Leather - Chemical determination of chromic oxide content - Part 2: Quantification by colorimetric determination (ISO 5398-2:2009)

ISO 5398-2:2009 describes the determination of chrome by colorimetric means. It is applicable to leathers which are expected to have chromic oxide contents in excess of 0,05 %. This is an analysis for total chromium in leather; it is not compound specific or specific to its oxidation state.

Overview

EN ISO 5398-2:2009 - Leather - Chemical determination of chromic oxide content - Part 2: Quantification by colorimetric determination - specifies a colorimetric laboratory method to quantify chromic oxide (Cr2O3) in leather. It is intended for leathers expected to contain chromic oxide in excess of 0.05% (w/w, dry basis). The method determines total chromium reported as chromic oxide; it is not compound-specific and does not distinguish chromium oxidation states.

Key topics and technical requirements

  • Scope and purpose: Quantification of chromium in leather by colorimetric means (spectrophotometry), suitable above 0.05% Cr2O3.
  • Sample preparation: Follow ISO sampling and grinding guidance (ISO 2418, ISO 4044) and weigh representative ground samples (typical mass given in the standard).
  • Ashing and fusion:
    • Pre-ash and ash sample (document specifies ashing at 750 °C ± 50 °C for 4 h).
    • Perform an alkaline fusion of the ash using a fusion mixture (sodium carbonate, potassium carbonate, sodium tetraborate) to solubilize chromium as hexavalent species.
  • Colorimetric reaction:
    • Convert chromium to hexavalent state, react with 1,5-diphenylcarbazide to form diphenylcarbazone.
    • Measure absorbance at 540 nm using a spectrophotometer (minimum 3 decimal places) in 1 cm cuvettes.
  • Reagents and apparatus:
    • Analytical-grade chemicals; distilled water per ISO 3696.
    • Required apparatus includes crucibles, muffle furnace, filtration device with ash-free paper, and a spectrophotometer.
  • Calibration and quality control:
    • Use potassium dichromate standard solutions for calibration.
    • Observe specified timing (measure 15 ± 5 minutes after reagent addition) and stability notes (e.g., solution stable up to 4 days for >0.5% Cr2O3).
  • Result expression: Results reported as percent chromic oxide on a dry-matter basis; repeatability and reporting requirements are included.

Applications and users

  • Intended for leather testing laboratories, quality control teams in tanneries, material compliance and regulatory testing.
  • Practical uses include:
    • Verifying chromium tanning levels for product specifications.
    • Quality assurance during leather processing.
    • Regulatory and safety checks where chromium content must be quantified.
  • Suitable when moderate laboratory equipment is available (spectrophotometer rather than ICP or AAS).

Foreword
ISO (the International Organization for Standardization) is a worldwide federation of national standards bodies
(ISO member bodies). The work of preparing International Standards is normally carried out through ISO
technical committees. Each member body interested in a subject for which a technical committee has been
established has the right to be represented on that committee. International organizations, governmental and
non-governmental, in liaison with ISO, also take part in the work. ISO collaborates closely with the
International Electrotechnical Commission (IEC) on all matters of electrotechnical standardization.
International Standards are drafted in accordance with the rules given in the ISO/IEC Directives, Part 2.
The main task of technical committees is to prepare International Standards. Draft International Standards
adopted by the technical committees are circulated to the member bodies for voting. Publication as an
International Standard requires approval by at least 75 % of the member bodies casting a vote.
Attention is drawn to the possibility that some of the elements of this document may be the subject of patent
rights. ISO shall not be held responsible for identifying any or all such patent rights.
ISO 5398-2 was prepared by the European Committee for Standardization (CEN) Technical Committee
CEN/TC 289, Leather, in collaboration with the Chemical Tests Commission of the International Union of
Leather Technologists and Chemists Societies (IUC Commission, IULTCS), in accordance with the
Agreement on technical cooperation between ISO and CEN (Vienna Agreement). It is based on IUC 8
originally published in J. Soc. Leather Tech. Chem., 49, p. 17, 1965, and declared an official method of the
