Overview
ASTM D7817-12(2021) is the standard test method for the enumeration of yeast and mold in raceway brine, brine-cured hides and skins, and pre-charge raceway liquor. Developed by ASTM International, this standard addresses the detection and quantification of salt-tolerant (halophilic) yeast and mold - microorganisms known to cause significant deterioration in hides and skins during brining and storage. The method outlines procedures using specific diluents and selective agar media to accurately count these organisms, providing critical data for quality assurance in leather processing and related industries.
Key Topics
- Halophilic Yeast and Mold: Focuses on salt-tolerant species, equated as halophilic organisms under the test conditions, which are responsible for spoilage and degradation of brine-cured materials.
- Enumeration Method: Involves serial dilution of samples (such as raceway brine or brine-cured hides and skins), plating on selective agar containing sodium chloride and an antibiotic to suppress bacterial interference, followed by incubation at 20-25°C for 5 days.
- Result Reporting: Counts are expressed as colony-forming units (cfu) per gram of sample for both yeast and mold. Results can be combined as needed.
- Precision and Repeatability: The standard includes guidance on repeatability limits, ensuring the reliability of results within and between laboratories.
- Safety and Compliance: Users are advised to implement appropriate safety, health, and environmental practices, and to be aware of regulatory considerations.
Applications
ASTM D7817-12(2021) plays a crucial role in the leather and hide industry, offering practical benefits such as:
- Monitoring Brine Quality: Enables leather manufacturers and tanneries to regularly evaluate the microbiological quality of raceway brine, preventing the proliferation of spoilage organisms.
- Quality Control in Brine-Cured Hides and Skins: Ensures that raw materials meet microbiological standards before further processing, reducing the risk of product loss due to microbial damage.
- Research and Development: Useful for studies aiming to develop improved preservation methods or new antimicrobial treatments for hides and skins.
- Regulatory Compliance: Supports compliance with international best practices and industry requirements on microbiological safety and product quality.
Related Standards
- ASTM D6715 - Practice for Sampling and Preparation of Fresh or Salt-Preserved (Cured) Hides and Skins for Chemical and Physical Tests: Provides detailed procedures for sample collection, crucial for accurate microbial testing.
- ASTM E691 - Practice for Conducting an Interlaboratory Study to Determine the Precision of a Test Method: Offers statistical guidance on evaluating repeatability and reproducibility.
- ASTM E177 - Practice for Use of the Terms Precision and Bias in ASTM Test Methods: Defines key statistical terms referenced in result analysis.
Practical Value
Implementing ASTM D7817-12(2021) helps organizations maintain the integrity and value of brine-cured hides and skins by controlling the levels of yeast and mold. Consistent use of this test method enhances process control, supports product safety, and reduces economic losses from spoilage. The standard’s focus on salt-tolerant microorganisms addresses a critical need in the hide processing sector, providing a reliable tool for safeguarding product quality from source to finished leather goods.
