ISO 8262-3|IDF 124-3:2005 specifies the reference method for the determination of the fat content of milk-based and of liquid, concentrated or dried milk products to which the Röse-Gottllieb method is not applicable; i.e. those containing distinct quantities of free fatty acids or those which are not completely soluble in ammonia owing to the presence of lumps or non-milk ingredients, such as custards, porridges or certain milk-based products for bakery purposes. The method is also applicable to fresh cheese types, such as cottage cheese and quarg, as well as to fresh cheeses with added fruit, syrup, muesli, etc. for which the Schmid-Bondzynski-Ratzlaff method is not suitable owing to the higher carbohydrate contents and/or extreme inhomogeneity.

La méthode est également applicable aux fromages frais tels que fromage blanc et pâte fraîche, ainsi qu'aux fromages frais contenant des fruits, du sirop, du «muesli», etc. pour lesquels la méthode Schmid-Bondzynski-Ratzlaff ne convient pas, en raison des teneurs plus élevées en hydrates de carbone et/ou d'une hétérogénéité extrême.

Введение
Настоящий международный стандарт был подготовлен в рамках разработки серии контрольных
методов, гармонизированных в наиболее возможной степени, для гравиметрического определения
содержания жира в молоке, молочных продуктах и пищевых продуктах на основе молока. Эти методы
основаны на принципах Розе-Готлиба (RG), Вейбулла-Бернтропа (WB) или Шмидта-Бондзински-
Ратзлафа (SBR) .
Для данной части ISO 8262IDF 124, рассматривающей нежирные продукты на основе молока и
нежирные жидкие, концентрированные или сухие молочные продукты и/или продукты, содержащие
нерастворимые немолочные компоненты, был выбран метод на основе принципа WB по следующим
причинам:
a) метод RG не применим в случае присутствия заметного количества свободных жирных кислот, а
также в том случае, когда продукт содержит комки и/или немолочные компоненты, не растворимые
в аммиаке, которые служат причиной неполной экстракции жира;
b) метод SBR не применим из-за значительного содержания лактозы, которая при кислотном
гидролизе дает рост соединений, экстрагируемых эфиром, и таким образом приводит к
завышенным значениям содержания жира;
c) метод WB, хотя также включает кислотный гидролиз, не оказывает неблагоприятного влияния на
образование соединений, экстрагируемых эфиром, так как кислый гидролизат фильтруют и
промывают, а сухой остаток на фильтре не содержит соединений, экстрагируемых петролейным
эфиром;
d) описанный метод уже используется для этой цели во многих странах.
Первоначально метод Вейбулла был разработан для хлеба; значительно модифицированный метод,
установленный в настоящем международном стандарте, разработан Бернтропом. Данная
модификация метода нашла широкое применение для определения содержания жира во многих типах
пищевых продуктов.

1 Область применения
Настоящая часть ISO 8262IDF 124 устанавливает контрольный метод определения содержания жира
в продуктах на основе молока и в жидких, концентрированных или сухих молочных продуктах, к
которым не применим метод Розе-Готлиба; т.е. в продуктах, содержащих заметные количества
свободных жирных кислот, или продуктах, которые не полностью растворимы в аммиаке из-за наличия
комков или немолочных компонентов, таких как сладкий крем, овсяная каша или некоторые продукты
на основе молока для хлебопекарной промышленности.
ПРИМЕЧАНИЕ 1 Контрольные методы Розе-Готлиба для определения содержания жира в молоке, сливках,
сгущенном молоке без сахара и с сахаром и сухих молочных продуктах установлены в ISO 1211, ISO 2450, ISO
1737 и ISO 1736 соответственно.
Данный метод также применим к свежим сырам, таким как домашний и кисло-молочный сыры, а также
к свежим сырам с добавками фруктов, сиропа, “мюслей” и т.д., для которых метод SBR не применим
из-за высокого содержания углеводов и/или чрезмерной неоднородности.
ПРИМЕЧАНИЕ 2 Контрольный метод Шмидта-Бондзински-Ратзлафа для определения содержания жира в сыре и
плавленых сырах с содержанием лактозы менее 5 % (массовая доля) в обезжиренном сухом веществе установлен
в ISO 1735.
2 Термины и определения
Применительно к настоящему документу применяются следующие термины и определения.

3 Принцип
Подвергают пробу для анализа гидролизу при кипении с разбавленной соляной кислотой. Фильтруют
горячий гидролизат через влажный бумажный фильтр для удержания жирных веществ, затем
экстрагируют жир с высушенного бумажного фильтра н-гексаном или петролейным эфиром. Удаляют
растворитель при перегонке или выпаривании и взвешивают экстрагированные вещества. (Этот метод
обычно известен как метод Вейбулла-Бернтропа.)
4 Реактивы и материалы
Используют только реактивы признанной аналитической чистоты, которые не оставляют заметного
остатка при определении установленным методом. Используют дистиллированную или

деионизированную воду или воду, по меньшей мере, эквивалентной чистоты.
4.1 Разбавленная соляная кислота, содержащая приблизительно 20 % (массовая доля) HCl, ρ
20
приблизительно 1,10 г/мл.
Разбавляют 100 мл концентрированной соляной кислоты (ρ = 1,18 г/мл) 100 мл воды и перемешивают.

4.2 Экстракционный растворитель, не содержащий воды: н-гексан или петролейный эфир с
пределами кипения от 30 °C до 60 °C.
Для проверки качества экстракционного растворителя перегоняют 100 мл растворителя из
экстракционной колбы (5.4), подготовленной в соответствии с 7.4. Используют пустую экстракционную
колбу, подготовленную тем же способом, для проверки массы (см. 10.1). Растворитель не должен
оставлять остатка массой более 1,0 мг.
Заменяют или перегоняют растворитель, если он не соответствует этому требованию.
4.3 Фильтровальная бумага, диаметром 150 мм, гофрированная, средней плотности,
предпочтительно обезжиренная.
Для проверки качества фильтровальной бумаги выполняют контрольный опыт, как указано в 7.3,
используя растворитель, удовлетворяющий требованию 4.2. Используют пустую экстракционную колбу
(5.4), подготовленную в соответствии с 7.4, для проверки массы (см. 10.1). Бумага не должна оставлять
остатка массой более 2,5 мг.
Заменяют фильтровальную бумагу, не удовлетворяющую этому требованию.
4.4 Синяя лакмусовая бумага.
4.5 Диатомовая земля (необязательно; см. 7.5.3).
4.6 Чистая лактоза (необязательно; см. 7.5.3).
4.7 Хлопковая вата, обезжиренная при экстрагировании растворителем (4.2) в течение 1,5 ч и высушенная.
5 Аппаратура
ПРЕДУПРЕЖДЕНИЕ — Поскольку при определении используются летучие воспламеняющиеся
растворители, необходимо, чтобы применяемая электроаппаратура соответствовала
требованиям законодательства в отношении опасностей, возникающих при использовании
таких растворителей.
Используют обычную лабораторную аппаратуру и, в частности, следующую.
5.1 Аналитические весы.
5.2 Смеситель, для гомогенизации лабораторной пробы, при необходимости. Например,
используют измельчитель пищевых продуктов или высокоскоростной смеситель с соответствующим
сосудом вместимостью 1 л, снабженным крышкой.
5.3 Экстракционный аппарат, непрерывного или полунепрерывного действия. Например, используют
аппарат типа Сокслета, состоящий из экстракционной колбы (плоскодонной, короткогорлой) вместимостью
150 мл, экстрактора с объемом сифонирования от 40 мл до 60 мл и эффективного обратного холодильника,
снабженного осушительной трубкой или пробкой из хлопковой ваты.
5.4 Экстракционные колбы, вместимостью 150 мл, плоскодонные и короткогорлые.

5.5 Экстракционные гильзы, изготовленные из обезжиренной фильтровальной бумаги, стекла,
оксида алюминия или политетрафторэтилена (PTFE), не вызывающих образования заметного остатка
при контрольном опыте, или изготовленные из целлюлозы, одной толщины, внутренним диаметром 22
мм и наружной длиной 80 мм, для использования с экстракционным аппаратом (5.3).
5.6 Водяные бани, способные поддерживать следующие температуры:
 40 °C - 60 °C (см. 7.1.1);
 30 °C - 40 °C (см. 7.1.2).
5.7 Нагреватель, для экстракционного аппарата. Например, используют водяную или песчаную
баню или нагревательную плитку, регулируемую с помощью термореле.
5.8 Кипелки, обезжиренные, например, стеклянные шарики или кусочки нехрупкого, непористого
фарфора или карбида кремния.
5.9 Коническая колба, вместимостью 250 мл, снабженная обратным холодильником,
предпочтительно типа Либиха.
5.10 Нагреватель, для конической колбы, снабженной холодильником. Например, используют
лабораторную сетку и газовую горелку, электрическую плитку или песчаную баню.
5.11 Фильтровальная воронка, пригодная для использования с гофрированной фильтровальной бумагой (4.3).
5.12 Химические стаканы с носиком, вместимостью 100 мл и 250 мл.
5.13 Аппарат для перегонки, в котором можно осторожно перегонять растворитель из колб при
температуре, не превышающей 100 °C.
5.14 Сушильный шкаф, с электрическим обогревом, полностью открытым(и) вентиляционным(и)
отверстием(ями), способный поддерживать во всем рабочем пространстве температуру 102 °C ± 2 °C.

ISO (the International Organization for Standardization) is a worldwide federation of national standards
bodies (ISO member bodies). The work of preparing International Standards is normally carried out through
ISO technical committees. Each member body interested in a subject for which a technical committee has

This edition of ISO 8262-3⎪IDF 124-3 cancels and replaces IDF 126A:1988, of which it constitutes a minor
revision.
ISO 8262⎪IDF 124 consists of the following parts, under the general title Milk products and milk-based
foods — Determination of fat content by the Weibull-Berntrop gravimetric method (Reference method):

Introduction
ISO 8262⎪IDF 124 has been prepared within the framework of producing a series of reference methods,
which are harmonized to the greatest possible extent, for the gravimetric determination of the fat content of
milk, milk products and milk-based foods. These methods are based on the Röse-Gottlied (RG) method, or
the Weibull-Berntrop (WB) method, or the Schmid-Bondzynski-Ratzlaff (SBR) principle.
For this part of ISO 8262⎪IDF 124, dealing with milk-based and with liquid, concentrated or dried milk products
in poor condition and/or containing insoluble non-milk ingredients, a method based on the WB principle has
been chosen for the following reasons:
a) the RG procedure is not suitable when a distinct quantity of free fatty acids is present, or when the
product contains lumps and/or non-milk ingredients insoluble in ammonia, since the extraction of fat is
incomplete;
b) the SBR procedure is not suitable owing to a considerable lactose content, which gives rise to some
ether-extractable compounds in the digestion with acid and thus gives too high values for the fat content;
c) the WB procedure, although it also includes acid digestion, is not adversely affected by the ether-
extractable compounds, since the acid digest is filtered and washed, and the dried residue on the filter
does not contain compounds that are extractable by light petroleum;
d) the method described is already used for this purpose in many countries.
The original Weibull method was designed for bread; a considerably modified method, as specified in this
International Standard, was developed by Berntrop. This version has found wide application for the
determination of fat in many types of food product.

1 Scope
This part of ISO 8262⏐IDF 124 specifies the reference method for the determination of the fat content of milk-
based and of liquid, concentrated or dried milk products to which the Röse-Gottllieb method is not applicable;
i.e. those containing distinct quantities of free fatty acids or those which are not completely soluble in
ammonia owing to the presence of lumps or non-milk ingredients, such as custards, porridges or certain milk-
based products for bakery purposes.
NOTE 1 Reference Röse-Gottlieb methods for the determination of the fat content of milk, of cream, of evaporated and
sweetened condensed milk, and of dried milk products are specified in ISO 1211, ISO 2450, ISO 1737 and ISO 1736
respectively.
The method is also applicable to fresh cheese types, such as cottage cheese and quarg, as well as to fresh
cheeses with added fruit, syrup, “muesli”, etc. for which the SBR method is not suitable owing to the higher
carbohydrate contents and/or extreme inhomogeneity.
NOTE 2 A reference Schmid-Bondzynski-Ratzlaff method for the determination of the fat content of cheese and
processed cheese products having lactose contents below 5 % (mass fraction) of the non-fat solids is specified in
ISO 1735.
2 Terms and definitions
For the purposes of this document, the following terms and definitions apply.

all the substances determined by the method specified in this part of ISO 8262⏐IDF 124
NOTE It is expressed as a mass fraction in percent.
3 Principle
A test portion is digested by boiling with dilute hydrochloric acid. The hot digest is filtered through a wetted
filter paper to retain fatty substances, then the fat is extracted from the dried filter paper using n-hexane or
light petroleum. The solvent is removed by distillation or evaporation and the substances are extracted and
weighed. (This is usually known as the Weibull-Berntrop principle.)

4 Reagents and materials
Use only reagents of recognized analytical grade that leave no appreciable residue when the determination is
carried out by the method specified. Use distilled or deionized water, or water of at least equivalent purity.
4.1 Dilute hydrochloric acid, containing approximately 20 % (mass fraction) of HCl, ρ approximately
20
1,10 g/ml.
Dilute 100 ml of concentrated hydrochloric acid (ρ = 1,18 g/ml) with 100 ml of water and mix.

4.2 Extraction solvent, free from water: n-hexane or light petroleum having any boiling range between
30 °C and 60 °C.
To test the quality of the extraction solvent, distil 100 ml of it from an extraction flask (5.4) prepared as
specified in 7.4. Use an empty extraction flask, prepared in the same way, to check the mass (see 10.1). The
solvent shall leave no residue greater than 1,0 mg.
Replace or distil the solvent if it does not meet this requirement.
4.3 Filter papers, of diameter 150 mm, pleated, medium grade, preferably defatted.
To test the quality of the filter paper, carry out a blank test as specified in 7.3, using a solvent satisfying the
requirement of 4.2. Use an empty extraction flask (5.4), prepared as specified in 7.4, to check the mass (see
10.1). The paper shall leave no residue greater than 2,5 mg.
Replace unsatisfactory filter papers.
4.4 Blue litmus paper.
4.5 Diatomaceous earth (optional; see 7.5.3).
4.6 Pure lactose (optional; see 7.5.3).
4.7 Cotton wool, defatted by extraction with the solvent (4.2) for 1,5 h and dried.
5 Apparatus
WARNING — Since the determination involves the use of volatile flammable solvents, electrical
apparatus employed may be required to comply with legislation relating to the hazards in using such
solvents.
Usual laboratory equipment and, in particular, the following.
5.1 Analytical balance.
5.2 Blender, for homogenizing the laboratory sample, if necessary. For example, use a food chopper or a
high-speed blender with a blender jar, of capacity 1 litre, fitted with a lid.
5.3 Extraction apparatus, continuous or semi-continuous. For example, use a Soxhlet type, consisting of
an extraction flask (flat-bottomed, short-necked) of capacity 150 ml, an extractor with a siphoning volume of
40 ml to 60 ml, and an efficient reflux condenser fitted with a drying tube or plug of cotton wool.
5.4 Extraction flasks, of capacity 150 ml, flat-bottomed and short-necked.
5.5 Extraction thimbles, made of defatted filter paper, glass, alumina or polytetrafluoroethylene (PTFE),
contributing no appreciable residue in the blank test, or made of cellulose, single thickness, of internal
diameter 22 mm and external length 80 mm, for use with the extraction apparatus (5.3).

5.6 Water baths, capable of being maintained at the following temperatures:
⎯ 40 °C to 60 °C (see 7.1.1);
⎯ 30 °C to 40 °C (see 7.1.2).
5.7 Heating apparatus, for the extraction apparatus. For example, use a water bath, sand bath or a
thermostatically controlled hotplate.
5.8 Boiling aids, fat-free, such as glass beads or pieces of non-friable, non-porous porcelain or silicon
carbide.
5.9 Conical flask, of capacity 250 ml, fitted with a reflux condenser, preferably of the Liebig type.

Introduction
L'ISO 8262-3⎪FIL 124-3 a été établie en vue de disposer d'une série de méthodes de référence, harmonisées
le mieux possible, pour la détermination par gravimétrie de la teneur en matière grasse du lait, des produits
laitiers et des aliments à base de lait. Ces méthodes reposent soit sur le principe de Röse-Gottlieb (RG), soit
sur celui de Weibull-Berntrop (WB), soit encore sur celui de Schmid-Bondzynski-Ratzlaff (SBR).
Pour la présente partie de l'ISO 8262-3⎪FIL 124-3, concernant le lait et les produits laitiers liquides qui ont été
concentrés ou séchés dans de mauvaises conditions et/ou contenant des composés insolubles non laitiers,
une méthode basée sur le principe de WB a été retenue pour les raisons suivantes:
a) lorsqu'une quantité élevée d'acides gras libres est présente ou lorsque le produit contient des grumeaux
et/ou des non-laitiers insolubles dans l'ammoniaque, l'extraction de la matière grasse est incomplète, de
sorte que la méthode RG ne convient pas;
b) une teneur généralement élevée en hydrates de carbone augmentant la quantité de composés
extractibles par l'éther lors de la minéralisation acide et donnant alors des valeurs trop élevées de la
teneur en matière grasse rend la méthode SBR inappropriée;
c) la méthode WB, bien qu'impliquant aussi une minéralisation acide, n'est pas troublée par ces composés
extractibles par l'éther puisque le minéralisat est filtré et lavé, et que le résidu séché sur le filtre ne
contient aucun composé extractible par l'éther de pétrole;
d) la méthode décrite est déjà utilisée à cette intention dans de nombreux pays.
La méthode originale Weibull était appliquée au pain. Une méthode grandement modifiée, telle que spécifiée
dans la présente Norme internationale, a été développée par Berntrop. Cette version a été largement
appliquée dans plusieurs pays pour la détermination de la teneur en matière grasse dans de nombreux
produits alimentaires.

1 Domaine d'application
La présente partie de l'ISO 8262⎪FIL 124 spécifie la méthode de référence pour la détermination de la teneur
en matière grasse du lait et des produits laitiers liquides concentrés ou séchés, pour lesquels la méthode
Röse-Gottlieb n'est pas applicable, car ces produits contiennent des quantités significatives d'acides gras
libres ou ne sont pas complètement solubles dans l'ammoniaque à cause de la présence de grumeaux, de
composés non laitiers, par exemple flans, bouillies d'avoine ou certains autres produits à base de lait destinés
à la boulangerie.
NOTE 1 Les méthodes de référence Röse-Gottlieb utilisées pour la détermination de la teneur en matière grasse du
lait, de la crème, du lait concentré sucré, ou de produits laitiers séchés, sont décrites respectivement dans l'ISO 1211,
l'ISO 2450, l'ISO 1737 et l'ISO 1736.
La méthode est également applicable aux fromages frais tels que fromage blanc et pâte fraîche, ainsi qu'aux
fromages frais contenant des fruits, du sirop, du «muesli», etc. pour lesquels la méthode SBR ne convient pas,
en raison des teneurs plus élevées en hydrates de carbone et/ou d'une hétérogénéité extrême.
NOTE 2 Une méthode de référence Schmid-Bondzynski-Ratzlaff, pour la détermination de la teneur en matière grasse
dans les fromages et fromages fondus ayant des teneurs en lactose inférieures à 5 % (fraction massique) des matières
solides non grasses, est spécifiée dans l'ISO 1735.
2 Termes et définitions
Pour les besoins du présent document, les termes et définitions suivants s'appliquent.
2.1

NOTE Elle est exprimée en fraction massique en pourcentage.
3 Principe
Minéralisation d'une prise d'essai en la portant à ébullition avec de l'acide chlorhydrique dilué, filtration du
minéralisat chaud sur papier-filtre humide pour retenir les substances grasses, extraction de la matière grasse
retenue sur le papier-filtre séché à l'aide de n-hexane ou d'éther de pétrole, élimination du solvant par
distillation ou évaporation, puis pesée des substances extraites. (Méthode habituellement connue sous le nom
de Weibull-Berntrop.)

4 Réactifs et matériaux
Sauf indication différente, utiliser uniquement des réactifs de qualité analytique reconnue et ne laissant pas de
résidu appréciable lorsque la détermination est effectuée selon la méthode spécifiée. L'eau utilisée doit être
de l'eau distillée ou de l'eau déminéralisée ou l'eau de pureté au moins équivalente.
4.1 Acide chlorhydrique, solution à environ 20 % (fraction massique) d'HCI, ρ = 1,10 g/ml environ.

Diluer 100 ml d'acide chlorhydrique concentré (ρ = 1,18 g/ml) avec 100 ml d'eau et mélanger.

4.2 Solvant d'extraction, exempt d'eau: n-hexane ou éther de pétrole ayant un point d'ébullition compris
entre 30 °C et 60 °C.
Pour vérifier la qualité du solvant d'extraction, distiller 100 ml à partir d'une fiole d'extraction (5.4) préparée
comme spécifié en 7.4. Utiliser une fiole vide d'extraction préparée de la même façon, pour les contrôles de
masse (voir 10.1). Le solvant ne doit pas laisser de résidus supérieurs à 1,0 mg.
Remplacer ou distiller le solvant s'il ne remplit pas cette condition.
4.3 Papiers-filtres, de 150 mm de diamètre, plissés, à filtration moyenne, de préférence dégraissés.
Pour vérifier la qualité du papier-filtre, effectuer un essai à blanc comme spécifié en 7.3, à l'aide d'un solvant
satisfaisant aux exigences de 4.2. Utiliser une fiole d'extraction vide (5.4), préparée comme spécifié en 7.4
pour les contrôles de masse (voir 10.1). Le papier ne doit pas laisser de résidus supérieurs à 2,5 mg.
Remplacer le papier-filtre non résistant.
4.4 Papier indicateur.
4.5 Terre de diatomées (facultatif; voir 7.5.3).
4.6 Lactose pur (facultatif; voir 7.5.3).
4.7 Tampon de coton, dégraissé par extraction à l'aide du solvant (4.2) pendant 1,5 h et séché.
5 Appareillage
AVERTISSEMENT — Pour les déterminations requérant l'utilisation de solvants volatils inflammables,
l'appareillage électrique utilisé devra satisfaire, le cas échéant, à la législation en matière de risques
liés à l'utilisation de ces solvants.
Matériel courant de laboratoire et, en particulier, ce qui suit.
5.1 Balance analytique.
5.2 Broyeur, pour le broyage de l'échantillon pour laboratoire, si nécessaire; par exemple hachoir
alimentaire ou broyeur à haute vitesse avec un bol d'une capacité de 1 litre, muni d'un couvercle.
5.3 Appareil d'extraction, continu ou semi-continu, par exemple de type Soxhlet, constitué d'une fiole
d'extraction (à fond plat et à col étroit) d'une capacité de 150 ml, d'un extracteur permettant de siphonner un
volume de 40 ml à 60 ml et d'un réfrigérant à reflux efficace, muni d'un tube à dessiccation ou d'un tampon de
coton.
5.4 Fioles d'extraction, de 150 ml de capacité, à fond plat et à col étroit.
5.5 Cartouches d'extraction, faites en papier-filtre dégraissé, en verre, en oxyde d'aluminium ou en
polytétrafluoroéthylène (PTFE), n'apportant aucun résidu lors de l'essai à blanc, ou faites en cellulose,

monocouche, de 22 mm de diamètre intérieur et de 80 mm de longueur, pour utilisation avec l'appareil
d'extraction (5.3).
5.6 Bains d'eau, réglables aux températures suivantes:
⎯ 40 °C à 60 °C (voir 7.1.1);
⎯ 30 °C à 40 °C (voir 7.1.2).
5.7 Appareil de chauffage, pour l'appareil d'extraction, par exemple bain d'eau, bain de sable ou plaque
chauffante thermostatée.
5.8 Régulateurs d'ébullition, exempts de matière grasse, tels que billes de verre, morceaux de porcelaine
non friable et non poreuse ou carbure de silicium.
5.9 Fiole conique, de 250 ml de capacité, munie d'un réfrigérant à reflux, de préférence de type Liebig.
