Leather - Determination of degradability by micro-organisms (ISO 20136:2017)

This document specifies a test method to determine the degree and rate of aerobic biodegradation of hides and skins of different animal origin, whether they are tanned or not, through the indirect determination of CO2 produced by the degradation of collagen.
The test material is exposed to an inoculum (activated sludge from tannery wastewater) in an aqueous medium.
The conditions established in this document correspond to optimum laboratory conditions to achieve the maximum level of biodegradation. However, they may not necessarily correspond to the optimum conditions or maximum level of biodegradation in the natural medium.
In general, the experimental procedure covers the determination of the degradation degree and rate of the material under controlled conditions, which allows the analysis of the evolved carbon dioxide produced throughout the test. For this purpose, the testing equipment complies with strict requirements with regard to flow, temperature and agitation control.
This method applies to the following materials:
— natural polymers of animal stroma (animal tissue/skins),
— animal hides and skins tanned (leather) using organic or inorganic tanning agents,
— leathers that, under testing conditions, do not inhibit the activity of microorganisms present in the
inoculum.

Dieses Dokument legt ein Prüfverfahren fest, mit dem durch die indirekte Bestimmung von CO2, das durch den Abbau von Kollagen entsteht, der Grad und die Geschwindigkeit des aeroben Bioabbaus von entweder gegerbten oder ungegerbten Fellen und Häuten unterschiedlichen tierischen Ursprungs bestimmt wird.
Die Prüfsubstanz wird einem Inokulum (Belebtschlamm aus Gerbereiabwässern) in einem wässrigen Medium ausgesetzt.
Die in diesem Dokument festgelegten Bedingungen entsprechen den optimalen Laborbedingungen zum Erreichen des maximalen Grades des Bioabbaus. Jedoch entsprechen sie möglicherweise nicht den optimalen Bedingungen oder dem maximalen Grad des Bioabbaus im natürlichen Medium.
Im Allgemeinen deckt das experimentelle Verfahren die Bestimmung des Grades und der Geschwindigkeit des Bioabbaus des Materials unter kontrollierten Bedingungen ab, welches durch die Analyse des freigesetzten produzierten Kohlenstoffdioxids ermöglicht wird, dass während der Prüfung gebildet wird. Für diesen Zweck entspricht die Prüfvorrichtung den strikten Anforderungen in Bezug auf die Regelung des Durchflusses, der Temperatur sowie der Bewegung.
Dieses Verfahren gilt für die folgenden Materialien:
-   natürliche Polymere von tierischem Stroma (tierisches Gewebe/tierische Häute);
-   tierische Felle und Häute, gegerbt (Leder) mit organischen oder anorganischen Gerbstoffen;
Leder, die unter Prüfbedingungen die Aktivität der im Inokulum vorhandenen Mikroorganismen nicht hemmen.

ISO 20136:2017 spécifie une méthode d'essai pour déterminer le degré et la vitesse de biodégradation aérobie de peaux de différents animaux, tannées ou non, par la détermination indirecte du CO2 produit par la dégradation du collagène.
Le matériau d'essai est exposé à un inoculum (boues activées d'eaux résiduaires de tannage) dans un milieu aqueux.
Les conditions établies dans le présent document correspondent aux conditions de laboratoire optimales pour obtenir le niveau maximal de biodégradation. Il se peut toutefois qu'elles ne correspondent pas aux conditions optimales ou au niveau maximal de biodégradation dans le milieu naturel.
De manière générale, le mode opératoire expérimental inclut la détermination du degré et de la vitesse de dégradation du matériau dans des conditions contrôlées, ce qui permet d'analyser le dégagement de dioxyde de carbone tout au long de l'essai. À cet effet, l'équipement d'essai répond à des exigences strictes concernant le contrôle du débit, de la température et de l'agitation.
La présente méthode s'applique aux matériaux suivants:
- les polymères naturels de stromas animaux (tissus/peaux d'animaux),
- les peaux d'animaux qui ont été tannées (cuir) en utilisant des agents de tannage organiques ou inorganiques,
- les cuirs qui, dans les conditions d'essai, n'ont pas d'effet inhibiteur sur l'activité des micro-organismes présents dans l'inoculum.

Ta dokument določa preskusno metodo za ugotavljanje stopnje in hitrosti aerobne biorazgradljivosti surovih kož različnega živalskega izvora, ustrojenih ali neustrojenih, z neposrednim določevanjem količine CO2, ki se proizvaja pri razgradnji kolagena.
Preskusni material je izpostavljen inokulumu (aktivno blato iz odpadne vode pri ustrojevanju) v vodnem mediju.
Pogoji, določeni v tem dokumentu, ustrezajo optimalnim laboratorijskim pogojem za dosego največje stopnje biorazgradljivosti. Vendar pa morda ne ustrezajo optimalnim pogojem ali največji stopnji biorazgradljivosti v naravnem mediju.
Na splošno preskusni postopek zajema ugotavljanje stopnje in hitrosti biorazgradljivosti materiala v nadzorovanih pogojih, kar omogoča analizo sproščenega ogljikovega dioksida, ki se proizvaja med preskusom. Za ta namen je oprema za preskušanje skladna s strogimi zahtevami glede toka, temperature in nadzora tresenja.
Ta metoda zadeva naslednje materiale:
– naravne polimere živalske strome (živalsko tkivo/kože),
– živalske surove kože, ustrojene(usnje) z organskimi ali anorganskimi sredstvi za strojenje,
– usnje, ki v preskusnih pogojih ne zavira delovanja mikroorganizmov, prisotnih v
inokulumu.

Introduction
One of the big problems faced by the footwear industry is waste treatment. Although this waste,
especially in the case of leather, is not considered hazardous by current legislation, it is however
produced in large quantities which present a problem for municipal landfill sites.
The aim of the tanning process is to avoid skin putrefaction and increase the resistence of the obtained
leather. For this purpose, chemical and biological agents are used which are involved in the denaturation
and hardening of the main stromal protein, collagen, thus also producing physicochemical changes in
the skin.
There is a wide range of different agents used for leather tanning, which can be based on organic
products, vegetable extracts or inorganic products, mostly metals.
The most used tanning agent in the footwear industry is Chromium (III), which gives the skin desirable
characteristics, such as elasticity, easy buffing and a good breathability and vapour permeability.
However, the traditional tanning industry, and especially chrome tanning, generates wastes that pose
an environmental threat. Also, chrome-tanned hides and skins have too long a lifespan, much larger
than the useful life of the final products. Therefore, the use of additives that are less harmful to the
environment and which generate products that have a certain ease of degradation, once the material
has achieved its purpose, would be preferred, thus minimising waste products.
Within this sector, the development of fast biodegradability quantification methods for leather that has
been treated with alternative tanning agents is needed in order to predict whether these materials are
more biodegradable than their predecessors. The methodology described in this document attempts to
allow the completion of this form of analysis in a test time of no more than 35 days.

5 Principle
5.1 Method A: assessment of biodegradation by manual titration
This test method determines the biodegradation percentage of tanned or untanned hides and skins
through the indirect measurement of CO evolved during the degradation of collagen, which is the

major constituent of the skin, by the action of the microorganisms present in tannery wastewater.
The CO evolved during the test is indirectly determined through the reaction of [Ba(OH) ] with

CO , which is precipitated as barium carbonate (BaCO ). The amount of CO evolved is determined

by titrating the remaining barium hydroxide with a 0,05 mol/l hydrochloric acid solution. These
measurements are taken on a daily basis throughout the test.
Biodegradability is assessed by indirectly measuring the CO evolved as a function of time and

calculating the biodegradation degree by the difference between the initial carbon percentage present
in collagen and the remaining soluble organic carbon content that has not been transformed into CO in
2
the course of the process.
The initial carbon percentage (C) present in the collagen under study is determined by the elemental
analysis of the test specimen. The biodegradation percentage does not include the amount of carbon
transformed into a new cellular biomass that has not been metabolized to carbon dioxide throughout
the test.
The tests shall be carried out using equipment able to provide the conditions needed to carry out the
test. Agitation, experiment temperature and CO -free air inflow should be controlled.

The test shall be carried out in duplicate in the presence of a positive control, which is made up of a
synthetic medium, microorganisms and collagen, and a negative control, which is made up only of a
synthetic medium and inoculum (activated sludge from tannery wastewater), allowing the assessment
of two different leather samples that can be evaluated in duplicate.
5.2 Method B: assessment of biodegradation by infrared detection
With this method, biodegradation is determined through the quantification of the CO evolved

throughout the degradation of collagen, by means of the direct IR detection and continuous monitoring
of the CO concentration. The equipment comprises a reaction unit made up of a closed set of

unidirectional gas flow recirculation tubes, an aerator immersed in the reaction fluid contained in
the reaction flask, a membrane pump for unidirectional flow that makes the gas go through the CO
2
concentration detector area, an IR sensor, and a data capture system connected to a computer.
This system is in its final development stage. The methodology will be added to this document
application at a later stage.
The initial percentage of carbon (C) present in the collagen under study is determined through the
elemental analysis of each sample. The percentage of biodegradation does not include the quantity of
carbon converted into a new cellular biomass which is not metabolised into carbon dioxide during the
course of the test.
The tests shall be carried out using equipment able to provide the conditions needed to carry out the
test. Agitation, experiment temperature and CO -free air inflow should be controlled.

The tests are conducted in duplicate in the presence of a positive control, composed of a synthetic
medium, microorganisms and collagen, and a negative control, composed only of a synthetic medium
and an inoculum, allowing the assessment of five different leather samples that can be evaluated in
duplicate.
6 Chemicals
The reagents employed in the tests are the same for the two methods used in this document (Method
A and Method B) only with some adjustments in the volume of the reaction flasks specific of each
methodology (method A: a final liquid volume of 2,68 l; method B: a final liquid volume of 1 l).

6.1 Deionised or ultrapure (Milli Q® ) water, free from toxic materials with resistivity >18 MΩ/cm.
6.2 Test medium: Use only analytical grade reagents.
6.2.1 Prepare synthetic stock solutions by dissolving each of the following in distilled water to 1 l:

6.3 Test specimens: Use collagen type I (Sigma or similar) as a positive control. Test specimens shall
be basically leather from the tanning industry used for the production of leather clothing.
6.4 Only for Method A: Barium hydroxide solution, 0,025 mol/l, is prepared dissolving 4,0 g
[Ba(OH) ] per litre of distilled water. Filter free of solid material, confirm molarity by titration with

standard acid, and store sealed as a clear solution to prevent absorption of CO from the air. It is

recommended that 5 l be prepared at a time when running a series of tests.
7 Apparatus and materials
The usual laboratory equipment and, in particular, the following:
7.1 Analytical balance, capable of reading to 0,000 1 g.
7.2 Pipettes, 5 ml to 25 ml capacity.
7.3 Micro-pipettes, 500 μl and 1 000 μl.
7.4 Volumetric flask, 1 l.
For each method, the following materials should be employed:
7.5 Method A: Assessment of biodegradation by manual titration
7.5.1 Biodegradation test equipment
The procedure is partially automated thanks to the equipment specially conceived for this test (see
Annex A and Figure A.1).
This equipment allows four test specimens to be analyzed in duplicate (two test specimens and two
controls). It also allows agitation, experiment temperature and CO -free air inflow to be controlled.

7.5.2 Autonomous CO -free air source, consisting of a noiseless compressor connected to a PSA

(pressure swing adsorption) system provided with a molecular sieve, from Peak Scientific, model PG14L.
7.5.3 Sepiolite to filter impurities and humidity from the ventilation system.
7.5.4 Test flasks

7.5.4.1 Eight 5 l Erlenmeyer® flasks (reaction flasks) for each test (two controls and two test
specimens per test). 5 000 ml H-SA V H40/45 Erlenmeyer® flasks shall be used, as well as V2 distilling
heads with GL18 threads and a filter pore no. 1 diffuser.
7.5.4.2 Connect the PSA equipment (7.5.2) to four glass flasks and two plastic flasks connected in
series using silicone tubing. The first three flasks shall contain 700 ml of 10 mol/l sodium hydroxide
(NaOH). The fourth flask shall be empty. The fifth flask shall contain 700 ml of 0,025 mol/l [Ba(OH) ].

7.6 Method B: Assessment of biodegradation by IR detection
7.6.1 Biodegradation equipment
The procedure is totally automated through equipment developed specially for these tests (see Annex
B and Figure B.1).
This equipment allows the analysis of up to seven samples, in duplicate, per test run (five samples and
two controls). The agitation is orbital and the temperature is controlled by an air cooling system. The
quantification of CO , produced during the leather biodegradation process, is carried out using CO
2 2
detection equipment that incorporate infrared sensors with a measuring range between 0 % and 5 %.
7.6.2 The supply of air (O ) free of carbon dioxide (CO ) is provided by a gas mixture of O :N at a ratio

of 30:70 injected directly into the reaction flasks for 30 min at a rate of 3 l/min.
7.6.3 Easy-to-use software for the capture, processing, and monitoring of signals, with the capacity to
store data for long periods of time.
7.6.4 The calibration of the CO detection equipment is carried out with special mixes of gases at

different concentrations of CO (1 %, 3 %, and 5 %) in addition to a gas mixture with 99,9 % O (oxygen

5,0) with zero CO concentration. At the end of the calibration, a linear calibration curve between 0 %
2
and 5 % is generated according to the linear equation of Y = AX + B and its respective coefficient of

determination (R ).
7.6.5 Calibration values are stored in a software program specially developed for these tests, which
additionally allows the test parameters to be controlled and all production data of CO generated during

the test in the different reaction flasks to be saved.
7.6.6 Test vessels
7.6.6.1 14 flasks with a useful volume of 2 l (reaction flasks) incorporating a distilling head and
an air diffuser which are used to conduct the tests (two controls and five samples in duplicate). The

Einleitung
Eines der großen Probleme der Schuhindustrie betrifft die Abfallbehandlung. Obwohl dieser Abfall, besonders
im Fall von Leder, durch die geltende Gesetzgebung als nicht gefährlich angesehen wird, fällt er jedoch in
großen Mengen an und stellt somit ein Problem für kommunale Mülldeponien dar.
Das Ziel des Gerbprozesses ist die Vermeidung der Fäulnis der Häute und die Erhöhung der Widerstands-
fähigkeit des erhaltenen Leders. Für diesen Zweck werden chemische und biologische Reagenzien ver-
wendet, die an der Denaturierung und Härtung des wichtigsten Gewebeproteins, dem Kollagen, beteiligt sind
und somit auch physikalisch-chemische Veränderungen in der Haut verursachen.
Es gibt eine Vielzahl von unterschiedlichen Mitteln, die in der Ledergerbung verwendet werden. Sie basieren
auf organischen Produkten, Pflanzenextrakten oder anorganischen Produkten, hauptsächlich Metalle.
Der am meisten in der Schuhindustrie verwendete Gerbstoff ist Chrom (III), das der Haut die gewünschten
Eigenschaften gibt, wie z. B. Elastizität, einfaches Schleifen (Polieren) sowie eine gute Atmungsfähigkeit und
Wasserdampfdurchlässigkeit. Jedoch erzeugt die traditionelle Gerbereiindustrie, und insbesondere die
Chromgerbung, Abfälle, die eine Umweltgefährdung darstellen. Außerdem haben chromgegerbte Felle und
Häute eine längere Lebensdauer, die wesentlich höher als die Nutzungsdauer der Endprodukte ist. Deshalb
wäre zur Verringerung der Abfallprodukte die Verwendung von Zusätzen zu bevorzugen, die weniger umwelt-
schädlich sind und die Produkte ergeben, die eine bestimmte leichtere Abbaubarkeit aufweisen, wenn das
Material seinen Zweck erfüllt hat.
In diesem Bereich ist die Entwicklung von schnellen Verfahren zur quantitativen Bestimmung der Bioab-
baubarkeit von mit alternativen Gerbstoffen behandeltem Leder für die Vorhersage notwendig, ob diese
Materialien leichter biologisch abbaubar als deren Vorgänger sind. Die in dieser Norm beschriebene Vor-
gehensweise versucht die Ausführung dieser Form der Analyse in einer Prüfzeit von nicht mehr als 35 Tagen
zu ermöglichen.

1 Anwendungsbereich
Diese Internationale Norm legt ein Prüfverfahren fest, mit dem durch die indirekte Bestimmung von CO , das

durch den Abbau von Kollagen entsteht, der Grad und die Geschwindigkeit des aeroben Bioabbaus von ent-
weder gegerbten oder ungegerbten Fellen und Häuten unterschiedlichen tierischen Ursprungs bestimmt wird.
Die Prüfsubstanz (das Leder) wird einem Inokulum aus Belebtschlamm (aktivierter Klärschlamm) in einem
wässrigen Medium ausgesetzt, wobei das Inokulum aus zum Gerben mit biologischen Mitteln genutzten
Tanks (Becken) stammt.
Die in dieser Internationalen Norm festgelegten Bedingungen entsprechen den optimalen Laborbedingungen
zum Erreichen des maximalen Grades des Bioabbaus. Jedoch entsprechen sie möglicherweise nicht den
optimalen Bedingungen oder dem maximalen Grad des Bioabbaus im natürlichen Medium.
Im Allgemeinen kann die Anwendung dieses Prüfverfahrens auf die Untersuchung des biologischen Abbaus
von Textilien und/oder von Polymermaterial, aus denen ein Schuh hergestellt ist, extrapoliert werden. Damit
das experimentelle Verfahren die Bestimmung des Grades und der Geschwindigkeit des Bioabbaus des
Materials unter kontrollierten Bedingungen durch die Analyse des freigesetzten Kohlendioxids, das während
der Prüfung gebildet wird, ermöglicht, muss die Prüfvorrichtung den strikten Anforderungen in Bezug auf die
Regelung des Durchflusses, der Temperatur sowie der Bewegung entsprechen.
Dieses Verfahren gilt für die folgenden Materialien:
 natürliche Polymere von tierischem Stroma (tierisches Gewebe/tierische Häute);
 tierische Felle und Häute, gegerbt (Leder) mit organischen oder anorganischen Gerbstoffen;
 Leder, die unter Prüfbedingungen die Aktivität der im Inokulum vorhandenen Mikroorganismen nicht
hemmen.

3 Kurzbeschreibung
3.1 Verfahren A: Beurteilung der Bioabbaubarkeit durch manuelle Titration
Dieses Prüfverfahren bestimmt den prozentualen Bioabbau von gegerbten und ungegerbten Fellen und
Häuten durch die indirekte Messung von CO , das während des Abbaus von Kollagen, dem Hauptbestandteil

der Haut, durch die Einwirkung der in den Gerbereiabwässern vorhandenen Mikroorganismen, gebildet wird.
Das während der Prüfung freigesetzte CO wird indirekt durch die Reaktion von Ba(OH) mit CO bestimmt,

das als Bariumcarbonat (BaCO ) ausfällt. Die Menge an gebildetem CO wird durch Titration des verbleiben-

den Bariumhydroxids mit 0,05 N Salzsäurelösung bestimmt. Diese Messungen werden täglich während der
gesamten Prüfung durchgeführt.
Die Bioabbaubarkeit wird durch die indirekte Messung des freigesetzten CO als Funktion der Zeit bestimmt

und der Grad des Bioabbaus durch die Differenz im Gehalt an löslichem organischem Kohlenstoff, der im
Laufe des Verfahrens nicht in CO umgewandelt wurde, berechnet.

Der im zu untersuchendem Kollagen vorhandene anfängliche prozentuale Kohlenstoffanteil (C ) wird durch

Elementaranalyse der Messprobe bestimmt. Der prozentuale Bioabbau schließt nicht die Menge an
Kohlenstoff ein, der in neue Zellmasse (Biomasse) umgewandelt und während der Prüfung nicht zu
Kohlendioxid umgesetzt wurde.

Die Prüfungen müssen mit der speziell für diese Analyse entwickelten Vorrichtung durchgeführt werden, die
die Analyse von 8 Proben je Prüfung sowie die Regelung/Kontrolle von Bewegung, Versuchstemperatur und
Zufuhr CO -freier Luft ermöglicht.

Die Prüfung muss in doppelter Ausführung in Gegenwart einer positiven Kontrolle, die aus einem
synthetischen Medium, Mikroorganismen und Kollagen hergestellt wird, sowie einer negativen Kontrolle, die
nur aus einem synthetischen Medium und dem Inokulum (Belebtschlamm aus Gerbereiabwässern) besteht,
durchgeführt werden. Somit ist eine Beurteilung von zwei verschiedenen Lederproben, die in einer
Doppelbestimmung analysiert wurden, möglich.
3.2 Verfahren B: Beurteilung der Bioabbaubarkeit durch Infrarot (IR)
Mit diesem Verfahren wird die Bioabbaubarkeit durch die quantitative Bestimmung des während des Abbaus
von Kollagen gebildeten CO mithilfe der direkten IR-Detektion und kontinuierlichen Überwachung der

CO -Konzentration bestimmt. Die Vorrichtung umfasst eine Reaktionseinheit, die aus einer geschlossenen

(dichten) Reihe von Schläuchen zur Rückführung des unidirektionalen Gasdurchflusses besteht, einen in die
Reaktionsflüssigkeit im Reaktionskolben eingetauchten Belüfter (Aerator), eine Membranpumpe für den
unidirektionalen Durchfluss, der das Durchströmen des Gases durch den Nachweisbereich für die
CO -Konzentration ermöglicht, einen IR-Sensor sowie ein mit einem Computer verbundenes

Datenerfassungssystem.
Dieses System befindet sich in seiner letzten Entwicklungsphase. Die Verfahrensweise wird zu einem
späteren Zeitpunkt der Anwendung dieser Internationalen Norm hinzugefügt.
Der im zu untersuchendem Kollagen vorhandene anfängliche prozentuale Kohlenstoffanteil (C ) wird durch

Elementaranalyse jeder Probe bestimmt. Der prozentuale Bioabbau schließt nicht die Menge an Kohlenstoff
ein, der in neue Zellmasse (Biomasse) umgewandelt und während der Prüfung nicht zu Kohlendioxid
umgesetzt wurde.
Die Prüfungen müssen mit der speziell für diese Analysen entwickelten Vorrichtung durchgeführt, die sowohl
die Analyse von bis zu 14 Proben je Prüfdurchgang als auch die Regelung/Kontrolle der Bewegung, der
Versuchstemperatur und die CO -freie Luftzufuhr (O ) ermöglicht.

Die Prüfungen werden in doppelter Ausführung in Gegenwart einer positiven Kontrolle, die aus einem
synthetischen Medium, Mikroorganismen und Kollagen zusammengesetzt ist, sowie einer negativen Kontrolle,
die nur aus einem synthetischen Medium und dem Inokulum (Belebtschlamm aus Gerbereiabwässern)
besteht, durchgeführt. Das ermöglicht die Beurteilung von fünf verschiedenen Lederproben, die in einer
Doppelbestimmung analysiert wurden.
4 Reagenzien
Die bei den Prüfungen verwendeten Reagenzien sind für die in dieser Norm beschriebenen Verfahren
(Verfahren A und Verfahren B) gleich, lediglich mit einigen Anpassungen an das Volumen der für jede
Verfahrensweise spezifischen Reaktionskolben (Verfahren A: ein endgültiges Flüssigkeitsvolumen von 2,68 l;
Verfahren B: ein endgültiges Flüssigkeitsvolumen von 1 l).
4.1 Deionisiertes oder ultrareines (Milli Q) Wasser, frei von toxischen Substanzen mit einem spezifischen
elektrischen Widerstand > 18 MΩ/cm.
4.2 Prüfmedium:
Es werden nur Reagenzien mit anerkannter Analysenreinheit verwendet.
4.2.1 Es werden synthetische Stammlösungen durch Auflösen der folgenden Reagenzien in destilliertem
Wasser bis auf 1 l hergestellt:

4.2.2 Das Prüfmedium muss aus den folgenden Reagenzien, verdünnt mit destilliertem Wasser bester
Qualität auf 1 l, bestehen:
4.2.2.1 Magnesiumsulfatlösung, 2 ml
4.2.2.2 Calciumchloridlösung, 2 ml
4.2.2.3 Phosphatpufferlösung, 4 ml
4.2.2.4 Eisenchloridlösung, 2 ml,
4.2.2.5 Ammoniumsulfatlösung, 2 ml
4.3 Prüfkörper:
Als positive Kontrolle wird Kollagen Typ I (Sigma oder ähnlich) verwendet. Prüfkörper (Messproben) müssen
grundsätzlich aus Leder aus der Gerbereiindustrie bestehen, das zur Herstellung von Lederbekleidung
verwendet wird.
4.4 Nur bei Verfahren A: Bariumhydroxidlösung, 0,025 N, wird durch Auflösen von 4,0 g Ba(OH) je Liter
2
destilliertes Wasser hergestellt. Festes Material in der Lösung wird abfiltriert, die Normalität (Konzentration)
wird durch Titration mit einer Säure-Standard-Lösung bestätigt und die Lösung wird als klare Lösung in einem
verschlossenen Kolben aufbewahrt, um die Absorption von CO aus der Luft zu verhindern. Es wird

empfohlen, dass 5 l zu dem Zeitpunkt hergestellt werden, an dem eine Reihe von Prüfungen durchgeführt
wird.
5 Prüfeinrichtung und Materialien
Die üblichen Laborgeräte:
5.1 Analysenwaage, geeignet für Ablesungen bis 0,000 1 g
5.2 Pipetten, Fassungsvermögen 5 ml bis 25 ml
5.3 Mikro-Pipetten, 500 µl und 1 000 µl

5.5 Verfahren A: Beurteilung der Bioabbaubarkeit durch manuelle Titration
5.5.1 Vorrichtung für die Prüfung Bioabbaubarkeit
Das Verfahren ist dank der speziell für diese Prüfung konzipierten Vorrichtung (siehe Anhang A, Bild A.1)
teilweise automatisiert.
Diese Vorrichtung ermöglicht die Analyse von 4 Messproben in doppelter Ausführung (2 Messproben und
2 Kontrollen). Sie ermöglicht ebenso die Regelung der Bewegung, der Versuchstemperatur und der Zufuhr
von CO -freier Luft.

5.5.2 Unabhängige Quelle für CO -freie Luft, bestehend aus einem geräuschlosen Kompressor, der mit
2
einem System zur Druckwechsel-Adsorption (PSA, en: pressure swing adsorption) verbunden ist, das mit
einem Molekularsieb, von Peak Scientific, Modell PG14L, ausgestattet ist.
5.5.3 Sepiolith, zum Herausfiltern von Verunreinigungen und Feuchtigkeit aus dem Belüftungssystem.
5.5.4 Prüfkolben
5.5.4.1 Acht 5-l-Erlenmeyerkolben (Reaktionskolben) für jede Prüfung (2 Kontrollen und 2 Messproben je
Prüfung). 5 000-ml-Erlenmeyerkolben, Modelltyp SAV H-40/45, müssen eingesetzt werden, sowie auch
V2-Destillieraufsätze mit GL-18-Gewinden und ein Poren-Diffusor Nr. 1.
5.5.4.3 Für jeden der Erlenmeyerkolben in 5.5.4.1 drei 0,25-l-Flaschen, die mit Silikonschläuchen in Reihe
verbunden sind und jeweils 100 ml Bariumhydroxidlösung, 0,025 N [BaOH ] enthalten, um CO zu binden

(Analysekolben).
5.5.4.2 Das PSA-Gerät (5.5.2) wird mit vier Kolben aus Glas und zwei Kolben aus Kunststoff mit Silikon-
schläuchen verbunden. Die ersten 3 Kolben müssen 700 ml 10 N Natriumhydroxid (NaOH) enthalten. Der
vierte Kolben muss leer sein. Der fünfte Kolben muss 700 ml 0,025 N [BaOH ] enthalten. Der sechste Kolben

muss leer sein.
5.5.4.4 Verschlüsse, für CO undurchlässiger flexibler Kunststoffschlauch, 100-ml-Büretten

5.5.4.5 Salzsäure, 0,05 N
5.6 Verfahren B: Beurteilung der Bioabbaubarkeit durch Infrarot (IR)
5.6.1 Vorrichtung für Bioabbaubarkeit
Das Verfahren ist durch die speziell für diese Prüfung konzipierte Vorrichtung (siehe Anhang B, Bild B.1)
vollständig automatisiert.
Diese Vorrichtung ermöglicht die Analyse von 7 Proben in doppelter Ausführung je Prüfdurchgang (5 Proben
und 2 Kontrollen). Die Bewegung ist kreisförmig und die Temperatur wird durch ein Luftkühlsystem geregelt.
Die quantitative Bestimmung des während des Prozesses des Lederabbaus entstehenden CO erfolgt durch

ein CO -Messgerät, das Infrarot-Sensoren mit einem Messbereich zwischen 0 % und 5 % enthält.

5.6.2 Die Versorgung mit Luft (O ), frei von Kohlendioxid (CO ), erfolgt mit einem Gasgemisch von O :N in
2 2 2 2
einem Verhältnis von 30:70, das für 30 min direkt in die Reaktionskolben mit einem Volumendurchfluss von
3 l/min eingeleitet wird.
5.6.3 Leicht anwendbare Software zur Erfassung, Verarbeitung und Überwachung von Signalen sowie mit
der Kapazität zur Speicherung von Daten über längere Zeiträume.

5.6.4 Die Kalibrierung des CO -Messgeräts erfolgt mit besonderen Gemischen von Gasen mit unter-

schiedlichen CO -Konzentrationen (1 %, 3 % und 5 %) zusammen mit einem Gasgemisch mit 99,9 % O
2 2
(Sauerstoff 5.0) mit einer CO -Konzentrationen von 0. Am Ende der Kalibrierung wird eine lineare Kalibrier-

kurve zwischen 0 % und 5 % entsprechend der linearen Gleichung von Y = AX + B und deren jeweiligem
2
Bestimmungskoeffizienten (R ) erstellt.
5.6.5 Die Kalibrierwerte werden in einem speziell für diese Prüfungen entwickelten Software-Programm
gespeichert, das es ermöglicht, zusätzlich die Prüfparameter zu regeln und alle Daten des während der
Prüfung in den verschiedenen Reaktionskolben gebildeten CO zu sichern.

5.7 CO -Nachweissystem und Datenerfassungssystem

Für jeden der Prüfkolben (6.2) ist ein CO -Messgerät erforderlich. Dieses CO -Messgerät wird mit einer

Erfassungs- und Signalverarbeitungseinheit verbunden, die die Signale von jedem Ausrüstungsteil verwalten
und diese an ein Computersystem zur Überwachung und Speicherung von Daten übermittelt. Auf diese Weise
werden die Werte des während der Prüfung in den Reaktionskolben insgesamt gebildeten CO auf einem

Computer gesichert und bleiben auf diesem verfügbar.
5.8 Prüfgefäße
5.8.1 14 Kolben mit einem Nutzvolumen von 2 l (Reaktionskolben), die einen Destillationsaufsatz und einen
Luftdiffusor umfassen, die zur Durchführung der Prüfungen (2 Kontrollen und 5 Proben in doppelter
Ausführung) genutzt werden. Die Erlenmeyerkolben müssen ein Fassungsvermögen von 2 000 ml und
3 Öffnungen (Anschlüsse) haben und dem Modelltyp SAV H-29/32 (SQ13) entsprechen. Sie müssen
V2-Destillationsaufsätze mit GL-14-Gewinden (6 mm am Lufteinlass und 8 mm am Luftauslass) und einen
Poren-Diffusor Nr. 1 haben. Das Volumen der Flüssigkeit (Kulturmedium + Inokulum) muss insgesamt 1 l
betragen.
5.8.2 Die Kolben müssen mit dem CO -Messgerät durch Nylon-Schläuche verbunden sein. Der Schlauch,

der den Auslass des CO -Messgeräts mit dem Einlassflansch des Destillationsaufsatzes bis zum Diffusor
2
verbindet, muss einen Durchmesser von 6 mm haben. Der Schlauch, der den Luftauslass des Kolbens mit
dem Einlass des CO -Messgeräts verbindet, sollte einen Durchmesser von 8 mm haben.

6 Durchführung
6.1 Sammlung und Herstellung des Inokulums
Es werden Proben (Abwasser) aus einem zum Gerben mit biologischen Mitteln belüfteten Tank (Becken) als
Inokulum verwendet. Die Probe muss frei von großen inerten Gegenständen sein, wie z. B. Lederstücken, die
manuell entfernt werden müssen.
Die Probe muss entnommen und unverzüglich in einem tragbaren Kühlbehälter an das Laboratorium einge-
sandt werden, damit deren ursprüngliche Eigenschaften erhalten bleiben. Zum Entfernen von Verunreini-
gungen wird die Probe dekantiert, und um die Menge an Schwebstoffen zu verringern, muss das Abwasser
über Glaswolle filtriert werden. Alternativ wird die Probe bei 1 500 U/min für 5 min zentrifugiert.
6.2 Herstellung der Prüfsubstanz und Referenzsubstanz
Die Aktivität des Inokulums wird während der Prüfung mithilfe einer biologisch abbaubaren Referenzsubstanz
überprüft, vorzugsweise mit Kollagen Typ 1 (Sigma oder ähnlich) in Form von Pulver und durch Messung der
CO -Bildung während dessen Abbaus. Die Referenzsubstanz muss am Ende der Prüfung zu 70 % oder mehr

abgebaut sein. Infolge der Möglichkeit, dass das Inokulum suspendierte organische Verbindungen enthält,
müssen Kolben, die das Inokulum und das Kulturmedium enthalten, als eine negative Kontrolle angewendet
werden. Die Werte für das in diesen Kolben gebildete CO müssen von den Werten, die sich in der positiven

Kontrolle und den Messproben gebildet haben, subtrahiert werden.

Für die Prüfungen der Bioabbaubarkeit müssen gegerbte Felle und Häute unterschiedlichen tierischen
Ursprungs, die üblicherweise in der Leder-, Polster- und Schuhindustrie eingesetzt werden, verwendet
werden.
6.3 Prüfbedingungen und Inkubationszeit
Sämtliche Prüfsubstanzen, Kolben, Kulturmedia usw. müssen vor der Verwendung autoklaviert werden.
Die gesamte Prüfdauer wird durch die Zeitdauer bestimmt, die für die positive Kontrolle (Kulturmedium +
Kollagen) erforderlich ist, um 70 % des maximalen Grades des Bioabbaus zu überschreiten.
Alle Messproben müssen in Form von Pulver eingebracht werden. Die Anfangskonzentration der Messproben
muss zwischen 0,18 g/l bis 0,19 g/l liegen, die absolute Menge für jede Prüfung ist 0,5 g (Verfahren A) und
0,18 g bis 0,19 g (Verfahren B).
6.4 Prüfvorrichtung
Die Prüfvorrichtung besteht aus kompakten (dichten) Einheiten, die speziell für die Prüfung der Bioabbau-
barkeit durch aerobe Mikroorganismen konzipiert wurden. Im Allgemeinen kann deren Anwendbarkeit auf die
Untersuchung des biologischen Abbaus von Textilien (Gewebe, Leder, Häute und Felle usw.) oder von
Polymermaterial, die in Schuhwaren enthalten sind, extrapoliert werden. Deshalb basiert das Prüfverfahren
auf der Grundlage der Bildung von CO während der Prüfung und auf der Bestimmung des Grades und der

Geschwindigkeit des Bioabbaus der Substanz unter kontrollierten Laborbedingungen. Für diesen Zweck
entspricht die Einheit den strikten Anforderungen in Bezug auf die Regelung des Durchflusses (CO -freie Luft,

bei Verfahren A geregelt durch ein PSA-System), die thermische Regelung sowie die Regelung der
Bewegung (hin- und hergehende Bewegung bei Verfahren A und kreisförmige Bewegung bei Verfahren B).
Diese Einheiten wurden entwickelt, um das experimentelle Verfahren zu vereinfachen.
6.4.1 Vorrichtung für die Beurteilung der Bioabbaubarkeit durch manuelle Titration (Verfahren A)
Der Prototyp funktioniert auf die Weise, dass die erzeugte Luft durch eine Reihe von sieben Erlenmeyer-
kolben (Vorbehandlungskolben) geleitet wird, die das verbliebene Kohlendioxid im aus dem PSA-Gerät
kommenden Luftstrom abfangen. Die Luft wird dann in 8 Leitungen aufgeteilt, die durch 8 Ventile geregelt
werden, die den Durchfluss unabhängig regeln können und wiederum 8 Erlenmeyerkolben (Reaktionskolben)
versorgen, die sich innerhalb des Behälters befinden. Der Auslass von jedem der 8 Erlenmeyerkolben ist
direkt mit einer Reihe von 3 Erlenmeyerkolben aus Glas (Analysekolben) verbunden, die jeweils 100 ml
Ba(OH) , 0,025 N, enthalten, aus denen die Ergebnisse ermittelt werden (siehe Anhang A, Bilder A.2 und

A.3).
Die Vorrichtung ist ebenfalls mit einem Thermostat ausgestattet, der die Regelung der Temperatur der
Reaktionskolben durch die Zirkulation von Wasser in einem geschlossenen Kreislauf ermöglicht. Die Prüfung
