Overview

EN ISO 10519:2015/A1:2025 is an amendment to the international standard for the determination of chlorophyll content in rapeseed using a spectrometric method. This update, published by the European Committee for Standardization (CEN), specifically adds a procedure for the preparation of the calibration curve to determine the k factor, as required in accurate chlorophyll analyses. The k factor is a crucial constant that depends on the path length and bandwidth of the spectrophotometer, and its correct determination ensures precision and comparability in measurement results.

Key Topics

  • Chlorophyll Content in Rapeseed: Establishes a standardized approach to assess chlorophyll levels, which is significant for oilseed quality and processing.
  • Spectrometric Analysis: Utilizes absorption measurements between 600 nm and 700 nm, emphasizing the impact of spectral bandwidth and instrument calibration.
  • Calibration Curve Preparation: Details the method for preparing a calibration curve using standard chlorophyll A solutions, enabling laboratories to determine the specific k factor for their spectrometers.
  • Reagents and Apparatus: Specifies the use of high-purity reagents-such as diethyl ether and chlorophyll A-and precise laboratory equipment including volumetric flasks, micropipettes, and spectrometers with known bandwidth.
  • Quality Assurance: Highlights the necessity to set and maintain constant bandwidth for accurate, repeatable results and underscores the relationship between instrument characteristics and analysis validity.

Applications

Implementing EN ISO 10519:2015/A1:2025 brings substantial benefits to a range of sectors involved in rapeseed handling and processing:

  • Quality Control in Oilseed Processing: Chlorophyll content can affect the taste, color, and shelf-life of rapeseed oil. Reliable measurement by standardized methods supports quality assurance and compliance with market and regulatory requirements.
  • Laboratory Standardization: Laboratories engaged in agricultural and food product testing can ensure their spectrometric measurements are internationally comparable by calibrating their instruments using the described procedure.
  • Research and Development: This standard empowers research institutions to generate robust data on rapeseed quality, supporting breeding programs, variety selection, and post-harvest technology improvements.
  • Trade and Compliance: Meeting the requirements of EN ISO 10519:2015/A1:2025 can facilitate national and international trade by demonstrating adherence to recognized methods for assessing oilseed quality.

Related Standards

For users of EN ISO 10519:2015/A1:2025, the following standards and guidelines may also be relevant to ensure integrated quality management and comprehensive oilseed analysis:

  • EN ISO 10519:2015 - The main standard for determination of chlorophyll content in rapeseed by spectrometric method.
  • ISO 5508 / ISO 5509 - Methods for analysis of oilseeds and vegetable oils, which may be used alongside chlorophyll determination for broader quality assessment.
  • CEN/TC 307 Standards - Covering oilseeds, vegetable oils, and derivatives, with a focus on sampling and analytical techniques.
  • Good Laboratory Practice (GLP) - For reliable implementation of analytical standards in industry and research settings.

By following EN ISO 10519:2015/A1:2025, stakeholders across the oilseed supply chain, laboratories, and regulatory bodies can uphold the highest standards of analytical accuracy, supporting product quality and market confidence.
