ISO 8968-1|IDF 20-1:2014 specifies a method for the determination of the nitrogen content and crude protein calculation of milk and milk products by the Kjeldahl principle, using traditional and block digestion methods. The methods are applicable to: liquid cow's (whole, partially skimmed or skimmed milk), goat's and sheep's whole milk; hard, semi-hard and processed cheese; dried milk and dried milk products (including milk-based infant formulae, milk protein concentrate, whey protein concentrate, casein and caseinate). The methods are not applicable to samples containing ammonium caseinate.

Les méthodes sont applicables: au lait de vache liquide (entier, demi-écrémé ou écrémé), au lait entier de chèvre et de brebis liquides; aux fromages à pâte dure, demi-dure ou molle; au lait en poudre et aux produits laitiers en poudre, notamment les laits infantiles, aux concentrés de protéines de lait, aux concentrés de protéines de lactosérum, aux caséines et aux caséinates. Elles ne sont pas applicables aux échantillons contenant du caséinate d'ammonium.

Принцип Кьельдаля и расчет содержания сырого белка
ПРЕДУПРЕЖДЕНИЕ — При применении настоящего международного стандарта могут
использоваться опасные материалы, операции и оборудование. В данном стандарте не
ставится цель описать все возможные проблемы, связанные с обеспечением безопасности при
его применении. Пользователь сам несет ответственность за меры безопасности и охрану
здоровья, а также определение применимости местных регулятивных ограничений перед его
использованием.
1 Область применения
Настоящий международный стандарт устанавливает метод определения содержания азота и расчета
содержания сырого белка в молоке и молочных продуктах по принципу Кьельдаля, используя
традиционный метод и метод минерализации в блоке.
Эти методы применимы к:
— питьевому коровьему (цельному, частично сепарированному или сепарированному молоку),
козьему и овечьему молоку;
― твердому, полутвердому и плавленому сыру;
― сухому молоку и сухим молочным продуктам (включая молочные смеси для детского питания,
концентраты молочного белка. концентраты сывороточного белка, казеины и казеинаты).
Данные методы не применимы к пробам, содержащим казеинат аммония.
ПРИМЕЧАНИЕ Могут быть получены неточные результаты при расчете содержания сырого белка в том
случае, если в продуктах, установленных в настоящем международном стандарте, присутствуют источники на
немолочной основе.

3 Термины и определения
Применительно к настоящему документу используются следующие термины и определения.
3.1
содержание азота
nitrogen content
массовая доля азота, определенная по установленной методике

массовая доля сырого белка, рассчитанная по установленной методике

4 Принцип
Пробу для анализа подвергают минерализации смесью концентрированной серной кислоты и сульфата
калия. Используя сульфат меди (II) в качестве катализатора превращают любой присутствующий
органический азот в сульфат аммония. Назначение сульфата калия состоит в повышении точки кипения
серной кислоты и создании более сильной окислительной смеси для минерализации. Добавляют
избыток гидроксида натрия к охлажденному минерализату для выделения аммиака. Выделяемый
аммиак отгоняют с водяным паром в избыток раствора борной кислоты и титруют титрованным
раствором соляной кислоты. Содержание азота рассчитывают по количеству полученного аммиака.
5 Реактивы
Если не указано иное, используют реактивы только установленной аналитической чистоты и
дистиллированную или деминерализованную воду или воду эквивалентной чистоты.
ПРИМЕЧАНИЕ Растворы, указанные в данной методике, могут отличаться от растворов, требуемых для
автоматических титраторов. Можно попытаться использовать эти растворы, но оператор сам несет
ответственность за соблюдение инструкций изготовителя оборудования.
5.1 Сульфат калия (K2SO4), не содержащий азота.
5.2 Сульфат меди (II), пентагидрат, раствор, c(CuSO45H O) = 5,0 г/100 мл.

Растворяют 5,0 г пентагидрата сульфата меди (II) в воде в мерной колбе с одной меткой вместимостью
100 мл. Разбавляют до метки водой и перемешивают.
5.3 Серная кислота (H2SO4), массовой долей от 95 % до 98 %, не содержащая азота
(приблизительно  = 1,84 г/мл).

5.4 Гидроксид натрия (NaOH), раствор, не содержащий азота, концентрацией 50 г гидроксида
натрия на 100 г.
В случае автоматических систем перегонки могут использоваться другие массовые доли раствора
гидроксида натрия при условии, что избыток гидроксида натрия распределяется в смеси для
перегонки; например, раствор гидроксида натрия с массовой долей 40 % может использоваться вместо
раствора гидроксида натрия с массовой долей 50 % там, где возникают проблемы, связанные с
закупоркой автоматической проточной системы. Следует учитывать общий объем такого раствора
гидроксида натрия, чтобы поддерживать соответствующие объемы перегонки.

5.5 Раствор индикатора
Растворяют 0,1 г метилового красного в 95 %-ном (объемная доля) растворе этанола в мерной колбе с
одной меткой вместимостью 50 мл (6.16). Разбавляют до объема 50 мл этанолом и перемешивают.
Растворяют 0,5 г бромкрезолового зеленого в 95 %-ном (объемная доля) растворе этанола в мерной
колбе с одной меткой вместимостью 250 мл (6.16). Разбавляют до объема 250 мл этанолом и
перемешивают. Смешивают одну часть раствора метилового красного с пятью частями раствора
бромкрезолового зеленого или объединяют и смешивают полностью оба раствора индикаторов.
5.6 Борная кислота, раствор, c(H BO )  40,0 г/л.

Растворяют 40,0 г борной кислоты (H3BO3) в 1 л горячей воды в мерной колбе с одной меткой
вместимостью 1 000 мл (6.16). Оставляют колбу с содержимым для охлаждения до температуры 20 °С.
Доводят до метки водой, добавляют 3 мл раствора индикатора (5.5) и перемешивают. Хранят раствор,
который должен иметь светло-оранжевую окраску, в склянке из боросиликатного стекла. При хранении
защищают раствор от света и источников паров аммиака.
В случае автоматических систем перегонки могут использоваться другие концентрации борной
кислоты после соответствующего обосновывания.
В случае использования электронного рН-метрического титрования с индикацией точки
эквивалентности можно опустить добавление раствора индикатора к раствору борной кислоты. С
другой стороны изменение окраски также можно использовать для проверки соответствующих
процедур титрования.
5.7 Соляная кислота, титрованный раствор, c(HCl)  (0,1 ± 0,000 5) моль/л.
Рекомендуется закупать реактив, титр которого был предварительно установлен изготовителем и
который соответствует или превышает требования настоящих технических условий. Часто
систематические ошибки (которые можно избежать), вносимые аналитиком при разбавлении
концентрированного исходного раствора кислоты, а затем при определении ее молярности, являются
причиной плохой воспроизводимости метода, рассматриваемого в этой части. Аналитик не должен
использовать раствор для титрования с концентрацией выше 0,1 моль/л, так как это будет уменьшать
общий титруемый объем на пробу и увеличивать неопределенность, выраженную в процентах, при
считывании значения по шкале бюретки. Все это будет негативно влиять на показатели повторяемости
и воспроизводимости данного метода.
Если вместо соляной кислоты используется серная кислота, то концентрация раствора серной кислоты
должна составлять 0,05 ± 0,000 3 моль/л.
5.8 Сульфат аммония [(NH ) SO ], с минимальным содержанием 99,9 % (массовая доля) на сухое

вещество.
Непосредственно перед использованием сушат сульфат аммония при температуре .102 °С ± 2 °С не
менее 2 ч. Охлаждают до комнатной температуры в эксикаторе.
5.9 Триптофан (C H N O ) или лизин гидрохлорид (C H ClN O ), с минимальным содержанием

99 % (массовая доля).
Не сушат эти реактивы в сушильном шкафу перед использованием.
5.10 Сахароза, с массовой долей азота не более 0,002 %.
Не сушат сахарозу в сушильном шкафу перед использованием.

6 Аппаратура
Обычная лабораторная аппаратура и, в частности, следующая.
6.1 Водяная баня, способная поддерживать температуру воды от 38 °С до 40 °С.
6.2 Аналитические весы, способные взвешивать с точностью до 0,1 мг.
6.3 Бюретка или автоматическая пипетка, способная подавать раствор сульфата меди (5.2)
порциями по 1,0 мл.
6.4 Градуированные мерные цилиндры, вместимостью 25 мл, 50 мл, 100 мл и 500 мл.
6.5 Конические колбы, вместимостью 500 мл.
6.6 Автоматическая бюретка, пригодной вместимости, например, 20 мл, с разрешающей
способностью по меньшей мере 0,004 мл, соответствующая требованиям ISO 8655-3. Альтернативно,
для анализа молока может использоваться бюретка вместимостью 50 мл с градуировкой через каждые
0,1 мл, соответствующая требованиям ISO 385, класс А.
ПРИМЕЧАНИЕ Для всех других продуктов ручная бюретка не дает разрешения, достаточного для достижения
требуемого количества значащих цифр.
6.7 Устройство для растирания
6.8 Колбы для минерализации (Кьельдаля), вместимостью 500 мл или 800 мл. Эти колбы
соответствуют используемой системе минерализации и требованиям изготовителя прибора для
минерализации (6.10 или 6.11).
6.9 Материал, способствующий равномерному кипению, например, твердые кусочки фарфора
или гранулы амфотерного алунда высокой чистоты, гладкие, с размером ячеек сита 10. Не используют
повторно материал, способствующий равномерному кипению.
ПРИМЕЧАНИЕ Кроме того, могут использоваться стеклянные шарики диаметром приблизительно 5 мм, но
они не могут поддерживать такое же эффективное кипение, как гранулы алунда, а также при минерализации с
использованием стеклянных шариков можно чаще столкнуться с проблемой пенообразования.
6.10 Аппарат для минерализации, удерживающий колбы Кьельдаля (6.8) в наклонном положении
(приблизительно 45°) и снабженный электронагревателями или газовыми горелками, которые не
нагревают колбы выше уровня их содержимого, и системой отвода дыма.
Источник тепла должен регулироваться для контроля максимального установочного значения
нагревателя, используемого при минерализации. Для оценки подвергают источник тепла
предварительному нагреву при установочном значении нагревателя. В случае газового нагревателя
период предварительного нагрева должен быть 10 мин, а для электронагревателя он должен
составлять 30 мин. Для каждого из нагревателей определяют установочное значение нагревателя,
которое доводит 250 мл воды, включая 5 – 10 кусочков материла, способствующего равномерному
кипению, от начальной температуры 25 °С до ее точки кипения за 5 – 6 мин. Это значение является
максимальным установочным значением нагревателя, используемым при минерализации.
6.11 Прибор для отгонки (традиционный метод), изготовленный из боросиликатного стекла или
другого подходящего материала, в котором может быть установлена колба для минерализации (6.8) с
эффективным брызгоотделителем, соединенным с холодильником с прямой внутренней трубкой и
отводящей трубкой, прикрепленной к его нижнему концу. Соединительные трубки и пробка(и) должны
быть плотно пригнаны и, предпочтительно, изготовлены из полихлоропрена.

ПРИМЕЧАНИЕ Прибор для отгонки, упомянутый выше, может быть заменен прибором конфигурации Parnas-

Wagner или другим подходящим оборудованием.
6.12 Блок для минерализации (метод минерализации в блоке), блок из алюминиевого сплава или
эквивалентный блок, снабженный терморегулятором и устройством для измерения температуры блока.
6.13 Пробирки для минерализации (метод минерализации в блоке), вместимостью 250 мл,
пригодные для использования в блоке для минерализации (6.12).
6.14 Вытяжной коллектор (метод минерализации в блоке), пригодный для использования с
пробирками для минерализации (6.13).
6.15 Центробежный скруббер или водоструйный насос или аспиратор (метод минерализации в
блоке), изготовленный из кислотоупорного материала, для использования с водопроводной сетью.
6.16 Мерные колбы, с одной меткой вместимостью 50 мл, 250 мл и 1 000 мл.
6.17 Установка для отгонки (метод минерализации в блоке), пригодная для перегонки с водяным
паром, ручная или полуавтоматическая, пригодная для установки пробирок для минерализации
вместимостью 250 мл (6.13) и конических колб вместимостью 500 мл (6.5).
6.18 Автоматический титратор, снабженный рН-метром.
рН-метр должен быть надлежащим образом откалиброван в диапазоне от рН 4 до рН 7 в соответствии
со стандартными лабораторными процедурами рН-калибровки. Бюретка автоматического титратора
должна соответствовать требованиям 6.6.
6.19 Шпатель или подходящее устройство.
6.20 Фильтровальная бумага, не содержащая азота, размеры и пористость которой подходит для
удерживания пробы для анализа сыра.
6.21 Освещенная плита магнитной мешалки.
7 Отбор проб
Отбор проб не является частью метода, рассматриваемого в данном международном стандарте.
Рекомендуемый метод отбора проб приводится в стандарте ISO 707IDF 50.
Важно, чтобы лаборатория получила пробу, которая является представительной и не была
повреждена или изменена при транспортировке или хранении.
8 Приготовление пробы для испытания
8.1 Цельное, частично сепарированное или сепарированное питьевое молоко
Помещают пробу для испытания на водяную баню (6.1), отрегулированную на температуру от 38 °С до
40 °С. Осторожно перемешивают путем переворачивания, не вызывая пенообразования или сбивания
сливок. Один раз основательно перемешивают пробу, охлаждают до комнатной температуры.
Продолжают, как указано в 9.1 или 9.2.

8.2 Твердый, полутвердый и плавленый сыр
Удаляют с поверхности сыра корковый, слизистый или заплесневелый слой таким образом, чтобы
получить пробу для испытания, представляющую сыр в обычном потребительском состоянии.
Протирают пробу для испытания с помощью устройства для растирания (6.7). Быстро перемешивают
всю массу и, предпочтительно, быстро протирают массу снова. Анализируют пробу для испытания как
можно быстрее после растирания.
С помощью шпателя (6.19) взвешивают требуемое количество (Таблица А.1) каждого подготовленного
протертого сыра на тарированный складчатый бумажный фильтр (6.20). Заворачивают сыр в

8.3 Сухое молоко и сухие молочные продукты
Оставляют пробу для испытания до достижения температуры от 20 °С до 25 °С перед переносом ее в
контейнер, внутренний объем которого приблизительно вдвое превышает объем пробы для испытания.
Сразу же закрывают контейнер, чтобы избежать изменения содержания влаги в пробе. Основательно
перемешивают пробу, многократно вращая и переворачивая контейнер.
Продолжают, как указано в 9.1 или 9.2.
9 Методики
9.1 Традиционный метод
9.1.1 Проба для анализа и предварительная обработка
Добавляют в чистую и сухую колбу для минерализации (6.8) от 5 до 10 кусочков материала,
способствующего равномерному кипению (6.9), 15,0 г сульфата калия (5.1), 1,0 мл раствора сульфата
меди (II) (5.2), количество подготовленной пробы для испытания (8.1, 8.2 или 8.3), как указано в

9.1.2 Определение
9.1.2.1 Минерализация
Включают систему отвода дыма аппарата для минерализации (6.10) до начала минерализации.
Нагревают колбу с ее содержимым (9.1.1) в аппарате для минерализации, используя достаточно низкое
установочное значение для нагревателя с тем, чтобы предотвратить бурное вспенивание обугленного
минерализата в горле колбы. Продолжают минерализацию при этом установочном значении до

максимальном установочном значении. Если кипение прозрачной жидкости не проявляется в виде
пузырьков, образующихся на поверхности горячей жидкости, то, возможно, температура нагревателя
слишком низкая. Общее время минерализации будет составлять от 1,8 ч до 3,25 ч.
Для определения точного времени кипения, требуемого для конкретных условий анализа молока в
данной лаборатории на данном комплекте аппаратуры, отбирают пробу молока с высоким
содержанием белка и высоким содержанием жира и определяют содержание белка в ней, используя
разное время кипения (от 1 ч до 2,5 ч) после осветления. Для других молочных продуктов требуются
пробы, состав которых аналогичен составу испытуемых проб. Средний результат содержания белка
увеличивается с возрастанием времени кипения, становится постоянным, а затем уменьшается, когда
время кипения становится слишком продолжительным. Выбирают такое время кипения, которое дает
максимальный результат содержания белка для испытуемого продукта.
В конце минерализации жидкость должна быть бесцветной и не содержать несгоревших частиц. Дают
минерализату остыть до комнатной температуры в открытой колбе в отдельном вытяжном шкафу в
течение приблизительно 25 мин. Если оставить колбу для охлаждения на горячем нагревателе, то
потребуется больше времени для достижения комнатной температуры. Охлажденный минерализат должен
представлять собой жидкость или жидкость с несколькими небольшими кристаллами на дне колбы в конце
25-минутного периода охлаждения. Нельзя оставлять неразбавленный минерализат в колбе на ночь.

1 Scope
This International Standard specifies a method for the determination of the nitrogen content and crude
protein calculation of milk and milk products by the Kjeldahl principle, using traditional and block
digestion methods.
The methods are applicable to:
— liquid cow’s (whole, partially skimmed or skimmed milk), goat’s and sheep’s whole milk;
— hard, semi-hard and processed cheese;
— dried milk and dried milk products (including milk-based infant formulae, milk protein concentrate,
whey protein concentrate, casein and caseinate).
The methods are not applicable to samples containing ammonium caseinate.
NOTE Inaccurate crude protein results will be obtained if non-milk sources of nitrogen are present in the
products specified in this International Standard.

4 Principle
A test portion is digested with a mixture of concentrated sulfuric acid and potassium sulfate. Using
copper sulfate (II) as a catalyst to thereby convert any organic nitrogen present to ammonium sulfate.
The function of the potassium sulfate is to elevate the boiling point of the sulfuric acid and to provide
a stronger oxidizing mixture for digestion. Excess sodium hydroxide is added to the cooled digest to
liberate ammonia. The liberated ammonia is steam distilled into the excess boric acid solution and
titration with hydrochloric acid standard volumetric solution is carried out. The nitrogen content is
calculated from the amount of ammonia produced.
5 Reagents
Use only reagents of recognized analytical grade, unless otherwise specified, and distilled or
demineralized water or water of equivalent purity.
NOTE The solutions specified in this procedure might be different than those required for the operation of
automated titrators. An effort is made to address those, but it is the responsibility of the operator to follow the
directions of the equipment manufacturer.
5.1 Potassium sulfate (K SO ), nitrogen free.

5.2 Copper (II) sulfate pentahydrate solution, c(CuSO .5H O) = 5,0 g/100 ml.

Dissolve 5,0 g of copper(II) sulfate pentahydrate in water in a 100 ml one-mark volumetric flask. Dilute
to the mark with water and mix.
5.3 Sulfuric acid (H SO ), with a mass fraction of between 95 % and 98 %, nitrogen-free (approximately

ρ = 1,84 g/ml).

5.4 Sodium hydroxide (NaOH) solution, nitrogen-free, containing 50 g of sodium hydroxide per
100 g.
With automated distillation systems, other mass fractions of sodium hydroxide may be used, provided
an excess of sodium hydroxide is dispensed to the distillation mixture; for example, a mass fraction of
40 % sodium hydroxide solution may be used instead of a mass fraction of 50 %, where plugging of the
automated flow system is a problem. The total volume of such sodium hydroxide solution should be
considered in order to maintain the suitable distillation volumes.
5.5 Indicator solution
Dissolve 0,1 g of methyl red in 95 % (volume fraction) ethanol in a 50 ml one-mark volumetric flask
(6.16). Dilute to 50 ml with ethanol and mix. Dissolve 0,5 g of bromocresol green in 95 % (volume
fraction) ethanol in a 250 ml one-mark volumetric flask (6.16). Dilute to 250 ml with ethanol and mix.
Mix one part of the methyl red solution with five parts of the bromocresol green solution or combine and
mix all of both solutions.
5.6 Boric acid solution, c(H BO ) = 40,0 g/l.

Dissolve 40,0 g of boric acid (H BO ) in 1 l of hot water in a 1 000 ml one-mark volumetric flask (6.16).

Allow the flask and its contents to cool to 20 °C. Adjust to the mark with water, add 3 ml of the indicator

solution (5.5) and mix. Store the solution, which will be light orange in colour, in a borosilicate glass
bottle. Protect the solution from light and sources of ammonia fume during storage.
With automated distillation systems, other boric acid concentrations may be used after validating
accordingly.
If using the electronic pH end point titration, the addition of the indicator solution to the boric acid
solution may be omitted. On the other hand, the change in colour may also be used as a check on proper
titration procedures.
5.7 Hydrochloric acid standard volumetric solution, c(HCI) = (0,1 ± 0,0005) mol/l.
It is recommended to purchase this material prestandardized by the manufacturer, which meets, or
exceeds, these specifications. Often, the systematic errors (which can be avoided) introduced by an
analyst diluting a concentrated stock acid and then determining the molarity of the acid, cause poor
reproducibility performance of the method in this part. The analyst should not use a solution for titration
that has a higher concentration than 0,1 mol/l, because this will reduce the total titration volume per
sample and the uncertainty in readability of the burette will become a larger percentage of the value.
This will have a negative impact on the method repeatability and reproducibility performance.
If sulfuric acid is substituted for hydrochloric acid, the solution should have a concentration of
0,05 ± 0,000 3 mol/l.
5.8 Ammonium sulfate [(NH ) SO ], minimum assay 99,9 % (mass fraction) on dried material.

Immediately before use, dry the ammonium sulfate at 102 °C ± 2 °C for not less than 2 h. Cool to room
temperature in a desiccator.
5.9 Tryptophan (C H N O ) or lysine hydrochloride (C H ClN O ), minimum assay 99 % (mass

fraction).
Do not dry these reagents in an oven before use.
5.10 Sucrose, with a mass fraction of nitrogen of not more than 0,002 %.
Do not dry the sucrose in an oven before use.
6 Apparatus
Usual laboratory apparatus and, in particular, the following.
6.1 Water bath, capable of maintaining a water temperature between 38 °C and 40 °C.
6.2 Analytical balance, capable of weighing to the nearest 0,1 mg.
6.3 Burette or automatic pipette, capable for delivering 1,0 ml portions of the copper sulfate solution
(5.2).
6.4 Graduated measuring cylinders, of capacity 25 ml, 50 ml, 100 ml and 500 ml.
6.5 Conical flasks, of capacity 500 ml.
6.6 Automatic burette, of suitable capacity e.g. 20 ml, with resolution of at least 0,004 ml, complying
with the requirements of ISO 8655-3. Alternatively, a burette, of capacity 50 ml, graduated at least at
every 0,1 ml, complying with the requirements of ISO 385, class A may be used for the analysis of milk.

NOTE The manual burette does not have sufficient resolution to achieve the required number of significant
figures for all other products.
6.7 Grinding device
6.8 Digestion flasks (Kjeldahl), of 500 ml or 800 ml capacities. Suitable to the digestion system to be
used and to the specifications of the manufacturer of the digestion apparatus (6.10 or 6.11).
6.9 Boiling aids, e.g. hard pieces of porcelain or high-purity amphoteric alundum (i.e. carbarundum)
granules, plain, mesh size 10. Do not reuse the boiling aids.
NOTE Glass beads of approximately 5 mm diameter can also be used, but they might not promote as efficient
boiling as the alundum granules and more foaming problems can be encountered during digestion with glass
beads.
6.10 Digestion apparatus, to hold the digestion flasks (6.8) in an inclined position (approximately
45 °), with electric heaters or gas burners, which do not heat the flasks above the level of their contents,
and with a fume extraction system.
The heater source should be adjustable to control the maximum heater setting to be used during
digestion. Preheat the heat source at the heater setting for evaluation. In the case of a gas heater, the
preheating period shall be 10 min and for an electric heater, it shall be 30 min. For each of the heaters,
determine the heater setting that brings 250 ml of water, including 5 to 10 boiling aids with an initial
temperature of 25 °C, to its boiling point in 5 min to 6 min. This is the maximum heater setting to be
used during digestion.
6.11 Distillation apparatus (traditional method), made of borosilicate glass or other suitable material
to which can be fitted a digestion flask (6.8) consisting of an efficient splash-head connected to an efficient
condenser with straight inner tube and an outlet tube attached to its lower end. The connecting tubing
and stopper(s) shall be close-fitting and preferably made of polychloroprene.

NOTE The distillation apparatus mentioned above can be replaced by the complete Parnas-Wagner
distillation configuration or other suitable equipment.
6.12 Digestion block (block digesting method), aluminium alloy block or equivalent block, fitted with
an adjustable temperature control and device for measuring block temperature.
6.13 Digestion tubes (block digesting method), of 250 ml in capacity, suitable for use with the
digestion block (6.12).
6.14 Exhaust manifold (block digesting method), suitable for use with the digestion tubes (6.13).
6.15 Centrifugal scrubber apparatus or filter pump or aspirator (block digesting method),
constructed of acid-resistant material, for use with mains water supply.
6.16 Volumetric flasks, one mark of 50 ml, 250 ml and 1 000 ml capacities.
6.17 Distillation unit (block digesting method), capable of steam distilling, manual or semi–automatic,
suited to accept the 250 ml digestion tubes (6.13) and the 500 ml conical flasks (6.5).

6.18 Automatic titrator provided with a pH-meter.
The pH-meter should be calibrated properly in the range of pH 4 to pH 7 following normal laboratory
pH-calibration procedures. The automatic titrator burette shall comply with the requirements of 6.6.
6.19 Spatula or suitable transfer device.
6.20 Filter paper, nitrogen–free, of dimensions and porosity suitable to hold the cheese test portion.
6.21 Illuminated magnetic stirrer plate.
7 Sampling
Sampling is not part of the method specified in this International Standard. A recommended sampling
method is given in ISO 707|IDF 50.
It is important that the laboratory receive a sample which is representative and has not been damaged
or changed during transport or storage.
8 Preparation of test sample
8.1 Whole, partially skimmed or skimmed liquid milk
Place the test sample in the water bath (6.1) set at 38 °C to 40 °C. Mix gently by inversion without causing
frothing or churning. Once the sample is mixed thoroughly, cool to room temperature.
Proceed as indicated in 9.1 or 9.2.
8.2 Hard, semi-hard and processed cheese
Remove the rind, smear or mouldy surface layer of the cheese, in such a way as to provide a test sample
representative of the cheese as it is usually consumed.
Grind the test sample by means of an appropriate device (6.7). Quickly mix the whole mass and, preferably,
grind the mass again quickly. Analyse the test sample as soon as possible after grinding.
Using the spatula (6.19), weigh the required amount (Table A.1) of either prepared ground cheese onto
a pre-folded, tared filter paper (6.20). Enclose cheese in filter paper and drop the filter paper containing
the cheese into the bottom of a digestion flask (6.8) or digestion tube (6.13) as indicated in 9.1.1 or 9.2.1.
NOTE Use of a filter paper can promote foam formation in block digestion systems. To avoid this, when using
the block digestion method (9.2), the filter paper can be omitted by weighing the sample into a suitable vessel,
weighing the cheese and vessel, transferring the cheese to the digestion vessel, reweighing the empty vessel and
determining the sample mass by subtracting the mass of the empty vessel from the mass of the cheese and vessel.
8.3 Dried milk and dried milk products
Let the test sample reach a temperature of between 20 °C and 25 °C before transferring to a container of
internal volume approximately twice the volume of the test sample. Close the container immediately to
avoid changing the moisture content of the sample. Thoroughly mix the sample by repeatedly rotating
and inverting the container.
Proceed as indicated in 9.1 or 9.2.

9.1 Traditional method
9.1.1 Test portion and pre-treatment
Add to a clean and dry digestion flask (6.8), 5 to 10 boiling aids (6.9), 15,0 g of potassium sulfate (5.1),
1,0 ml of copper (II) sulfate solution (5.2), the amount of prepared test sample (8.1, 8.2 or 8.3) as indicated
in Table A.1, weighed to the nearest 0,1 mg, and 25 ml of sulfuric acid (5.3) while using sulfuric acid to
wash down any copper (II) sulfate solution, potassium sulfate or test portion left on the neck of the flask.
Gently mix the contents of the digestion flask.
As an alternative to 5.1 and 5.2, commercially available tablets containing, for example 15 g of potassium
sulfate and 0,05 g of copper (II) sulfate pentahydrate, may be used, provided
a) the tablets contain a quantity of potassium sulfate such that the required amount can be dispensed
using an integer number of whole tablets to maintain a similar salt to acid (5.3) ratio. For instance,
three tablets each containing 5 g of potassium sulfate, may be used with 20 ml of sulfuric acid (5.3),
and
b) the tablets do not contain salts of toxic metals, such as selenium or mercury.
9.1.2 Determination
9.1.2.1 Digestion
Turn on the fume extraction system of the digestion apparatus (6.10) prior to beginning the digestion.
Heat the digestion flask and its contents (9.1.1) on the digestion apparatus using a heater setting low
enough such that charred digest does not foam up the neck of the digestion flask. Digest at this heat
setting until white fumes appear in the flask after approximately 20 min. Increase the heater setting
to half way to the maximum setting determined in 6.10 and continue the heating period for 15 min.
At the end of the 15-min period increase the heat to maximum setting determined in 6.10. After the
digest clears (clear with light blue-green colour), continue boiling for 1 h to 2,5 h at maximum setting. If
visible boiling of the clear liquid is not apparent as bubbles forming at the surface of the hot liquid, the
temperature of the heating device might be too low. The total digestion time will be between 1,8 h and
3,25 h.
To determine the specific boiling time required for analysis conditions in a particular laboratory using
a particular set of apparatus, select for milk analysis a high-protein, high-fat milk sample and determine
its protein content using different boiling times (1 h to 2,5 h) after clearing. Other milk products require
samples of compositions similar to those being tested. The mean protein result increases with increasing
boil time, becomes constant and then decreases when boil time is too long. Select the boil time that
yields the maximum protein result for the product tested.
At the end of digestion, the digest shall be clear and free of undigested material. Allow the digest to cool
to room temperature in an open flask in a separate hood over a period of approximately 25 min. If the
flask is left on the hot heating device to cool, it will take longer to reach room temperature. The cooled
digest should be liquid or liquid with a few small crystals at the bottom of the flask at the end of the 25-
min cooling period. Do not leave the undiluted digest in the flasks overnight. The undiluted digest might
crystallize during this period and it will be very difficult to get the crystallized digest back into solution.
NOTE Excessive crystallization after 25 min is the result of undue acid loss during digestion and can result
in low test values. Undue acid loss is caused by excessive fume aspiration or by an excessively long digestion time
caused by an incorrect maximum burner setting.
Add 300 ml of water to the 500 ml digestion flasks or 400 ml of water when using the 800 ml digestion
flasks. Wash down the neck of the flask during the addition of water. Mix the contents thoroughly
ensuring that any crystals that separate out are dissolved. Allow the mixture to cool again to room

ATTENTION — La présente Norme internationale peut impliquer l’utilisation de produits
et la mise en œuvre de modes opératoires et d’appareillages à caractère dangereux. La
présente Norme internationale ne prétend pas aborder tous les risques liés à son utilisation.

pratiques d’hygiène et de sécurité appropriées et de déterminer l’applicabilité des restrictions

1 Domaine d’application
La présente Norme internationale spécifie une méthode de détermination de la teneur en azote et de
calcul de la teneur en protéines brutes du lait et des produits laitiers selon la méthode Kjeldahl, à l’aide
des méthodes de minéralisation classiques et en bloc.
Les méthodes sont applicables:
— au lait de vache liquide (entier, demi-écrémé ou écrémé), au lait entier de chèvre et de brebis liquides;
— aux fromages à pâte dure, demi-dure ou molle;
— au lait en poudre et aux produits laitiers en poudre, notamment les laits infantiles , aux concentrés
de protéines de lait, aux concentrés de protéines de lactosérum, aux caséines et aux caséinates.
Elles ne sont pas applicables aux échantillons contenant du caséinate d’ammonium.
NOTE Des résultats erronés seront obtenus pour les protéines brutes si des sources d’azote autres que le lait
sont présentes dans les produits spécifiés dans la présente Norme internationale.

4 Principe
Minéralisation d’une prise d’essai avec un mélange d’acide sulfurique concentré et de sulfate de
potassium, en utilisant du sulfate de cuivre (II) comme catalyseur pour convertir ainsi l’azote organique
présent en sulfate d’ammonium. La fonction du sulfate de potassium est d’élever le point d’ébullition de
l’acide sulfurique et de permettre d’obtenir un mélange oxydant plus fort pour la minéralisation. Addition
d’hydroxyde de sodium excédentaire au minéralisat refroidi pour libérer de l’ammoniac. Distillation à
la vapeur de l’ammoniac libéré dans un excédent de solution d’acide borique, puis titration en utilisant
une solution volumétrique standard d’acide chlorhydrique. Calcul de la teneur en azote à partir de la
quantité d’ammoniac produite.
5 Réactifs
Sauf indication contraire, utiliser uniquement des réactifs de qualité analytique reconnue ainsi que de
l’eau distillée ou déminéralisée, ou de l’eau de pureté équivalente.
NOTE Les solutions spécifiées dans le présent mode opératoire peuvent être différentes de celles requises
pour le fonctionnement des dispositifs de titration automatiques. L’objectif est de les présenter, mais il incombe à
l’opérateur de respecter les instructions du fabricant de l’équipement.
5.1 Sulfate de potassium (K SO ), exempt d’azote.

5.2 Solution de sulfate de cuivre (II) pentahydraté, c(CuSO .5H O) = 5,0 g/100 ml.

Dans une fiole jaugée de 100 ml, dissoudre 5,0 g de sulfate de cuivre (II) pentahydraté dans de l’eau.
Compléter au volume avec de l’eau, puis mélanger.
5.3 Acide sulfurique (H SO ), ayant une concentration (M/M) comprise entre 95 % et 98 %, sans

azote (ρ = environ 1,84 g/ml).

5.4 Solution d’hydroxyde de sodium (NaOH), exempte d’azote, contenant 50 g d’hydroxyde de
sodium pour 100 g.
Avec les systèmes de distillation automatiques, d’autres concentrations (M/M) d’hydroxyde de sodium
peuvent être utilisées à condition qu’un excédent d’hydroxyde de sodium soit ajouté au mélange de
distillation, par exemple une concentration (M/M) de 40 % de solution d’hydroxyde de sodium peut être
utilisée à la place d’une concentration (M/M) de 50 %, dans le cas où l’on rencontre des problèmes de
colmatage. Il convient de tenir compte du volume total de cette solution d’hydroxyde de sodium pour
maintenir les volumes de distillation à un niveau approprié.
5.5 Solution d’indicateur
Dans une fiole jaugée de 50 ml à un repère (6.16), dissoudre 0,1 g de rouge de méthyle dans de l’éthanol
à 95 % (rapport de volume). Diluer à 50 ml avec de l’éthanol et mélanger. Dans une fiole jaugée de 250 ml
à un repère (6.16), dissoudre 0,5 g de vert de bromocrésol dans de l’éthanol à 95 % (rapport de volume).
Diluer à 250 ml avec de l’éthanol et mélanger. Mélanger un volume de la solution de rouge de méthyle
avec cinq volumes de la solution de vert de bromocrésol ou combiner et mélanger l’ensemble des deux
solutions.

5.6 Solution d’acide borique, c(H BO ) = 40,0 g/l.

Dans une fiole jaugée de 1 000 ml (6.16), dissoudre 40,0 g d’acide borique (H BO ) dans 1 l d’eau chaude.

Laisser refroidir la fiole et son contenu à 20 °C. Compléter au volume avec de l’eau, ajouter 3 ml de
la solution d’indicateur (5.5) et mélanger. Conserver la solution, qui doit être orange clair, dans une
bouteille en verre de borosilicate. Durant le stockage, protéger la solution de la lumière et des sources
de vapeur d’ammoniac.
Avec les systèmes de distillation automatiques, d’autres concentrations en acide borique peuvent être
utilisées si elles ont été validées.
En cas de titration avec point final en pH, l’ajout de la solution d’indicateur à la solution d’acide borique
n’est pas obligatoire. D’autre part, le changement de couleur peut aussi servir à contrôler le mode
opératoire de titration.
5.7 Solution volumétrique standard d’acide chlorhydrique, c(HCl) = (0,1 ± 0,0005) mol/l.
Il est recommandé d’acheter ce matériau déjà ajusté à cette concentration et répondant à ces spécifications.
Bien souvent, les erreurs systématiques (qui peuvent être évitées) introduites par un analyste qui dilue
un acide concentré, puis détermine la molarité de l’acide, peuvent diminuer la reproductibilité de la
méthode. Il convient que l’analyste n’utilise pas de solution de titration de concentration supérieure
à 0,1 mol/l car cela réduirait le volume total de titration par échantillon, et l’incertitude de lecture de
la burette représenterait un pourcentage plus élevé de la valeur. Cela aura un impact négatif sur les
performances de répétabilité et de reproductibilité de la méthode.
Si de l’acide sulfurique est utilisé à la place de l’acide chlorhydrique, il convient que la solution présente
une concentration de 0,05 ± 0,0003 mol/l.
5.8 Sulfate d’ammonium [(NH ) SO ], ayant une pureté minimale de 99,9 % (concentration (M/M))

sur la matière sèche.
Immédiatement avant l’emploi, sécher le sulfate d’ammonium à 102 °C ± 2 °C pendant au moins 2 h.
Laisser refroidir à température ambiante dans un dessiccateur.
5.9 Tryptophane (C H N O ) ou chlorhydrate de lysine (C H ClN O ), ayant une pureté

minimale de 99 % (concentration (M/M)).
Ne pas sécher ces réactifs dans une étuve avant l’emploi.
5.10 Saccharose, dont la concentration (M/M) en azote est inférieure à 0,002 %.
Ne pas sécher le saccharose avant l’emploi.
6 Appareillage
Matériel courant de laboratoire et, en particulier, ce qui suit.
6.1 Bain d’eau, pouvant être maintenu à une température comprise entre 38 °C et 40 °C.
6.2 Balance analytique, précise à 0,1 mg près.
6.3 Burette ou pipette automatique, permettant d’obtenir des doses de 1,0 ml de solution de sulfate
de cuivre (5.2).
6.4 Éprouvettes graduées, d’une capacité de 25 ml, 50 ml, 100 ml et 500 ml.
6.5 Fioles coniques, d’une capacité de 500 ml.

6.6 Burette automatique, de capacité appropriée (par exemple 20 ml), d’une résolution d’au moins
0,004 ml, conforme aux exigences de l’ISO 8655-3. Il est également possible d’utiliser une burette d’une
capacité de 50 ml, graduée au moins tous les 0,1 ml, conforme aux exigences de l’ISO 385, classe A, pour
l’analyse du lait.
NOTE Les burettes manuelles n’ont pas une résolution suffisante pour atteindre le nombre requis de chiffres
significatifs pour tous les autres produits.
6.7 Broyeur
6.8 Ballons de minéralisation (Kjeldahl), d’une capacité de 500 ml ou 800 ml. Adaptés au système de
minéralisation à utiliser et conformes aux spécifications du fabricant de l’appareil de minéralisation (6.10
ou 6.11).
6.9 Corps facilitant l’ébullition, par exemple pièces de porcelaine dures ou granules d’alundon
amphotères (carbarundon) lisses, d’une pureté élevée et d’une taille de mailles de 10. Ne pas réutiliser
les corps facilitant l’ébullition.
NOTE Des billes de verre d’environ 5 mm de diamètre peuvent également être utilisées, mais celles-ci peuvent
être moins efficaces pour l’ébullition que les granules d’alundon, et des problèmes de formation de mousse pendant
la minéralisation peuvent davantage se poser avec les billes de verre.
6.10 Appareil de minéralisation, pour maintenir les ballons de minéralisation (6.8) en position inclinée
(environ 45°), pourvu de résistances électriques ou de becs à gaz ne chauffant pas les ballons au-delà du
niveau de leur contenu, ainsi que d’un système d’évacuation des fumées.
Il convient que la source chauffante soit réglable pour permettre de contrôler le réglage maximal de
l’élément chauffant à appliquer durant la minéralisation. Préchauffer la source chauffante au réglage
de l’élément chauffant à évaluer. La durée de préchauffage doit être de 10 min dans le cas d’un bec à
gaz et de 30 min dans le cas d’un élément chauffant électrique. Déterminer, pour chacun des éléments
chauffants, le réglage qui permet de porter à ébullition 250 ml d’eau et 5 à 10 corps facilitant l’ébullition
(en partant d’une température initiale de 25 °C) en 5 min à 6 min. Ce réglage correspond au réglage
maximal de l’élément chauffant à appliquer durant la minéralisation.
6.11 Appareil de distillation (méthode classique), en verre de borosilicate ou autre matière appropriée,
pouvant être équipé d’un ballon de minéralisation (6.8), se composant d’une tête antiprojections
efficace, relié à un condenseur efficace avec tube intérieur droit et un tube d’écoulement fixé à son
extrémité inférieure. Le tube de connexion et le(s) bouchon(s) doivent être étanches et de préférence en
polychloroprène.
NOTE L’appareil de distillation mentionné ci-dessus peut être remplacé par le montage de distillation de

Parnas-Wagner complet ou par un autre équipement approprié.
6.12 Bloc de minéralisation (méthode de minéralisation en bloc), bloc en alliage d’aluminium ou
bloc équivalent, équipé d’un régulateur de température et d’un dispositif de mesure de la température du
bloc.
6.13 Tubes de minéralisation (méthode de minéralisation en bloc), d’une capacité de 250 ml,
compatibles avec le bloc de minéralisation (6.12).
6.14 Tubulure d’échappement (méthode de minéralisation en bloc), compatible avec les tubes de
minéralisation (6.13).

6.15 Séparateur centrifuge ou pompe ou aspirateur à filtre (méthode de minéralisation en bloc),
constitué d’un matériau résistant à l’acide, utilisable avec l’alimentation en eau principale.
6.16 Fioles jaugées, à un repère, d’une capacité de 50 ml, 250 ml et 1 000 ml.
6.17 Unité de distillation (méthode de minéralisation en bloc), permettant une distillation à la
vapeur, manuelle ou semi-automatique, compatible avec les tubes de minéralisation de 250 ml (6.13) et
les fioles coniques de 500 ml (6.5).
6.18 Dispositif de titration automatique pourvu d’un pH-mètre
Il convient que le pH-mètre soit correctement étalonné dans la gamme de pH 4 à pH 7 selon les méthodes
normales d’étalonnage du pH en laboratoire. La burette du dispositif de titration automatique doit être
conforme aux exigences énoncées en 6.6.
6.19 Spatule ou dispositif de transfert approprié
6.20 Papier filtre, exempt d’azote, de dimensions et de porosité appropriées pour contenir la prise
d’essai de fromage.
6.21 Agitateur magnétique éclairé.
7 Échantillonnage
L’échantillonnage ne fait pas partie de la méthode spécifiée dans la présente Norme internationale. Une
méthode d’échantillonnage recommandée est indiquée dans l’ISO 707|FIL 50.
Il est important que le laboratoire reçoive un échantillon représentatif et n’ayant pas été endommagé ou
modifié durant le transport ou le stockage.
8 Préparation de l’échantillon pour essai
8.1 Lait liquide entier, demi-écrémé ou écrémé
Placer l’échantillon pour essai dans le bain d’eau (6.1) réglé entre 38 °C et 40 °C. Bien mélanger en
retournant sans formation de mousse ni barattage. Une fois que l’échantillon est bien mélangé, le laisser
refroidir à température ambiante.
Procéder comme indiqué en 9.1 ou 9.2.
8.2 Fromage à pâte dure, demi-dure ou molle
Enlever la croûte, la graisse ou la couche de surface moisie du fromage, de façon à fournir un échantillon
pour essai représentatif du fromage couramment consommé.
Broyer l’échantillon pour essai à l’aide d’un dispositif approprié (6.7). Mélanger rapidement toute la
masse et broyer à nouveau la masse rapidement, de préférence. Après avoir broyé l’échantillon pour
essai, l’analyser dès que possible.
Avec la spatule (6.19), peser la quantité requise (Tableau A.1) de fromage broyé préparé sur un papier
filtre taré et préalablement plié (6.20). Enrober le fromage dans le papier filtre et déposer le papier filtre

contenant le fromage au fond d’un ballon de minéralisation (6.8) ou d’un tube de digestion (6.13) comme
indiqué en 9.1.1 ou 9.2.1.
NOTE L’utilisation d’un papier filtre peut favoriser la formation de mousse dans les systèmes de minéralisation
en bloc. Pour éviter ce phénomène, le papier filtre peut, lors de l’utilisation de la méthode de minéralisation en
bloc (9.2), ne pas être utilisé en pesant l’échantillon dans un récipient approprié, en pesant ensuite le fromage et le
récipient en transférant le fromage dans le récipient de minéralisation, et en pesant à nouveau le récipient à vide.
On déterminera ainsi la masse de l’échantillon en soustrayant la masse du récipient à vide de la masse du fromage
et du récipient.
8.3 Lait en poudre et produits laitiers en poudre
Laisser l’échantillon pour essai atteindre une température comprise entre 20 °C et 25 °C avant de le
transvaser dans un récipient d’un volume interne environ égal au double du volume de l’échantillon
pour essai. Fermer immédiatement le récipient pour éviter de modifier la teneur en eau de l’échantillon.
Mélanger vigoureusement l’échantillon en faisant tourner et en retournant plusieurs fois le récipient.
Procéder comme indiqué en 9.1 ou 9.2.
9 Mode opératoire
9.1 Méthode classique
9.1.1 Prise d’essai et prétraitement
Dans un ballon de minéralisation (6.8) propre et sec, introduire 5 à 10 corps facilitant l’ébullition (6.9),
15,0 g de sulfate de potassium (5.1), 1,0 ml de solution de sulfate de cuivre (II) (5.2), la quantité de
l’échantillon pour essai préparé (8.1, 8.2 or 8.3) comme indiqué dans le Tableau A.1, pesé à 0,1 mg près,
et 25 ml d’acide sulfurique (5.3). À cet effet, utiliser l’acide sulfurique pour entraîner tout résidu de la
solution de sulfate de cuivre (II), du sulfate de potassium ou de la prise d’essai restant sur le col du
ballon. Mélanger doucement le contenu du ballon de minéralisation.
Des comprimés disponibles dans le commerce contenant, par exemple, 15 g de sulfate de potassium et
0,05 g de sulfate de cuivre (II) pentahydraté, peuvent être utilisés comme alternative aux dispositions
des paragraphes 5.1 et 5.2, à condition que :
a) les comprimés contiennent une quantité de sulfate de potassium telle que la quantité requise peut
être distribuée en utilisant un nombre entier de comprimés entiers permettant de maintenir un
rapport sel/acide (5.3) à un niveau similaire.
