SIST EN ISO 15301:2001 is the Slovenian adoption of EN ISO 15301:2001, an ISO method for measuring sediment in crude animal and vegetable fats and oils by centrifugation. It tells laboratories how to prepare the sample, run the centrifuge test, read the sediment layer, and calculate the sediment content for crude oils and fats obtained by extraction and/or crushing.

What does SIST EN ISO 15301:2001 specify?

SIST EN ISO 15301:2001 specifies a laboratory method for determining the sediment in crude fats or oils that can be separated by centrifugal force. It is intended for crude oils in the stated sediment range and is not used for fats that are not liquid at 20 C.

The document is organized into clauses 1 to 11 covering scope, normative reference, term and definition, principle, apparatus, sampling, sample preparation, procedure, expression of results, precision, and test report. It also includes annexes for calculation, precision evidence, and European reference mapping.

AnnexWhat it covers
Annex ACalculation of the rotational frequency of the centrifuge
Annex BInterlaboratory test results and comparison with the 96 h method
Annex ZANormative references to international publications with their European publications

What are the key requirements of SIST EN ISO 15301:2001?

SIST EN ISO 15301:2001 is a duplicate-laboratory centrifuge test. The most important requirements are sample preparation, controlled centrifuging, reading the sediment layer correctly, and reporting the result in a traceable way.

Sample handling and preparation

Clause 6 says the laboratory should receive a representative sample that has not been changed during transport or storage, and the sample should be stored in glass or polyethylene terephthalate (PET) bottles. Clause 7 requires preparation of the test sample in accordance with ISO 661, then warming it if needed to 20 C to 25 C and redistributing any settled material before testing. In practice, this matters because the sediment must be part of the test portion, not left behind in the bottle.

Centrifuge setup and tubes

Clause 5 specifies 100 ml centrifuge tubes that are pear-shaped or cone-shaped, made of thoroughly annealed glass and fitted with a stopper, together with fat- and oil-resistant buckets and a controllable centrifuge. The centrifuge must deliver the stated radial acceleration at the narrow part of the tubes, and Annex A gives the calculation for rotational frequency from the swing diameter. For the user, this means the centrifuge settings are part of the method and must be known, not guessed.

Procedure and reading the result

Clause 8 calls for two tubes, each filled with 100 ml of prepared test sample, then centrifuged for 1 h 5 s. The sediment volume is read from the tube, with different reading accuracy depending on the amount, and the result should be corrected if separation is incomplete in a pear-shaped tube. The method also requires recording the centrifuge acceleration or swing diameter and frequency, plus the temperature before and after centrifuging, so the test can be checked later.

Result expression and precision

Clause 9 converts the measured sediment volume into sediment content using the tube mass and the test portion mass, then averages the two tube results and reports the final value to the nearest 1 ml per 100 g. Clause 10 gives repeatability and reproducibility limits based on interlaboratory testing, with details in Annex B. In practice, this tells the laboratory how to judge whether two results are close enough to agree.

Test report

Clause 11 lists the information that must be recorded in the report: sample identification, known sampling method, method reference, centrifuge settings, temperatures, any nonstandard operating details or incidents, and the result or the two results if repeatability was checked. That makes the result usable for quality control, dispute checking, or comparison with another laboratory.

What terms does SIST EN ISO 15301:2001 define?

Sediment is the insoluble material in a crude fat or oil that can be separated by centrifuging and collected as the unclear layer at the bottom of the tube.

Crude fats or oils are the materials to which the method applies when they are obtained by extraction and/or crushing and remain liquid at 20 C.

Relative radial acceleration is the centrifugal acceleration used to set the centrifuge performance at the narrow part of the tube.

Repeatability is the closeness of two results obtained in the same laboratory by the same operator with the same equipment over a short time.

Reproducibility is the closeness of two results obtained in different laboratories by different operators using different equipment.

Test report is the written record of the sample, method, settings, temperatures, observations, and final result.

Who uses SIST EN ISO 15301:2001?

SIST EN ISO 15301:2001 is used by laboratories testing crude animal and vegetable fats and oils, especially in the fats-and-oils and oilseed sectors. It is relevant to quality-control staff, laboratory managers, and analysts who need a fast check of sediment after extraction or crushing. It is also useful where results must be compared between laboratories or documented for trade and internal quality control.

Which standards are used with SIST EN ISO 15301:2001?

ISO 661 is the main normative reference and gives the preparation of the test sample. ISO 5555 is cited for recommended sampling, even though sampling itself is outside the method. ISO 5725-2 is used for the interlaboratory testing approach reported in Annex B, and Annex ZA maps the international references to their European publications.

What does the SIST EN ISO 15301:2001 document contain?

  • Two tube drawings: a pear-shaped tube and a cone-shaped tube.
  • A detailed procedure for weighing tubes, adding 100 ml of sample, centrifuging, and reading the sediment layer.
  • Annex A with the calculation for centrifuge rotational frequency.
  • Annex B with interlaboratory precision tables and a comparison with the 96 h standing method.
  • A test-report list that supports routine testing, method validation, and audit trail documentation.

Foreword
ISO (the International Organization for Standardization) is a worldwide federation of national standards bodies (ISO
member bodies). The work of preparing International Standards is normally carried out through ISO technical
committees. Each member body interested in a subject for which a technical committee has been established has
the right to be represented on that committee. International organizations, governmental and non-governmental, in
liaison with ISO, also take part in the work. ISO collaborates closely with the International Electrotechnical
Commission (IEC) on all matters of electrotechnical standardization.
International Standards are drafted in accordance with the rules given in the ISO/IEC Directives, Part 3.
Draft International Standards adopted by the technical committees are circulated to the member bodies for voting.
Publication as an International Standard requires approval by at least 75 % of the member bodies casting a vote.
Attention is drawn to the possibility that some of the elements of this International Standard may be the subject of
patent rights. ISO shall not be held responsible for identifying any or all such patent rights.
International Standard ISO 15301 was prepared by Technical Committee ISO/TC 34, Food products, Subcommittee
