Animal and vegetable fats and oils - Gas chromatography of fatty acid methyl esters - Part 3: Preparation of methyl esters using trimethylsulfonium hydroxide (TMSH) (ISO 12966-3:2009)

This part of ISO 12966 specifies a rapid base-catalysed transesterification method for fats and oils with trimethylsulfonium hydroxide (TMSH) to prepare fatty acid methyl esters. The method is exclusively applicable to the preparation of methyl esters of fats and oils for gas liquid chromatographic (GLC) analysis. It is applicable to all fats and oils including milk fat and blends containing milk fat. Isomerization of unsaturated fatty acids only occurs to a minor extent and isomerized fatty acids are only present at the determination limit. As isomerization takes place, the procedure is not recommended for conjugated linoleic acid (CLA). As CLA is not correctly analysed, this method is not applicable to the determination of the complete fatty acid composition of milk fat samples. Only about 70 % to 80 % of the free fatty acids are esterified. In the case of conjugated cyclopropyl and cyclopropenyl fatty acids, side reactions may occur, but these do not interfere with the determination of the fatty acids.

Diese Internationale Norm legt ein Schnellverfahren zur basekatalysierten Umesterung von Fetten und Ölen mit Trimethylsufoniumhydroxid (TMSH) für die Herstellung von Fettsäuremethylestern fest [1–5]. Das Verfahren ist ausschließlich zur Herstellung von Methylestern von Fetten oder Ölen für die GLC Analyse anwendbar. Es ist anwendbar bei allen Fetten und Ölen, einschließlich Milchfett und Mischungen, die Milchfette enthalten. Eine Isomerisierung von ungesättigten Fettsäuren tritt nur in geringem Umfang auf und die Menge der vor¬handenen isomerisierten Fettsäuren liegt bei der Bestimmungsgrenze. Da eine Isomerisierung stattfindet, wird das Verfahren nicht für konjugierte Linolsäure (en: conjugated linoleic acid, CLA) empfohlen. Freie Fettsäuren werden nur zu etwa 70 % bis 80 % verestert.
Im Fall von konjugierten, Cyclopropyl- und Cyclopropenyl-Fettsäuren treten möglicherweise Nebenreaktionen auf, die sich nicht störend auf die Bestimmung der Fettsäuren auswirken. Lipide, die Hydroxylgruppen ent-halten, werden jedoch partiell in korrespondierende O Methylderivate umgewandelt, die bei der GLC Tren-nung möglicherweise mit den Fettsäuremethylestern interferieren [6]. Im vorderen Teil des Chromatogramms (Bereich von C4) können Peaks auftreten, die durch das Reagens bedingt sind. Diese Peaks werden nicht berücksichtigt.
Diese Internationale Norm beruht auf der Deutschen Einheitsmethode C VI 11e (98) [5].

L'ISO 12966-3:2009 spécifie une méthode de transestérification rapide des corps gras par catalyse basique avec de l'hydroxyde de triméthylsulfonium, pour préparer des esters méthyliques d'acides gras. Cette méthode s'applique exclusivement à la préparation des esters méthyliques des corps gras pour l'analyse chromatographique en phase gazeuse-liquide. Elle s'applique à tous les corps gras y compris les matières grasses laitières et les mélanges contenant des matières grasses laitières. L'isomérisation des acides gras insaturés ne se produit que rarement et les acides gras isomérisés ne sont présents que dans la limite de détermination. Puisqu'il y a isomérisation, ce mode opératoire n'est pas recommandé pour l'acide linoléique conjugué (CLA). Puisque le CLA n'est pas correctement analysé, cette méthode ne s'applique pas à la détermination de la composition complète d'acide gras des échantillons de matières grasses laitières.
Seulement environ 70 % à 80 % des acides gras libres sont estérifiés. Dans le cas d'acides gras conjugués cyclopropyl et cyclopropényl, des réactions secondaires peuvent se produire mais n'interfèrent pas dans la détermination des acides gras.

Ta del ISO 12966 določa hitro bazično katalizirano transesterifikacijsko metodo za maščobe in olja s TMSH za pripravo metilestrov maščobnih kislin. Metoda velja izključno za pripravo metilestrov maščob in olj za analizo s plinsko tekočinsko kromatografijo (GLC). Velja za vse maščobe in olja, vključno z mlečno maščobo in mešanicami, ki vsebujejo mlečno maščobo. Do izomerizacije nenasičenih maščobnih kislin pride le v manjšem obsegu in izomerizirane maščobne kisline so prisotne v meji določanja. Zaradi izomerizacije postopek ni priporočljiv za konjugirano linolno kislino (CLA). products" in collaboration with Technical Committee CEN/TC 307 “Oilseeds, vegetable and animal fats and oils
and their by-products - Methods of sampling and analysis” the secretariat of which is held by AFNOR.
This European Standard shall be given the status of a national standard, either by publication of an identical
text or by endorsement, at the latest by June 2010, and conflicting national standards shall be withdrawn at
the latest by June 2010.

Foreword
ISO (the International Organization for Standardization) is a worldwide federation of national standards bodies
