Microbiology of the food chain - Detection of Trichinella larvae in meat by artificial digestion method (ISO 18743:2015)
ISO 18743:2015 specifies a method of detection of Trichinella spp. muscle stage larvae in meat of individual animal carcasses intended for human consumption. It is applicable to the examination of meat from domestic and sylvatic animal species, which can be infected by nematodes of the genus Trichinella.
This method does not allow the identification of the species or genotype of detected parasites; species or genotype identification can be carried out by molecular methods.
The method described in this International Standard is intended to be used in conjunction with the guidelines in the OIE Manual of Diagnostic Tests and Vaccines and by the International Commission on Trichinellosis (ICT) for Trichinella testing and the inspection of carcasses intended for human consumption, unless it has been demonstrated by other means that the animal was not at risk for exposure to Trichinella.
The artificial digestion/magnetic stirrer method is considered to be the standard method because it has proven to give the most reliable results in validation studies.
NOTE Provided equivalence with the method described within this International Standard can be documented, alternative methods can be used for analysis.

In der vorliegenden Internationalen Norm wird ein Verfahren festgelegt, das zum Nachweis von Larven von Trichinella spp. in der Muskelphase (Muskellarven) im Fleisch von einzelnen Schlachttierkörpern, die für den menschlichen Verzehr bestimmt sind, angewendet werden kann. Es ist anwendbar zur Untersuchung von Fleisch von Haus- und Wildtierarten (aus dem domestischen und silvatischen Zyklus), die mit Nematoden der Gattung Trichinella infiziert sein können.
Dieses Verfahren ermöglicht keine Bestimmung der Art oder des Genotyps der nachgewiesenen Parasiten; eine Identifizierung kann mit molekularen Verfahren erfolgen.
Das in dieser Internationalen Norm beschriebene Verfahren ist in Verbindung mit den Leitlinien im OIE-Diagnosehandbuch und von der ICT zur Untersuchung auf Trichinella und zur Kontrolle von Schlachttierkörpern, die für den menschlichen Verzehr bestimmt sind, anzuwenden, sofern nicht auf andere Weise nachgewiesen wurde, dass das Tier kein Risiko bezüglich einer Trichinella-Exposition darstellte.
Das Magnetrührverfahren für die künstliche Verdauung wird als „Goldstandard“ (Maßstab) angesehen, weil in Validierungsstudien nachgewiesen wurde, dass es zu den zuverlässigsten Ergebnissen führt.
Vorausgesetzt, dass die Gleichwertigkeit mit dem in dieser Internationalen Norm beschriebenen Verfahren dokumentiert werden kann, dürfen alternative Verfahren zur Analyse angewendet werden.

ISO 18743:2015 décrit une méthode de détection de larves musculaires de Trichinella spp. dans la viande provenant de carcasses individuelles d'animaux destinées à la consommation humaine. Elle est applicable à l'examen de viande d'espèces animales domestiques et sauvages qui peuvent être infectées par des nématodes du genre Trichinella.
Cette méthode ne permet pas d'identifier l'espèce ou le génotype des parasites détectés; l'identification de l'espèce ou du génotype peut être réalisée à l'aide de méthodes moléculaires.
La méthode décrite dans la présente Norme internationale est destinée à être utilisée en association avec les lignes directrices du Manuel des tests de diagnostic et des vaccins de l'OIE et celles émises par la Commission internationale sur la trichinellose (ICT) pour la recherche de Trichinella et l'inspection de carcasses destinées à la consommation humaine, sauf s'il a été prouvé par d'autres moyens que l'animal ne risquait pas d'avoir été exposé à Trichinella.
La méthode de digestion artificielle utilisant un agitateur magnétique est considérée comme la méthode normalisée puisqu'il a été démontré, lors d'études de validation, qu'elle donne les résultats les plus fiables.

Einleitung
Trichinella spp. sind die Verursacher der Trichinellose beim Menschen, einer Erkrankung, die eine Gefahr für
die öffentliche Gesundheit darstellt und als Folge davon auch ein wirtschaftliches Problem in der
Schweineerzeugung. Aufgrund der zoonotischen Bedeutung dieser Infektion in vielen Ländern
konzentrieren sich die Bemühungen hauptsächlich auf Bekämpfung und/oder Tilgung von Trichinella bei
Hausschweinen, die die wichtigste Quelle für Infektionen beim Menschen weltweit sind.
Verdauungsverfahren zum Nachweis von Trichinella-Larven in Muskelproben von Schweinen und anderen
empfänglichen Tierarten, die für den menschlichen Verzehr bestimmt sind (z. B. Pferde, Wildschweine,
Walrosse und Bären), sind effektive Verfahren zur Vorbeugung einer klinischen Trichinellose beim
Menschen. Aufgrund der begrenzten Empfindlichkeit von Verdauungsverfahren können infizierte Tiere mit
nur sehr geringer Larvenanzahl in Muskelproben, die möglicherweise ein Risiko für subklinische Infektionen
beim Menschen darstellen können, durch diese Verfahren nicht erkannt werden.
WARNUNG — Anwender dieser Internationalen Norm sollten mit der üblichen Laborpraxis vertraut
sein. Diese Internationale Norm gibt nicht vor, alle unter Umständen mit der Anwendung des
