1 Scope
This International Standard specifies a method for the determination of the alkaline phosphatase activity in
pasteurized whole milk, semi-skimmed milk, skimmed milk, cream and flavoured milks using a
chemiluminescent (EPAS) method.
The method is applicable to milk and milk-based drinks from cows, sheep, buffalo and goats.
The method is also suitable for any liquid-based sample if diluted in such a way that the diluted alkaline
phosphatase activity has less than 7 000 milliunits per litre.
NOTE There has been a successful collaborative trial with cow, sheep, buffalo, and goat whole milk, as well as
skimmed cow milk (< 0,5 % fat), 20 % fat cream and 2 % fat chocolate milk (all mass fractions).
2 Terms and definitions
For the purposes of this document, the following terms and definitions apply.
2.1
alkaline phosphatase activity
ALP
activity of the alkaline phosphatase present in the product, determined according to the procedure described
in this International Standard
[4], [5]
NOTE The alkaline phosphatase activity is expressed as milliunits of enzyme activity per litre (mU/l) .
2.2
unit of alkaline phosphatase activity
amount of alkaline phosphatase enzyme that catalyses the transformation of 1 µmol of stable aromatic
substrate per minute
3 Principle
The alkaline phosphatase activity is measured by photo-activation of the hydrolysed product followed by an
instrumental measurement of photo-activation. In the presence of alkaline phosphatase, a stable aromatic
dioxetane-phosphate substrate is hydrolysed at 35 °C ± 1 °C to produce a photo-activated (chemiluminescent)
product. The photo-activation of the product is amplified by a macromolecular enhancing component. The
hydrolysis reaction is stopped after a specified incubation time (3 min). The amount of chemiluminescent
product thus produced is measured and converted to enzyme units by a luminometer. Luminometer calibration
is based on calibration using tablets with known enzyme activity.

4 Reagents
Use only reagents of recognized analytical grade, unless otherwise specified, and distilled or demineralized
water or water of equivalent purity.
4.1 Non-chemiluminescent dioxetane ester substrate [0,2 mol/l 3-(2'-spiroadamantanane)-4-methoxy-4-
(3''-phosphate phenyl-1,2 dioxetane disodium salt in DEAE buffer with 1 % fluorosine], is commercially ®

available [e.g. as Charm reagent AP liquid].
It is recommended to store the substrate at between 0 °C and 7 °C. If stored in amber plastic vials and at 4 °C,
the substrate remains stable for 6 months. If stored at 30 °C, the substrate is stable for 24 h only.
When using the substrate in the assay, keep it at between 0 °C and 7 °C or on ice.
4.2 Stopping solution
Prepare the stopping solution by mixing the same amount of 0,15 mol/l 2-amino-2-methyl-1-propanol and
0,02 % benzalkonium chloride, pH 10,7. The stopping solution shall be at room temperature (18 °C to 24 °C)
prior to use. For assay consistency when not using a thermoprobe, record and maintain the temperature to
within 0,5 °C of the solution temperature used at calibration.
NOTE The stopping solution is used to stop enzymatic hydrolysis of the dioxetane ester substrate (4.1).
Commercially supplied stopping solution has a shelf life of 1 year when kept at 4 °C, or of 2 months when kept
at room temperature. For daily use, room temperature storage is recommended.
4.3 Working calibrators, for example, calibration tablets (dried raw milk with measured phosphatase
content in tablet form to rehydrate in milk) with a phosphatase activity of 875 µU/l ± 26 µU/l. Rehydrate the
tablets in three different volumes of milk-based drink presenting no phosphatase activity or negative test
sample (7.2) to create a standard calibration curve.
Commercially supplied calibration tablets may be stored at 4 °C for 2 years.
4.3.1 Fluid white milk calibrators
In each of three 50 ml test tubes (5.9), marked A , B and C , respectively, dissolve one calibrator tablet in

100 µl of distilled water.
Add 20 ml to tube A , 5 ml to tube B and 2,5 ml to tube C of fluid white milk products (presenting no

phosphatase activity) or negative test sample (7.2). This makes calibration standards A , B and C with a

phosphatase activity of A = 44 mU/l, B = 175 mU/l and C = 350 mU/l, respectively. Cap the tubes and

shake their contents vigorously. Allow the tube contents to rehydrate under refrigeration for 10 min. Mix well
before use.
4.3.2 Cream and flavoured-milk calibrators
In each of three 50 ml test tubes (5.9), marked A , B and C , respectively, dissolve one calibrator tablet in

100 µl of distilled water.
Add 10 ml to tube A , 5 ml to tube B and 2,5 ml to tube C of cream or flavoured-milk products (presenting no

phosphatase activity) or negative test sample (7.2), making calibration standards A , B , C with phosphatase

activities of A = 88 mU/l, B = 175 mU/l and C = 350 mU/l, respectively. Cap the tubes and shake their

contents vigorously. Allow the tube contents to rehydrate under refrigeration for 10 min. Mix well before use.

4.4 Positive control
As positive control, use freeze-dried alkaline phosphatase in a 15 ml amber bottle. Rehydrate the positive
control with 10 ml of a representative dairy drink presenting no phosphatase activity, or with a prepared
negative test sample (7.2). Rehydrated positive control contains 450 mU/l of phosphatase enzyme.
Allow the control to stand for 10 min to rehydrate. Vigorously shake before use.
The rehydrated positive control is stable when stored at between 0 °C and 7 °C for 48 h. Positive control
rehydrated with fluid milk is stable frozen at or below −15 °C for 2 months. Thaw the positive control in water
at room temperature. Vigorously shake the control to homogenize before use. Do not re-freeze.
5 Apparatus
Usual laboratory equipment and, in particular, the following.
5.1 Luminometer, capable of operating at a wavelength of 540 nm, with linear outputs being converted by ®

internal software into enzyme activity [e.g. Charm Luminometer models NovaLum, Luminator K or T ].
A vial adapter is needed with NovaLum and Lum-T model. NovaLum and Lum-T should be used in the
propped up position. A temperature probe supported by NovaLum should be used to measure the stopping
solution (4.2) temperature.
Measurements should be optimized according to the manufacturer's recommendations for the apparatus used.
5.2 Mini-vials, disposable, made of non-luminescent plastic, with caps, and of capacity 2 ml.
5.3 Fixed volume pipette, of capacity 100 µl.
5.4 Fixed-volume dispenser, capable of dispensing 1,0 ml. Check that the volume of water dispensed is
accurate to 1,00 g ± 0,05 g before use.
5.5 One-mark volumetric flasks, of capacity 100 ml.
5.6 Analytical balance, capable of weighing to the nearest 1 mg.
5.7 Incubator block or dry well bath, capable of operating at 35 °C ± 1 °C and 63 °C ± 0,2 °C with wells of
mini-vial dimensions.
5.8 Water bath, adjustable, capable of operating at 63 °C ± 0,2 °C and 95 °C ± 2 °C.
5.9 Test tubes, of capacity 50 ml, of diameter 13 mm and length 100 mm, with leakproof caps.
5.10 Printer, with connection cables for printout of results.
6 Sampling
A representative sample should have been sent to the laboratory. It should not have been damaged or
changed during transport or storage.
Sampling is not part of the method specified in this International Standard. A recommended sampling method
is given in ISO 707⎪IDF 50.

7 Preparations
7.1 Test samples
7.1.1 General
Carefully mix all test samples prior to use. The ambient temperature of the room should be between 18 °C and
24°C.
Samples should be tested at refrigeration temperature (0 to 7) °C or on ice during assay.
7.1.2 Pasteurized samples
Use the test samples as obtained in amounts as required.
7.1.3 Raw milk
Pipette 1 ml (or an appropriate amount such that the activity after dilution is less than 7 000 mU/l) of test
sample into a 100 ml one-mark volumetric flask (5.5). Dilute to the mark with alkaline phosphatase-free milk
(7.2). Mix carefully.
7.1.4 Flavoured-milk products and cream products
Use the test samples as obtained in amounts as required. Test samples of viscous products may require a
pipetting accuracy determination using mass (100 µl = 100 mg). Wipe the pipette tips after drawing the sample.
Possibly, modification of the pipette tip (cutting wider opening) might be required for accurate dispensing.
7.2 Alkaline phosphatase-free milk
Prepare alkaline phosphatase-free milk (negative test sample) by heating 35 ml or necessary volume of a test
portion (7.1.2, 7.1.3 or 7.1.4) in a tube in a water bath (5.8) set at 95 °C. Allow the test portion to reach 95 °C
and keep it at this temperature for 1 min. Then cool rapidly.
The negative sample is used as the 0 (zero) calibrator and shall have a mean value of less than 5 mU/l (or
less than 15 mU/l with flavoured-milk products and cream products) in the appropriately calibrated
luminometer channel. If stored at 4 °C, the negative sample may be kept for 48 h.
Fluid white milks may be kept for up to 6 months if stored frozen at or below −15 °C. Thaw the negative
sample in water at room temperature. Vigorously shake the sample to make it homogeneous before use. Do
not re-freeze.
It should be noted that some milk products, such as sheep milk, will precipitate and separate when the
negative test sample is prepared at 95 C for 1 min. In this case, a lower temperature for a longer time is
required, e.g. 63 °C for 30 min.
8 Procedure (see Annex A)
8.1 Calibration
8.1.1 Establish a calibration curve for each type of product to be tested. Begin by setting the luminometer
background (B ) to 100 and the correction (C ) to 100.

Consult the luminometer manual for instructions on making adjustments. Calibration curves are stable and
need to be run when new batch lots/lot numbers of the dioxetane ester substrate (4.1) and stopping buffer
(4.2) are used. A flowchart for the calibration is given in Annex A.

NOTE The calibration mode on some luminometers automatically guides users through steps 8.1.1 to 8.2.2. (The
calibration menu is item 8 of the main menu.) Select alkaline phosphatase calibration. Select channel to calibrate. Select
calibrate. Select test portion to calibrate, for example milk, cream or chocolate (flavoured-milk product) or other. The
luminometer prompts the user for the next steps.
8.1.2 Limit calibration assays to three tubes per assay. Use the fixed-volume pipette (5.3), provided with a
clean tip, to add 100 µl of the dioxetane ester substrate (4.1) to the bottom of three mini-vials (5.2).
8.1.3 For the test portions obtained in 7.1.2 to 7.1.4, add, with the fixed-volume pipette (5.3) provided with a
clean tip, 100 µl of negative test sample (7.2) to each vial. Deliver the contents of the sample to the bottom of
the tube to ensure all of the sample contacts the substrate. Mix the contents of the vials in the vial rack with a
back and forth motion for about 10 times in 5 s.
8.1.4 Place the mini-vials (8.1.3) in the incubator block (5.7) set at 35 °C for 3 min. At the end of incubation,
add, with the fixed-volume dispenser (5.4), within 15 s, 1,0 ml of stopping solution (4.2) to the contents of all
mini-vials.
8.1.5 Remove the mini-vials from the incubator. Cap each vial and shake each vigorously for 5 s.
If necessary unscrew the cap and attach the vial to the luminometer adapter. Insert the vial into the
luminometer. Read and record the readings of the luminometer (5.1) at count completion (beep). Repeat with
each vial. Do not touch the liquid solution with the adapter to avoid contaminating the next vial. If the solution
does contact the vial, rinse it with water and dry before re-use. Calculate the mean negative value (N). Repeat
the determination specified in 8.1.2 to 8.1.5 if any one value varies by more than 30 % of the mean.
8.1.6 For the test portions obtained in 7.1.2 to 7.1.3, repeat steps 8.1.2 through 8.1.5 with calibrator solution
C (4.3.1), each in triplicate. For the test portion obtained in 7.1.4, repeat steps 8.1.2 through 8.1.5 with
calibrator solution C (4.3.2), each in triplicate. Calculate the mean calibrator C or C value (C ). Repeat this

8.1.7 Calculate the correction value, C , using the mean calibration value, C (see 8.1.6), and the mean

1 Domaine d'application
La présente Norme internationale spécifie une méthode de détermination de l'activité de la phosphatase
alcaline dans les laits pasteurisés entier, demi écrémé, écrémé, la crème et les laits aromatisés, en utilisant
une méthode chimiluminescente par photoactivation enzymatique (EPAS).
La présente méthode est applicable au lait de vache, de brebis, de bufflonne et de chèvre ainsi qu'aux
boissons à base de lait de ces espèces.
Cette méthode convient également pour tout échantillon à base liquide, dilué de sorte que l'activité de la
phosphatase alcaline diluée présente une teneur inférieure à 7 000 milliunités par litre.
NOTE L'étude collaborative de cette méthode a été réalisée avec succès sur du lait entier de vache, de brebis, de
bufflonne et de chèvre, ainsi que sur du lait de vache écrémé (< 0,5 % de matière grasse), de la crème à 20 % de matière
grasse et du lait chocolaté à 2 % de matière grasse (toutes fractions massiques).
2 Termes et définitions
Pour les besoins du présent document, les termes et définitions suivants s'appliquent.
2.1

2.2

3 Principe
L'activité de la phosphatase alcaline est mesurée par photo-activation du produit hydrolysé, suivie d'un
mesurage instrumental de la photoactivation. En présence de phosphatase alcaline, un substrat aromatique
dioxétane-phosphate stable est hydrolysé à (35 ± 1) °C pour donner un produit photoactivé
(chimiluminescent). La photoactivation du produit est amplifiée par un activateur macromoléculaire. La
réaction d'hydrolyse est arrêtée après un temps d'incubation défini (3 min). La quantité de produit
chimiluminescent ainsi obtenue est mesurée et convertie en unités enzymatiques par un luminomètre.
L'étalonnage du luminomètre s'effectue en utilisant des tablettes d'activité enzymatique connue.

4 Réactifs
Utiliser uniquement des réactifs de qualité analytique reconnue, sauf spécification contraire, et de l'eau
distillée ou déminéralisée ou bien de l'eau de pureté équivalente.
4.1 Substrat d'ester de dioxétane non chimiluminescent, [0,2 mol/l 3-(2'-spiroadamantanane)-4-
méthoxy-4-(3''-phényl phosphate-1,2 sel de disodium de dioxétane dans une solution tampon DEAE à 1 % de

fluorosine] est disponible dans le commerce [liquide Charm reagent AP , par exemple].
Il est recommandé de stocker le substrat à une température comprise entre 0 °C et 7 °C. Si stocké dans des
flacons en plastique ambré et à 4 °C, le substrat reste stable pendant 6 mois. S'il est conservé à 30 °C, le
substrat ne reste stable que pendant 24 h.
Lorsqu'on utilise le substrat dans le cadre de l'essai, le maintenir à une température comprise entre 0 °C et
7 °C ou sur de la glace.
4.2 Solution d'arrêt
Préparer la solution d'arrêt en mélangeant la même quantité de 0,15 mol de 2-amino-2-méthyl-1-propanol et
de chlorure de benzalkonium à 0,02 %, pH 10,7. La solution d'arrêt doit être à température ambiante (18 °C à
24 °C) avant utilisation. Pour assurer la cohérence de l'essai, lorsque l'on n'utilise pas de sonde de
température, enregistrer et maintenir la température à la température de la solution utilisée lors de
l'étalonnage, à 0,5 °C près.
NOTE La solution d'arrêt stoppe l'hydrolyse enzymatique du substrat d'ester de dioxétane non chimiluminescent (4.1).
La solution d'arrêt disponible dans le commerce peut être conservée pendant 1 an à 4 °C ou 2 mois à
température ambiante. Pour une utilisation quotidienne, il est recommandé de conserver la solution à
température ambiante.
4.3 Étalons de travail
Comme exemple d'étalons de travail, on peut citer les tablettes d'étalonnage (lait cru sec ayant une teneur
mesurée en phosphatase, sous forme de tablettes à réhydrater dans du lait) ayant une activité
phosphatasique de 875 µU/l ± 26 µU/l. Rehydrater les tablettes dans trois volumes différents de boisson à
base de lait ne présentant aucune activité phosphatasique ou dans un échantillon pour essai négatif (7.2) afin
de créer une courbe d'étalonnage.
Les tablettes d'étalonnage disponibles dans le commerce peuvent être conservées à 4 °C pendant 2 ans.
4.3.1 Étalons pour les laits liquides non aromatisés
Dans trois tubes à essai de 50 ml chacun (5.9), marqués A , B et C , dissoudre respectivement une tablette

d'étalonnage dans 100 µl d'eau distillée.
Ajouter 20 ml au tube A , 5 ml au tube B et 2,5 ml au tube C de produit laitier liquide non aromatisé

(exempts d'activité phosphatasique) ou d'échantillon pour essai négatif (7.2). On obtient ainsi des étalons A ,
B et C présentant respectivement une activité phosphatasique de A = 44 mU/l, B = 175 mU/l et

C = 350 mU/l. Boucher les tubes et les agiter vigoureusement. Laisser le contenu des tubes se réhydrater au
réfrigérateur pendant 10 min. Bien mélanger avant emploi.

4.3.2 Étalons pour la crème et le lait aromatisé
Dans trois tubes à essai de 50 ml chacun (5.9), marqués A , B et C , dissoudre respectivement une tablette

d'étalonnage dans 100 µl d'eau distillée.
Ajouter 10 ml au tube A , 5 ml au tube B et 2,5 ml au tube C de crème ou de produit laitier aromatisé

(exempts d'activité phosphatasique) ou d'échantillon pour essai négatif (7.2) de façon à obtenir des étalons A ,
B et C présentant respectivement une activité phosphatasique de A = 88 mU/l, B = 175 mU/l et

C = 350 mU/l. Boucher les tubes et les agiter vigoureusement. Laisser le contenu des tubes se réhydrater au
réfrigérateur pendant 10 min. Bien mélanger avant emploi.
4.4 Témoin positif
Comme témoin positif, utiliser de la phosphatase alcaline sèche, congelée et placée dans une bouteille
ambrée de 15 ml. Réhydrater le témoin positif avec 10 ml d’une boisson représentative à base de lait exempte
d'activité phosphatasique, ou avec un échantillon pour essai négatif préalablement préparé (7.2). Le témoin
positif réhydraté contient 450 mU/l d'enzyme phosphatase.
Laisser le témoin reposer pendant 10 min pour permettre sa réhydratation. Bien agiter avant emploi.
Lorsque le témoin positif réhydraté est conservé entre 0 °C et 7 °C, il reste stable pendant 48 h. S'il est
réhydraté à l'aide de lait liquide, le témoin positif congelé à une température inférieure ou égale à – 15 °C
reste stable pendant 2 mois. Décongeler le témoin positif dans de l'eau à température ambiante. L'agiter
vigoureusement pour l'homogénéiser avant l'emploi. Ne pas recongeler.
5 Appareillage
Matériel courant de laboratoire et, en particulier, ce qui suit.
5.1 Luminomètre, pouvant fonctionner à une longueur d'onde de 540 nm, dont les sorties linéaires sont ®
converties par logiciel intégré en activité enzymatique (par exemple modèles de luminomètres Charm
1)
NovaLum, Luminator K ou T Charm . Un raccord de flacon est nécessaire avec les modèles NovaLum et
Lum T. Les modèles NovaLum et Lum T sont utilisés avec un support. Il est recommandé d'utiliser une sonde
de température prévue par le modèle NovaLum pour mesurer la température de la solution d'arrêt (4.2).
Il est recommandé d'optimiser les mesurages selon les recommandations du fabricant de l'appareillage utilisé.
5.2 Miniflacons, jetables, en plastique non luminescent, pourvus de bouchons et ayant une capacité
de 2 ml.
5.3 Pipette à volume fixe, d'une capacité de 100 µl.
5.4 Distributeur à volume fixe, capable de distribuer 1,0 ml. Vérifier que le volume d'eau délivré est précis
à 1,00 g ± 0,05 g avant emploi.
5.5 Fioles jaugées à un trait, d'une capacité de 100 ml.
5.6 Balance analytique, pouvant peser à 1 mg près.
5.7 Bloc d'incubation ou bain à puits sec, pouvant fonctionner à 35 °C ± 1 °C et 63 °C ± 0,2 °C, dont les
puits ont la dimension des miniflacons.
5.8 Bain d'eau, réglable, pouvant fonctionner à 63 °C ± 0,2 °C et à 95 °C ± 2 °C.
5.9 Tubes à essai, d'une capacité de 50 ml, de 13 mm de diamètre et de 100 mm de longueur, munis de
bouchons étanches.
5.10 Imprimante, avec câbles de raccordement pour imprimer les résultats.

6 Échantillonnage
Il est recommandé d'envoyer un échantillon représentatif au laboratoire. Il convient que cet échantillon ne soit
pas endommagé, ni modifié lors du transport ou de l'entreposage.
L'échantillonnage ne fait pas partie de la méthode indiquée dans la présente Norme internationale. Une
méthode d'échantillonnage recommandée est indiquée dans l'ISO 707⎪FIL 50.
7 Préparations
7.1 Échantillons pour essai
7.1.1 Généralités
Mélanger soigneusement tous les échantillons pour essai avant emploi. Il convient que la température
ambiante de la pièce soit comprise entre 18 °C et 24 °C.
Il convient que les échantillons soient soumis à l'essai à la température de réfrigération (de 0 °C à 7 °C) ou
soient posés sur de la glace pendant l'essai.
7.1.2 Échantillons pasteurisés
Utiliser les échantillons pour essai tels quels et dans les quantités nécessaires.
7.1.3 Lait cru
À l'aide d'une pipette, prélever 1 ml (ou le volume permettant d'obtenir, après dilution, une activité inférieure
à 7 000 mU/l) d'échantillon pour essai et le déposer dans une fiole jaugée à un trait de 100 ml (5.5). Diluer à
la marque avec du lait exempt de phosphatase alcaline (7.2). Mélanger soigneusement.
7.1.4 Produits laitiers aromatisés et produits à base de crème
Utiliser les échantillons pour essai tels quels et dans les quantités nécessaires. Les échantillons pour essai de
produits visqueux sont susceptibles de nécessiter une détermination de la précision du pipetage en utilisant la
masse (100 µl = 100 mg). Essuyer l'embout des pipettes après prélèvement de l'échantillon. Il peut
éventuellement être nécessaire de le modifier (en réalisant une ouverture plus large) afin de permettre une
distribution précise.
7.2 Lait exempt de phosphatase alcaline
Préparer du lait exempt de phosphatase alcaline (échantillon pour essai négatif) en chauffant 35 ml ou le
volume nécessaire de prise d'essai (7.1.2, 7.1.3 ou 7.1.4) dans un tube placé dans un bain d'eau (5.8) porté à
95 °C. Laisser la prise d'essai atteindre 95 °C et maintenir cette température pendant 1 min. Refroidir
rapidement.
L'échantillon négatif est utilisé comme étalon 0 (zéro) et doit avoir une valeur moyenne inférieure à 5 mU/l (ou
inférieure à 15 mU/l pour les produits laitiers aromatisés et les produits à base de crème) dans le canal,
correctement étalonné, du luminomètre. À une température de 4 °C, l'échantillon négatif peut être conservé
48 h.
Il est possible de conserver les laits liquides non aromatisés jusqu'à 6 mois en les congelant à une
température inférieure ou égale à – 15 °C. Décongeler le témoin négatif dans de l'eau à température
ambiante. L'agiter vigoureusement pour l'homogénéiser avant l'emploi. Ne pas recongeler.
Il convient de noter que certains produits laitiers comme le lait de brebis précipitent et se séparent lors de la
préparation de l'échantillon pour essai négatif à 95 °C pendant 1 min. Dans ce cas, une température inférieure
est nécessaire pendant une durée plus longue, par exemple 63 °C pendant 30 min.

8 Mode opératoire (voir Annexe A)
8.1 Étalonnage
8.1.1 Établir une courbe d'étalonnage pour chaque type de produit devant être soumis à l'essai.
Commencer par régler le fond du luminomètre (B ) à 100 et la correction (C ) à 100.

Consulter le manuel du luminomètre à propos des réglages. Les courbes d'étalonnage sont stables et doivent
être exécutées à chaque utilisation d'un nouveau lot ou un nouveau numéro de lot de substrat d'ester de
dioxétane non chimiluminescent (4.1) et d'une nouvelle solution tampon d'arrêt (4.2). Un schéma fonctionnel
représentant l'étalonnage est donné dans l'Annexe A.
NOTE Sur certains luminomètres, le mode d'étalonnage guide automatiquement l'utilisateur de l'étape 8.1.1 à
l'étape 8.2.2. (Le menu d'étalonnage est indiqué au point 8 du menu principal.) Sélectionner «étalonnage de la
phosphatase alcaline». Sélectionner «canal à étalonner». Sélectionner «étalonner». Sélectionner «prise d'essai à
étalonner», par exemple lait, crème ou chocolat (produit laitier aromatisé) ou autre. Le luminomètre guide l'utilisateur pour
les étapes suivantes.
8.1.2 Limiter les essais d'étalonnage à trois tubes par essai. Utiliser la pipette à volume fixe (5.3) munie
d'un embout propre pour ajouter 100 µl de substrat d'ester de dioxétane non chimiluminescent (4.1) au fond
des trois miniflacons (5.2).
8.1.3 Pour les prises d'essai obtenues de 7.1.2 à 7.1.4, ajouter, à l'aide de la pipette à volume fixe (5.3)
dont l'embout doit être propre, 100 µl d'échantillon pour essai négatif (7.2) dans chaque flacon. Introduire la
quantité d'échantillon jusqu'au fond du tube de façon à garantir que la totalité de l'échantillon est en contact
avec le substrat. Mélanger le contenu des flacons en place sur le porte-flacons en imprimant pendant environ
dix reprises et en 5 s un mouvement de va-et-vient.
8.1.4 Placer les miniflacons (8.1.3) dans l'incubateur (5.7) réglé à 35 °C pendant 3 min. Dans les 15 s qui
suivent la fin de l'incubation, ajouter à l'aide du distributeur à volume fixe (5.4) 1,0 ml de solution d'arrêt (4.2)
au contenu de tous les miniflacons.
8.1.5 Sortir les miniflacons de l'incubateur. Les boucher et les agiter vigoureusement pendant 5 s. Si
nécessaire, dévisser le bouchon et ajuster le flacon sur le raccord du luminomètre. Introduire le flacon dans le
luminomètre. Relever et enregistrer les valeurs du luminomètre (5.1) à la fin du comptage (signal sonore).
Répéter ce mode opératoire avec chaque flacon. Ne pas mettre le raccord en contact avec la solution de
liquide pour éviter de contaminer le flacon suivant. Si, malgré tout, la solution entre en contact avec le flacon,
rincer celui-ci à l'eau et le sécher avant réutilisation. Calculer la valeur négative moyenne (N). Si l'une des
valeurs obtenues diffère de plus de 30 % de la moyenne, recommencer la détermination spécifiée de 8.1.2 à
8.1.5.
8.1.6 Pour les prises d'essai obtenues de 7.1.2 à 7.1.3, répéter à trois reprises les étapes 8.1.2 à 8.1.5 en
utilisant la solution d'étalonnage C (4.3.1). Pour la prise d'essai obtenue en 7.1.4, répéter à trois reprises les
étapes 8.1.2 à 8.1.5 en utilisant la solution d'étalonnage C (4.3.2). Calculer la valeur moyenne de l'étalon C

ou C (C ). Si l'une des valeurs obtenues diffère de plus de 30 % de la moyenne, recommencer la

8.1.7 Calculer la valeur de correction , C , en utilisant la valeur moyenne de l'étalon, C , (voir 8.1.6) et la
