. Динамический диапазон применения
данной части ISO/TS 17379 составляет от 0,5 мкг/л до 20 мкг/л. Пробы, содержащие селен в более
высокой концентрации, чем применяемый диапазон, можно анализировать при соответствующем
разбавлении. Маловероятно обнаружить этим методом органоселеновые соединения.
Чувствительность данного метода зависит от выбранных рабочих условий.
Важно использовать реактивы высокой чистоты во всех случаях минимальной концентрации селена.

3 Сущность метода
Аликвотное количество пробы обрабатывают концентрированной соляной кислотой (5.2). Se(VI)
предварительно восстанавливают до Se(IV) осторожным кипячением с обратным холодильником в растворе
HCl концентрацией 6 моль/л в течение 1 ч. Необходимо следить за тем, чтобы не потерять летучие
соединения селена. Подходящее оборудование показано на Рисунке B.3. Растворы пробы затем
обрабатывают тетрагидроборатом натрия для получения ковалентного газообразного гидрида (SeH ). Этот

гидрид и избыток водорода удаляют из емкости, где генерируются гидриды, в периодическом режиме, а из
сепаратора газ/жидкость – в непрерывном режиме и подают в нагретую кварцевую ячейку. После атомизации
определяют оптическую плотность селена при длине волны, λ = 196,0 нм. Эта процедура автоматизирована с
помощью автодозатора и управляющей компьютерной программы.
Можно использовать другие способы измерения, при условии возможности продемонстрировать
критерии эффективности, которые равноценны или выше критериев эффективности лаборатории
пользователя (см. Приложение С).
4 Помехи
Техника получения гидридов подвержена помехам со стороны переходных и легко восстанавливаемых
металлов. Для большинства проб природной воды такой тип помех не должен быть значительным.
Пользователю следует осуществить определение степени извлечения на типичных водах, а также
определить максимальные концентрации потенциально мешающих элементов, пользуясь
подходящими методами. Если есть признаки таких помех, то следует оценить их уровень, измерив
степень извлечения.
Условия реакции, установленные в данной части ISO/TS 17379, подобраны таким образом, чтобы
сократить до минимума любые помехи.

Следующие элементы не вызывают помех, при условии, что не превышаются концентрации,
установленные в Таблице 1.

Если концентрации, указанные в Таблице 1 превышены, то может потребоваться применение метода
добавления стандарта (9.3). Предположив, что содержание селена достаточно велико,
предпочтительно разбавить пробу для анализа.

Металлы, которые легко восстанавливаются тетрагидроборатом натрия могут также вызвать помехи. В
частности, сюда входят хром, железо, медь, никель и свинец. Если концентрации этих элементов,
указанные в Таблице 2, превышены, то может возникнуть значительное снижение абсорбции.

ПРИМЕЧАНИЕ Если используются системы периодического действия, то массовые концентрации, которые
значительно ниже установленных в Таблице 1 и Таблице 2, могут тоже вызвать помехи.
5 Реактивы и стандарты
В ходе анализа, если нет иных указаний, пользуются только реактивами признанной аналитической кислоты.
Реактивы могут содержать селен в качестве примеси. Все реактивы должны иметь концентрацию
содержащегося в них селена ниже той, которая дает значение холостого опыта для метода, будучи
выше наиболее низкой представляющей интерес концентрации.
Реактивы необходимо готовить по инструкциям изготовителя, пользуясь в качестве примера
следующими реактивами.
5.1 Вода, соответствующая классу 1 по ISO 3696, для приготовления и разбавления проб.
5.2 Соляная кислота, ρ(HCl) = 1,16 г/мл.
5.3 Соляная кислота, c(HCl) = 1 моль/л.
5.4 Гидроксид натрия, NaOH.
5.5 Раствор тетрагидробората натрия, ρ(NaBH ) = 13 г/л.

Растворяют подходящий объем тетрагидробората натрия (13 г считается достаточным для системы,
показанной в Приложении B) в 500 мл воды (5.1) и добавляют (4,0 ± 0,1) г гидроксида натрия (5.4).
Доводят до 1000 мл водой (5.1). Готовят раствор в день использования и емкость с раствором не
закупоривают ввиду создания избыточного давления за счет выделения водорода.
ПРИМЕЧАНИЕ 1 Концентрация NaBH зависит от коллектора генератора гидридов и скорости потока. См.

5.6 Азотная кислота, ρ(HNO ) = 1,40 г/мл.

5.7 Холостой раствор
Для каждых 1 000 мл готовят раствор, содержащий (300 ± 3) мл соляной кислоты (5.2). Доводят до
объема водой (5.1).
При поточной системе холостой раствор проходит как фон. Поскольку холостой раствор может содержать
обнаружимые уровни примесей селена, важно пользоваться одними и теми же реактивами для приготовления
пробы и стандарта, а также для приготовления холостого раствора. Сигнал от аналита накладывается на этот
сигнал, как только пробу вводят в цикл измерения. Концентрация селена в холостом растворе должна быть
меньше нижней границы рассматриваемого диапазона концентраций.
5.8 Стандартные растворы селена.
5.8.1 Исходный раствор A селена, ρ[Se(IV)] = 1 000 мг/л.
Используют количественный исходный раствор с содержанием Se(IV) в размере (1 000 ± 2) мг/л.
Этот раствор считается устойчивым в течение не менее одного года.
ПРИМЕЧАНИЕ Можно использовать другие исходные растворы, при условии, что неопределенность
измерения не будет высокой.
Альтернативно, используют исходный раствор, приготовленный из химических веществ высокой чистоты.
Помещают 1,094 г селенита натрия (Na SeO ), просушенного в течение 2 ч при температуре (105 ± 3) °C в

мерной колбе вместимостью 500 мл.
Добавляют 200 мл воды (5.1) и 200 мл соляной кислоты (5.3) и полностью растворяют селенит натрия
при помешивании.
Доводят до 500 мл водой (5.1) и тщательно перемешивают.
5.8.2 Стандартный раствор селена B, ρ[Se(IV)] = 10 мг/л.
Пипеткой переносят (1 ± 0,01) мл исходного раствора селена A (5.8.1) в мерную колбу вместимостью
100 мл, добавляют (30 ± 0,5) мл соляной кислоты (5.2) и наполняют до метки водой (5.1).
Этот раствор стабилен в течение одного месяца.
5.8.3 Стандартный раствор селена C, ρ[Se(IV)] = 100 мкг/л.
Пипеткой переносят (1 ± 0,01) мл стандартного раствора селена B (5.8.2) в мерную колбу
вместимостью 100 мл, добавляют (30 ± 0,5) мл соляной кислоты (5.2) и наполняют до метки водой (5.1).
Этот раствор стабилен в течение одной недели.
5.8.4 Стандартный раствор селена D, ρ[Se(IV)] = 10 мкг/л.
Пипеткой переносят (10 ± 0,1) мл стандартного раствора селена C (5.8.3) в мерную колбу из
боросиликатного стекла вместимостью 100 мл. Наполняют до метки холостым раствором (5.7).
Этот раствор необходимо готовить в день использования.
5.8.5 Исходный раствор селена E, ρ[Se(VI)] = 1 000 мг/л.
Растворяют(2,392 ± 0,002) г селената натрия (Na SeO ) после сушки при температуре (105 ± 3) °С в

течение 2 ч в 500 мл воды (5.1), затем переносят раствор количественно в мерную колбу
вместимостью 1 000 мл и доводят до метки водой (5.1).

Этот стандарт используют для приготовления подходящего стандарта селена (VI) для проверки
количественного извлечения Se(VI).
Этот раствор стабилен не менее шести месяцев.
5.8.6 Градуировочные растворы селена.
Необходимо использовать не менее пяти независимых градуировочных растворов. Выполняют
градуировку в соответствии с ISO 8466-1.
Готовят не менее пяти градуировочных растворов из стандартного раствора селена С (5.8.3),
покрывающих весь рабочий диапазон ожидаемых концентраций селена.
Например, для диапазона концентраций от 1 мкг/л до 5 мкг/л, поступают следующим образом.
Пипеткой переносят в пять мерных колб вместимостью 100 мл объемы (1 ± 0,01) мл, (2 ± 0,02) мл, (3 ±
0,03) мл, (4 ± 0,04) мл и (5 ± 0,05) мл, соответственно, стандартного раствора селена С (5.8.3).
Добавляют 30 мл соляной кислоты (5.2).
Доводят до 100 мл водой (5.1) и тщательно перемешивают.
Эти градуировочные растворы будут содержать 1 мкг/л, 2 мкг/л, 3 мкг/л, 4 мкг/л, и 5 мкг/л селена
соответственно.
Их необходимо готовить в день использования.
Допускается применением пипеток с поршнем, что облегчает приготовление малых объемов
градуировочных растворов. Также допускается применение разбавителей.
После построения градуировочной кривой количество стандартов, используемых повседневно, можно
уменьшить. Любое подобное изменение не должно сказываться на результате, полученном в
испытаниях, или на порядке следования с другими пробами.
6 Аппаратура
Обычное лабораторное оборудование и, в частности, следующее.
Оборудование перечисляется для руководства. В общем, необходимо следовать инструкциям изготовителя.
6.1 Атомно-абсорбционный спектрометр.
6.1.1 Общие положения. Рекомендуется использовать атомно-абсорбционный спектрометр,
оснащенный системой образования гидридов атомайзером с нагретой осушающей трубкой
(предпочтительно оснащенный системой коррекции фона). Автоматические поточные системы [анализ
с вводом пробы в поток (FIA) или непрерывный анализ потока (CFA)] для образования гидридов
удобны и чаще используются, хотя системы периодического действия также пригодны. Следующий
пример показывает режим непрерывного потока.
6.1.2 Источник излучения для определения селена. Используют источник излучения, который
излучает при длине волны λ = 196,0 нм для селена с соответствующим спектральным излучением.
6.2 Подача газа. Используют аргон степени чистоты, установленной изготовителем.
Подачу газа следует оснастить двухступенчатым регулятором, а аргон подавать под давлением,
рекомендованным изготовителем.
Рекомендуется применять газоочиститель, состоящий из активированного угля.

Также можно использовать газообразный азот, но в результате чувствительность значительно снизится.
6.3 Лабораторная посуда.
6.3.1 Общие требования. Для определения селена при очень низких концентрациях критическим
вопросом является загрязнение и потери. Потенциальные источники загрязнения включают
нетщательно промытое лабораторное оборудование и общее загрязнение в рамках помещений
лаборатории. Для работы с пробой с микропримесями должно использоваться чистое рабочее
помещение лаборатории.
Перед применением всю посуду многократного использования, соприкасающуюся с пробой,
необходимо мыть.
Лабораторную посуду сначала необходимо замочить в моющей смеси с азотной кислотой (5.6), как
минимум, на 24 ч и пять раз промыть водой (5.1).
После этого, снова наполняют лабораторную посуду соляной кислотой, c(HCl) = 1 моль/л (5.3) и
оставляют на следующие 24 ч.
Пластиковая лабораторная посуда однократного применен6ия не требует специальной очистки при
условии, что в этом материале содержится пренебрежимо малое количество селена.
6.3.2 Склянки для хранения и обработки проб. Используют пробоотборные емкости из
боросиликатного стекла, кварца или пластмасс [например, из политетрафторэтилена (ПТФЭ),
фторированного этилен-пропилена (FEP)], которые не абсорбируют и не десорбируют аналит.
6.3.3 Резервуар прибора для реактивов. Реактивы подаются с помощью перистальтического
насоса из стеклянных склянок с реактивами по трубкам из ПТФЭ. Все трубки насоса должны быть
совместимы с используемыми реактивами и не должны адсорбировать или десорбировать аналит.
6.3.4 Виалы автодозатора. Пользуются виалами из полистирола или материалов, установленных в
(6.3.2).
6.4 Оборудование для обработки проб.
6.4.1 Пипетка с вытеснением воздуха. Пользуются системой микропипетки, обеспечивающей
подачу объемов от 10 мкл до 1 000 мкл с ассортиментом не содержащих металлов наконечников
однократного применения.
6.4.2 Весы. Аналитические весы, обеспечивающие точное взвешивание (стандартов) до ±0,1 мг; и
весы с чашкой, установленной сверху, для приготовления растворов, точностью до ±0,1 г.
7 Отбор и подготовка проб
7.1 Техника пробоотбора
Пробоотбор осуществляют в соответствии с ISO 5667-1, ISO 5667-3, ISO 5667-5, ISO 5667-6,
ISO 5667-8, и ISO 5667-11, пользуясь емкостями, установленными в 6.3.2.
Для определения селена в пробах воды подкисляют воду до значения достаточно использовать соляную кислоту (5.2) (3 ± 0,1) мл на литр. Обеспечивают pH меньше 2; в
противном случае добавляют еще соляной кислоты по мере необходимости.
Для всех типов проб соответствующим образом готовят и анализируют холостую пробу. Используют
емкости одного типа и количество кислоты, которое использовалось в пробе.

7.2 Предварительное восстановление
7.2.1 Общие положения
Поскольку только Se(IV) быстро и количественно реагирует в используемых условиях получения
гидридов, Se(VI) необходимо восстановить до Se(IV) перед этапом образования гидридов.

Точно переносят аликвоту пробы (от 40 мл до 50 мл) в емкость вместимостью 100 мл.
Добавляют (30 ± 0,5) мл соляной кислоты (5.2).
Перемешивают и осторожно кипятят с обратным холодильником в течение 1 ч.
Переносят в мерную колбу и разбавляют до 100 мл водой (5.1).
Если применяются другие объемы проб, используют реактивы и оборудование, соответствующее
выбранным объемам.
Для большей точности пробу можно также отмерять по массе. В этом случае, объем вычисляют по
плотности и массе и записывают его.
7.2.3 Пробы, требующие дополнительного вываривания
Пробы, содержащие значительное количество твердых частиц и/или органически связанного селена,
требуют дополнительного этапа вываривания в соответствии с ISO 15587-1. Хотя этот вопрос выходит
за рамки данной части ISO/TS 17379, пробы можно анализировать с помощью аналогичной процедуры,
при условии приготовления надлежащим образом подходящей к реактивам матрицы, используя
правильную пропорцию азотной кислоты (5.6) и соляной кислоты (5.2). Холостые и стандартные

8 Установка приборов
Компонуют оборудование в соответствии с описанием в инструкциях изготовителя. Следующее
относится к проточным системам; в отношении приборов для анализа при вводе пробы в поток или
периодических анализаторов см. инструкции изготовителя.
Проверяют трубки на износ и надежность насоса каждый день использования системы и заменяют по
мере необходимости. Все трубки, соединяющие автодозатор, парогенератор и детектор, должны
сохраняться минимальной длины.
Наполняют резервуары для реактивов холостым раствором (5.7) и раствором тетрагидробората
натрия (5.5), соответственно.
Устанавливают систему поточного парогенератора в соответствии с инструкциями изготовителя.
Необходимо обеспечить поток реактивов в рамках принятых допусков. Как только установятся
стабильные условия, можно продолжать анализ. И стандарты и пробы должны измеряться
количественно при одинаковых реологических характеристиках.
Для проб, выпадающих за пределы градуировки для установки данного диапазона, разбавляют пробу
для попадания концентрации в диапазон градуировки и анализируют снова. Если пробу разбавляют, то
разбавителем необходимо брать холостой раствор (5.7), т.e. совпадающую матрицу.

растворы (холостой раствор (5.7), градуировочные растворы (5.8.6) и пробы (7.2) вводят в поточную
систему согласно инструкциям изготовителя и выполняют измерения.

Introduction
This part of ISO/TS 17379 is intended for use by analysts experienced with the handling of trace elements
at very low concentrations.
Inorganic selenium normally occurs in two oxidation states; Se(VI) and Se(IV). It is essential to convert all
selenium species to the Se(IV) state prior to generating the hydrides. Selenium(VI) does not form a hydride.
In natural water sources, selenium compounds generally occur in very small quantities, typically
less than 1 µg/l. Higher concentrations may be found, e.g. in industrial waste water. Selenium occurs
naturally in organic and inorganic compounds and may have oxidation states –II, 0, IV, and VI.
In order to fully decompose all of the selenium compounds, a digestion procedure is necessary. Digestion
can only be omitted if it is certain that the selenium in the sample can form a covalent hydride without
the necessity of a pre-oxidation digestion step.
The user should be aware that particular problems could require the specification of additional
marginal conditions.

1 Scope
This part of ISO/TS 17379 specifies a method for the determination of selenium. The method is
applicable to drinking water, surface water, ground water, and rain water. The dynamic range of this
part of ISO/TS 17379 is approximately 0,5 µg/l to 20 µg/l. Samples containing selenium at higher
concentrations than the application range can be analysed following appropriate dilution. The method
is unlikely to detect organoselenium compounds.
The sensitivity of this method is dependent on the selected operating conditions.
It is important to use high purity reagents in all cases with minimum levels of selenium.

ISO 15587-1, Water quality — Digestion for the determination of selected elements in water — Part 1: Aqua
regia digestion
3 Principle
An aliquot of sample is treated with concentrated hydrochloric acid (5.2). Se(VI) is pre-reduced to
Se(IV) by gently refluxing in 6 mol/l HCl for 1 h. Care is necessary to avoid any losses of volatile selenium
components. A suitable apparatus is shown in Figure B.3. The sample solutions are then treated with sodium
tetrahydroborate to generate the covalent gaseous hydride (SeH ). The hydride and excess hydrogen are

swept out of the generation vessel in the batch mode and out of the gas/liquid separator in the continuous
mode into a heated silica cell. After atomization the absorbance of selenium is determined at a wavelength,
λ = 196,0 nm. The procedure is automated by means of an auto sampler and control software.
Other measurement techniques may be applicable provided that the performance criteria can be
adequately demonstrated or exceeded by the user laboratory (see Annex C).
4 Interferences
The hydride generation technique is prone to interferences by transition and easily reducible metals.
For the majority of natural water samples, this type of interference should not be significant. The user
should carry out recovery tests on typical waters and also determine the maximum concentrations of
potentially interfering elements, using appropriate methods. If such interferences are indicated, the
level of interferences should be assessed by performing spike recoveries.
The reaction conditions set out in this part of ISO/TS 17379 have been chosen so that any interferences
are reduced to a minimum.

These elements do not cause interferences provided the concentrations specified in Table 1 are not exceeded.

If the concentrations in Table 1 are exceeded, it may be necessary to use the standard addition method
(9.3). Assuming that the selenium content is high enough, an appropriate dilution of the water sample
is preferred.
Metals which are readily reduced by sodium tetrahydroborate may also cause interferences. In particular,
these include chromium, iron, copper, nickel, and lead. If the concentrations of these elements specified
in Table 2 are exceeded, a significant decrease of absorption may occur.

5 Reagents and standards
During the analysis, unless otherwise stated, use only reagents of recognized analytical grade.
Reagents may contain selenium as an impurity. All reagents shall have selenium concentrations below
that which would result in a selenium blank value for the method being above the lowest level of interest.
Reagents shall be prepared to manufacturer’s recommendations using the following series as an example.
5.1 Water, complying with grade 1 as defined in ISO 3696, for all sample preparation and dilutions.
5.2 Hydrochloric acid, ρ(HCl) = 1,16 g/ml.
5.3 Hydrochloric acid, c(HCl) = 1 mol/l.
5.4 Sodium hydroxide, NaOH.
5.5 Sodium tetrahydroborate solution, ρ(NaBH ) = 13 g/l.

Dissolve an appropriate amount of sodium tetrahydroborate (13 g has proven suitable for the system
illustrated in Annex B) in 500 ml water (5.1) and add (4,0 ± 0,1) g of sodium hydroxide (5.4). Dilute
to 1 000 ml with water (5.1). Prepare on day of use and do not keep in a closed container because of
pressure build-up due to hydrogen evolution.
NOTE 1 The concentration of NaBH is dependent on the hydride generator manifold and flow rate conditions.

Prepare a nitric acid cleaning mixture by diluting nitric acid [ρ(HNO ) = 1,40 g/ml] with an equal volume

of water (5.1) by carefully adding the acid to the water.
5.7 Reagent blank.
For each 1 000 ml, prepare a solution containing (300 ± 3) ml of hydrochloric acid (5.2). Dilute to volume
with water (5.1).
On the continuous flow system, the reagent blank solution is run as background. Since the blank solution
may contain detectable trace levels of selenium, it is important that the same reagents be used for both
sample and standard preparation, as well as for preparation of the reagent blank. The analyte signal is
superimposed on this signal once the sample is introduced into the measurement cycle. The selenium
concentration of the blank solution should be less than the lower level of interest.

5.8 Selenium standard solutions.
5.8.1 Selenium stock solution A, ρ[Se(IV)] = 1 000 mg/l.
Use a quantitative stock solution with a selenium content of (1 000 ± 2) mg/l.
This solution is considered to be stable for at least one year.
NOTE Other stock solutions may be available and can be used providing the uncertainty of the measurement
is not compromised.
Alternatively, use a stock solution prepared from high purity grade chemicals:
Place 1,094 g of sodium selenite (Na SeO ) dried for 2 h at (105 ± 3) °C into a 500 ml volumetric flask.

Add 200 ml of water (5.1) and 200 ml of hydrochloric acid (5.3) and dissolve the sodium selenite
completely by stirring.
Dilute to 500 ml with water (5.1) and mix thoroughly.
5.8.2 Selenium standard solution B, ρ[Se(IV)] = 10 mg/l.
Pipette (1 ± 0,01) ml of selenium stock solution A (5.8.1) into a 100 ml volumetric flask, add (30 ± 0,5) ml
of hydrochloric acid (5.2) and fill up to the mark with water (5.1).
This solution is stable for one month.
5.8.3 Selenium standard solution C, ρ[Se(IV)] = 100 µg/l.
Pipette (1 ± 0,01) ml of selenium standard solution B (5.8.2) into a 100 ml volumetric flask, add (30 ±
0,5) ml of hydrochloric acid (5.2) and fill up to the mark with water (5.1).
This solution is stable for one week.
5.8.4 Selenium standard solution D, ρ[Se(IV)] = 10 µg/l.
Pipette (10 ± 0,1) ml of selenium standard solution C (5.8.3) into a 100 ml borosilicate volumetric flask.
Fill up to the mark with reagent blank solution (5.7).
This solution shall be prepared freshly on the day of use.
5.8.5 Selenium stock solution E, ρ[Se(VI)] = 1 000 mg/l.
Dissolve (2,392 ± 0,002) g of sodium selenate (Na SeO ) after drying at (105 ± 3) °C for 2 h in 500 ml

water (5.1), then transfer the solution quantitatively to a 1 000 ml volumetric flask and fill up to the
mark with water (5.1).
Use this standard to prepare a suitable selenium(VI) standard to check quantitative recovery of
selenium(IV).
The solution is stable for at least six months.
5.8.6 Selenium calibration solutions.
A minimum of five independent calibration solutions shall be used. Perform the calibration in accordance
with ISO 8466-1.
Prepare a minimum of five selenium calibration solutions from the selenium standard solution C (5.8.3)
covering the working range of expected selenium concentrations.
For the operating range from 1 µg/l to 5 µg/l, for example, proceed as follows.

Pipette into a series of five 100 ml volumetric flasks (1 ± 0,01) ml, (2 ± 0,02) ml, (3 ± 0,03) ml, (4 ±
0,04) ml, and (5 ± 0,05) ml, respectively, of selenium standard solution C (5.8.3).
Add 30 ml of hydrochloric acid (5.2).
Dilute to 100 ml with water (5.1) and mix thoroughly.
These calibration solutions contain 1 µg/l, 2 µg/l, 3 µg/l, 4 µg/l, and 5 µg/l selenium respectively.
They shall be prepared on the day of use.
The use of piston pipettes is permitted and enables the preparation of lower volumes of calibration
solutions. The application of dilutors is also allowed.
Once a calibration pattern has been established, the number of standards used routinely may be reduced.
Any such change shall not alter the result obtained from tests or the ranking with other samples.
6 Apparatus
Usual laboratory equipment and in particular the following.
The following are set out as guidelines. In general, the manufacturer’s instructions shall be followed.
6.1 Atomic absorption spectrometer.
6.1.1 General. An atomic absorption spectrometer equipped with a hydride generation system and
a heated silica tube atomizer or a graphite furnace atomizer (preferably equipped with a background
correction system) is recommended. Automated flow systems [flow injection analysis (FIA) or
continuous flow analysis (CFA)] for hydride generation are suitable and more usual but batch systems
are also adequate. The description below follows a continuous flow regime.
6.1.2 Radiation source for the determination of selenium. Use a radiation source which emits at
wavelength λ = 196,0 nm for selenium with an adequate spectral radiance.
6.2 Gas supply. Use argon with a grade specified by the manufacturer.
The gas supply shall be with a two stage regulator and the argon supplied at a pressure recommended
by the manufacturer.
The use of a gas purifier consisting of activated carbon is recommended.
Nitrogen gas may also be used but has reduced sensitivity.
6.3 Laboratory-ware.
6.3.1 General requirements. For the determination of selenium at very low concentrations,
contamination and loss are of critical consideration. Potential contamination sources include improperly
cleaned laboratory apparatus and general contamination within the laboratory environment. A clean
laboratory work area, designated for trace element sample handling shall be used.
All re-usable laboratory-ware in contact with the sample shall be cleaned prior to use.
Laboratory-ware shall be soaked in the nitric acid cleaning mixture (5.6) for at least 24 h and rinsed five
times with water (5.1).
Following this, refill laboratory-ware with hydrochloric acid, c(HCl) = 1 mol/l (5.3) and leave for 24 h.
Disposable (single-use) plastics laboratory-ware does not require special cleaning, provided that
negligible selenium contamination in that material is demonstrated.

6.3.2 Storage and sample processing bottles. Use sampling vessels constructed of silica, borosilicate
glass or suitable plastics materials] e.g. polytetrafluorethylene (PTFE), perfluoro (ethene-propene)
(FEP)] that neither absorb nor desorb the analyte under test.
6.3.3 Instrument reagent reservoir. The reagents are delivered via a peristaltic pump from glass
reagent bottles through PTFE transfer lines. All pump tubing shall be compatible with reagents in use
and neither adsorb nor desorb the analyte under test.
6.3.4 Auto-sampler vials. Use vials of polystyrene or materials specified in 6.3.2.
6.4 Sample processing equipment.
6.4.1 Air displacement pipette. Use a micropipette system capable of delivering volumes from 10 µl
to 1 000 µl with an assortment of metal-free, disposable pipette tips.
6.4.2 Balances. Analytical balance, capable of accurately weighing (standards) to ±0,1 mg; and a top-
pan balance, for preparation of solutions, accurate to ±0,1 g.
7 Sampling and sample preparation
7.1 Sampling technique
Carry out the sampling as specified in ISO 5667-1, ISO 5667-3, ISO 5667-5, ISO 5667-6, ISO 5667-8, and
ISO 5667-11, using sampling vessels as specified in 6.3.2.
For the determination of selenium in aqueous samples, acidify at time of sampling to acid (5.2) (3 ± 0,1) ml per litre is sufficient for most samples. Ensure that the pH is less than 2; otherwise,
add more hydrochloric acid as required.
For all types of samples, prepare an appropriate field blank and analyse as required. Use the same type
of vessel and quantity of acid as used in the sample.
7.2 Pre-reduction
7.2.1 General
Since only Se(IV) reacts quickly and quantitatively under the conditions used in the hydride technique,
Se(VI) has to be reduced to Se(IV) prior to the hydride generation step.
7.2.2 Standard procedure for water samples with no solid or organically bound selenium
Pre-treat water samples, free from solid materials and organically bound selenium, field blanks, and
blank solutions in the following way.
Accurately transfer an aliquot of the sample (40 ml to 50 ml) to a 100 ml volumetric flask.
Add (30 ± 0,5) ml of hydrochloric acid (5.2).
Reflux gently for 1 h.
Transfer to a volumetric flask and dilute to 100 ml with water (5.1).
If other sample volumes are applied, use reagents and equipment adequate for the chosen volumes.
For greater accuracy, the sample can also be dispensed by mass. In this case, calculate the volume from
the density and the mass and record the volume.

7.2.3 Samples requiring additional digestion
Samples that contain significant amounts of solid material and/or organically bound selenium require
an additional digestion step as specified in ISO 15587-1. This is outside the scope of this part of
ISO/TS 17379, but samples may be analysed using a similar procedure provided that correctly matrix-
matched reagents are prepared using the correct proportion of nitric acid (5.6) and hydrochloric acid
(5.2). Blanks and standard solutions shall also be matrix matched.
8 Instrumental set-up
Configure the instrumentation as described in the instrument manufacturer’s manual. The following
relates to continuous flow systems, for details on flow injection instruments or batch analysers refer to
manufacturer’s instruction manual.
Check tubing for wear and pumping reliability each day that the system is used and replace if necessary. All
tube distances between the auto-sampler, vapour generator, and detector shall be kept to a minimum length.
Fill the reagent reservoirs with reagent blank solution (5.7) and sodium tetrahydroborate solution (5.5),
respectively.
Set up the continuous flow vapour generator according to the manufacturer’s instructions. Ensure that
the reagent flows are within the accepted tolerances. Once stable conditions are established, analysis
can proceed. Both standards and samples shall be quantified using the same flow characteristics.
For samples which are above the calibration for a given range setting, re-analyse after dilution of the
sample into the calibration range. If the sample is diluted, then the dilutent shall be the reagent blank
(5.7), i.e. matrix matched.
Samples which are digested shall be matrix matched against standards and blanks to provide reliable data.
9 Procedure
9.1 General requirements
Before beginning the measurement procedure, set the instrument parameters according to the
manufacturer’s instructions (wavelength λ = 196,0 nm).
Align the silica tube atomizer or, if using the graphite furnace, atomization condition an iridium-coated
graphite tube according to the manufacturer’s instructions.
If a flow system is used, hydrochloric acid (5.3) shall be used as carrier solution. See recommendations
of the manufacturer.
The sodium tetrahydroborate solution (5.5) and all pre-reduced test solutions (reagent blank solution
(5.7), calibration solutions (5.8.6) and samples (7.2) are introduced to the flow system according to the
manufacturer’s instructions and measurements are carried out.
Measure the absorbance of each solution at least twice.
9.2 Analysis using the method of standard calibration
A standard calibration shall only be applied if matrix effects can be excluded. Otherwise samples can be
diluted, assuming that the selenium content is high enough. If not, use the standard addition method (9.3).
Plot a calibration graph for the analysis using the following procedure.
Prepare selenium calibration solutions as described in 5.8.6.

Determine the absorbance references of the selenium calibration solutions (5.8.6) and of the undigested,
but pre-reduced reagent blank solution (5.7).

Introduction
This part of ISO/TS 17379 is intended for use by analysts experienced with the handling of trace elements
at very low concentrations.
Inorganic selenium normally occurs in two oxidation states; Se(VI) and Se(IV). It is essential to convert all
selenium species to the Se(IV) state prior to generating the hydrides. Selenium(VI) does not form a hydride.
In natural water sources, selenium compounds generally occur in very small quantities, typically
less than 1 µg/l. Higher concentrations may be found, e.g. in industrial waste water. Selenium occurs
naturally in organic and inorganic compounds and may have oxidation states –II, 0, IV, and VI.
In order to fully decompose all of the selenium compounds, a digestion procedure is necessary. Digestion
can only be omitted if it is certain that the selenium in the sample can form a covalent hydride without
the necessity of a pre-oxidation digestion step.
The user should be aware that particular problems could require the specification of additional
marginal conditions.

1 Scope
This part of ISO/TS 17379 specifies a method for the determination of selenium. The method is
applicable to drinking water, surface water, ground water, and rain water. The dynamic range of this
part of ISO/TS 17379 is approximately 0,5 µg/l to 20 µg/l. Samples containing selenium at higher
concentrations than the application range can be analysed following appropriate dilution. The method
is unlikely to detect organoselenium compounds.
The sensitivity of this method is dependent on the selected operating conditions.
It is important to use high purity reagents in all cases with minimum levels of selenium.

ISO 15587-1, Water quality — Digestion for the determination of selected elements in water — Part 1: Aqua
regia digestion
3 Principle
An aliquot of sample is treated with concentrated hydrochloric acid (5.2). Se(VI) is pre-reduced to
Se(IV) by gently refluxing in 6 mol/l HCl for 1 h. Care is necessary to avoid any losses of volatile selenium
components. A suitable apparatus is shown in Figure B.3. The sample solutions are then treated with sodium
tetrahydroborate to generate the covalent gaseous hydride (SeH ). The hydride and excess hydrogen are

swept out of the generation vessel in the batch mode and out of the gas/liquid separator in the continuous
mode into a heated silica cell. After atomization the absorbance of selenium is determined at a wavelength,
λ = 196,0 nm. The procedure is automated by means of an auto sampler and control software.
Other measurement techniques may be applicable provided that the performance criteria can be
adequately demonstrated or exceeded by the user laboratory (see Annex C).
4 Interferences
The hydride generation technique is prone to interferences by transition and easily reducible metals.
For the majority of natural water samples, this type of interference should not be significant. The user
should carry out recovery tests on typical waters and also determine the maximum concentrations of
potentially interfering elements, using appropriate methods. If such interferences are indicated, the
level of interferences should be assessed by performing spike recoveries.
The reaction conditions set out in this part of ISO/TS 17379 have been chosen so that any interferences
are reduced to a minimum.

These elements do not cause interferences provided the concentrations specified in Table 1 are not exceeded.

If the concentrations in Table 1 are exceeded, it may be necessary to use the standard addition method
(9.3). Assuming that the selenium content is high enough, an appropriate dilution of the water sample
is preferred.
Metals which are readily reduced by sodium tetrahydroborate may also cause interferences. In particular,
these include chromium, iron, copper, nickel, and lead. If the concentrations of these elements specified
in Table 2 are exceeded, a significant decrease of absorption may occur.

5 Reagents and standards
During the analysis, unless otherwise stated, use only reagents of recognized analytical grade.
Reagents may contain selenium as an impurity. All reagents shall have selenium concentrations below
that which would result in a selenium blank value for the method being above the lowest level of interest.
Reagents shall be prepared to manufacturer’s recommendations using the following series as an example.
5.1 Water, complying with grade 1 as defined in ISO 3696, for all sample preparation and dilutions.
5.2 Hydrochloric acid, ρ(HCl) = 1,16 g/ml.
5.3 Hydrochloric acid, c(HCl) = 1 mol/l.
5.4 Sodium hydroxide, NaOH.
5.5 Sodium tetrahydroborate solution, ρ(NaBH ) = 13 g/l.

Dissolve an appropriate amount of sodium tetrahydroborate (13 g has proven suitable for the system
illustrated in Annex B) in 500 ml water (5.1) and add (4,0 ± 0,1) g of sodium hydroxide (5.4). Dilute
to 1 000 ml with water (5.1). Prepare on day of use and do not keep in a closed container because of
pressure build-up due to hydrogen evolution.
NOTE 1 The concentration of NaBH is dependent on the hydride generator manifold and flow rate conditions.

Prepare a nitric acid cleaning mixture by diluting nitric acid [ρ(HNO ) = 1,40 g/ml] with an equal volume

of water (5.1) by carefully adding the acid to the water.
5.7 Reagent blank.
For each 1 000 ml, prepare a solution containing (300 ± 3) ml of hydrochloric acid (5.2). Dilute to volume
with water (5.1).
On the continuous flow system, the reagent blank solution is run as background. Since the blank solution
may contain detectable trace levels of selenium, it is important that the same reagents be used for both
sample and standard preparation, as well as for preparation of the reagent blank. The analyte signal is
superimposed on this signal once the sample is introduced into the measurement cycle. The selenium
concentration of the blank solution should be less than the lower level of interest.

5.8 Selenium standard solutions.
5.8.1 Selenium stock solution A, ρ[Se(IV)] = 1 000 mg/l.
Use a quantitative stock solution with a selenium content of (1 000 ± 2) mg/l.
This solution is considered to be stable for at least one year.
NOTE Other stock solutions may be available and can be used providing the uncertainty of the measurement
is not compromised.
Alternatively, use a stock solution prepared from high purity grade chemicals:
Place 1,094 g of sodium selenite (Na SeO ) dried for 2 h at (105 ± 3) °C into a 500 ml volumetric flask.

Add 200 ml of water (5.1) and 200 ml of hydrochloric acid (5.3) and dissolve the sodium selenite
completely by stirring.
Dilute to 500 ml with water (5.1) and mix thoroughly.
5.8.2 Selenium standard solution B, ρ[Se(IV)] = 10 mg/l.
Pipette (1 ± 0,01) ml of selenium stock solution A (5.8.1) into a 100 ml volumetric flask, add (30 ± 0,5) ml
of hydrochloric acid (5.2) and fill up to the mark with water (5.1).
This solution is stable for one month.
5.8.3 Selenium standard solution C, ρ[Se(IV)] = 100 µg/l.
Pipette (1 ± 0,01) ml of selenium standard solution B (5.8.2) into a 100 ml volumetric flask, add (30 ±
0,5) ml of hydrochloric acid (5.2) and fill up to the mark with water (5.1).
This solution is stable for one week.
5.8.4 Selenium standard solution D, ρ[Se(IV)] = 10 µg/l.
Pipette (10 ± 0,1) ml of selenium standard solution C (5.8.3) into a 100 ml borosilicate volumetric flask.
Fill up to the mark with reagent blank solution (5.7).
This solution shall be prepared freshly on the day of use.
5.8.5 Selenium stock solution E, ρ[Se(VI)] = 1 000 mg/l.
Dissolve (2,392 ± 0,002) g of sodium selenate (Na SeO ) after drying at (105 ± 3) °C for 2 h in 500 ml

water (5.1), then transfer the solution quantitatively to a 1 000 ml volumetric flask and fill up to the
mark with water (5.1).
Use this standard to prepare a suitable selenium(VI) standard to check quantitative recovery of
selenium(IV).
The solution is stable for at least six months.
5.8.6 Selenium calibration solutions.
A minimum of five independent calibration solutions shall be used. Perform the calibration in accordance
with ISO 8466-1.
Prepare a minimum of five selenium calibration solutions from the selenium standard solution C (5.8.3)
covering the working range of expected selenium concentrations.
For the operating range from 1 µg/l to 5 µg/l, for example, proceed as follows.

Pipette into a series of five 100 ml volumetric flasks (1 ± 0,01) ml, (2 ± 0,02) ml, (3 ± 0,03) ml, (4 ±
0,04) ml, and (5 ± 0,05) ml, respectively, of selenium standard solution C (5.8.3).
Add 30 ml of hydrochloric acid (5.2).
Dilute to 100 ml with water (5.1) and mix thoroughly.
These calibration solutions contain 1 µg/l, 2 µg/l, 3 µg/l, 4 µg/l, and 5 µg/l selenium respectively.
They shall be prepared on the day of use.
The use of piston pipettes is permitted and enables the preparation of lower volumes of calibration
solutions. The application of dilutors is also allowed.
Once a calibration pattern has been established, the number of standards used routinely may be reduced.
Any such change shall not alter the result obtained from tests or the ranking with other samples.
6 Apparatus
Usual laboratory equipment and in particular the following.
The following are set out as guidelines. In general, the manufacturer’s instructions shall be followed.
6.1 Atomic absorption spectrometer.
6.1.1 General. An atomic absorption spectrometer equipped with a hydride generation system and
a heated silica tube atomizer or a graphite furnace atomizer (preferably equipped with a background
correction system) is recommended. Automated flow systems [flow injection analysis (FIA) or
continuous flow analysis (CFA)] for hydride generation are suitable and more usual but batch systems
are also adequate. The description below follows a continuous flow regime.
6.1.2 Radiation source for the determination of selenium. Use a radiation source which emits at
wavelength λ = 196,0 nm for selenium with an adequate spectral radiance.
6.2 Gas supply. Use argon with a grade specified by the manufacturer.
The gas supply shall be with a two stage regulator and the argon supplied at a pressure recommended
by the manufacturer.
The use of a gas purifier consisting of activated carbon is recommended.
Nitrogen gas may also be used but has reduced sensitivity.
6.3 Laboratory-ware.
6.3.1 General requirements. For the determination of selenium at very low concentrations,
contamination and loss are of critical consideration. Potential contamination sources include improperly
cleaned laboratory apparatus and general contamination within the laboratory environment. A clean
laboratory work area, designated for trace element sample handling shall be used.
All re-usable laboratory-ware in contact with the sample shall be cleaned prior to use.
Laboratory-ware shall be soaked in the nitric acid cleaning mixture (5.6) for at least 24 h and rinsed five
times with water (5.1).
Following this, refill laboratory-ware with hydrochloric acid, c(HCl) = 1 mol/l (5.3) and leave for 24 h.
Disposable (single-use) plastics laboratory-ware does not require special cleaning, provided that
negligible selenium contamination in that material is demonstrated.

6.3.2 Storage and sample processing bottles. Use sampling vessels constructed of silica, borosilicate
glass or suitable plastics materials] e.g. polytetrafluorethylene (PTFE), perfluoro (ethene-propene)
(FEP)] that neither absorb nor desorb the analyte under test.
6.3.3 Instrument reagent reservoir. The reagents are delivered via a peristaltic pump from glass
reagent bottles through PTFE transfer lines. All pump tubing shall be compatible with reagents in use
and neither adsorb nor desorb the analyte under test.
6.3.4 Auto-sampler vials. Use vials of polystyrene or materials specified in 6.3.2.
6.4 Sample processing equipment.
6.4.1 Air displacement pipette. Use a micropipette system capable of delivering volumes from 10 µl
to 1 000 µl with an assortment of metal-free, disposable pipette tips.
6.4.2 Balances. Analytical balance, capable of accurately weighing (standards) to ±0,1 mg; and a top-
pan balance, for preparation of solutions, accurate to ±0,1 g.
7 Sampling and sample preparation
7.1 Sampling technique
Carry out the sampling as specified in ISO 5667-1, ISO 5667-3, ISO 5667-5, ISO 5667-6, ISO 5667-8, and
ISO 5667-11, using sampling vessels as specified in 6.3.2.
For the determination of selenium in aqueous samples, acidify at time of sampling to acid (5.2) (3 ± 0,1) ml per litre is sufficient for most samples. Ensure that the pH is less than 2; otherwise,
add more hydrochloric acid as required.
For all types of samples, prepare an appropriate field blank and analyse as required. Use the same type
of vessel and quantity of acid as used in the sample.
7.2 Pre-reduction
7.2.1 General
Since only Se(IV) reacts quickly and quantitatively under the conditions used in the hydride technique,
Se(VI) has to be reduced to Se(IV) prior to the hydride generation step.
7.2.2 Standard procedure for water samples with no solid or organically bound selenium
Pre-treat water samples, free from solid materials and organically bound selenium, field blanks, and
blank solutions in the following way.
Accurately transfer an aliquot of the sample (40 ml to 50 ml) to a 100 ml volumetric flask.
Add (30 ± 0,5) ml of hydrochloric acid (5.2).
Reflux gently for 1 h.
Transfer to a volumetric flask and dilute to 100 ml with water (5.1).
If other sample volumes are applied, use reagents and equipment adequate for the chosen volumes.
For greater accuracy, the sample can also be dispensed by mass. In this case, calculate the volume from
the density and the mass and record the volume.

7.2.3 Samples requiring additional digestion
Samples that contain significant amounts of solid material and/or organically bound selenium require
an additional digestion step as specified in ISO 15587-1. This is outside the scope of this part of
ISO/TS 17379, but samples may be analysed using a similar procedure provided that correctly matrix-
matched reagents are prepared using the correct proportion of nitric acid (5.6) and hydrochloric acid
(5.2). Blanks and standard solutions shall also be matrix matched.
8 Instrumental set-up
Configure the instrumentation as described in the instrument manufacturer’s manual. The following
relates to continuous flow systems, for details on flow injection instruments or batch analysers refer to
manufacturer’s instruction manual.
Check tubing for wear and pumping reliability each day that the system is used and replace if necessary. All
tube distances between the auto-sampler, vapour generator, and detector shall be kept to a minimum length.
Fill the reagent reservoirs with reagent blank solution (5.7) and sodium tetrahydroborate solution (5.5),
respectively.
Set up the continuous flow vapour generator according to the manufacturer’s instructions. Ensure that
the reagent flows are within the accepted tolerances. Once stable conditions are established, analysis
can proceed. Both standards and samples shall be quantified using the same flow characteristics.
For samples which are above the calibration for a given range setting, re-analyse after dilution of the
sample into the calibration range. If the sample is diluted, then the dilutent shall be the reagent blank
(5.7), i.e. matrix matched.
Samples which are digested shall be matrix matched against standards and blanks to provide reliable data.
9 Procedure
9.1 General requirements
Before beginning the measurement procedure, set the instrument parameters according to the
manufacturer’s instructions (wavelength λ = 196,0 nm).
