Introduction
By using standard conditions of temperature, salinity and pH at low biocide concentrations in the surrounding
artificial seawater, a repeatable value of the release rate under the specified laboratory conditions can be
determined using the method given in this part of ISO 15181, which can be used for quality assurance and
material selection purposes. The actual release rate of biocides from antifouling paints on ships’ hulls into the
environment will, however, depend on many factors, such as ship operating schedules, length of service,
berthing conditions, paint condition, as well as temperature, salinity, pH, pollutants and biological community
in a particular area.
The results of this test do not reflect environmental biocide release rates for antifouling products and are not
suitable for direct use in the process of generating environmental-risk assessments, producing environmental-
loading estimates or for establishing release rate limits for regulatory purposes. In comparison with copper
and organotin release rates obtained either by direct or indirect measurements of the copper release rate from
ships’ hulls and from measurements made on panels exposed in harbours, all available data indicate that the
results obtained using this generic test method significantly overestimate the release rates of biocides under
in-service conditions. Published results demonstrate that the results of this test method are generally higher
by a factor of about 10 or more for several commercial antifouling coatings than direct in situ measurements of
[1, 2]
copper and organotin release rates from in-service ship hulls . A similar relationship is expected to be
found for other biocides. Realistic estimates of the biocide release rate from a ship’s hull under in-service
conditions can only be obtained from this test method if this difference is taken into account.
Where the results of this test method are used in the process of generating environmental-risk assessments,
producing environmental-loading estimates or for regulatory purposes, it is most strongly recommended that
the relationship between laboratory release rates and actual environmental inputs be taken into account to
allow a more accurate estimate of the biocide release rate from antifouling coatings under real-life conditions

to be obtained. This can be accomplished through the application of appropriate correction factors .

3 Principle
The amount of tolylfluanid released from a test cylinder into artificial seawater by the method given in
ISO 15181-1 is determined by promoting the degradation of the tolylfluanid in the leachate under controlled
conditions and subsequently quantifying the concentration of the degradation product, dimethyltolylsulfamide
(DMST), by a high-performance liquid chromatography (HPLC) method or by an alternative analytical method,
provided that it has a limit of quantitation for tolylfluanid in artificial seawater of 7 µg/l or less. The release rate
of the biocide under the specified laboratory conditions is then calculated as tolylfluanid.
The amount of dichlofluanid released from a test cylinder into artificial seawater by the method given in
ISO 15181-1 is determined by promoting the controlled degradation of the dichlofluanid in the leachate, and
quantifying the concentration of the degradation product, dimethylphenylsulfamide (DMSA), by a similar HPLC
method or by an alternative analytical method, provided that it demonstrates a limit of quantitation for
dichlofluanid in artificial seawater of 7 µg/l or less. The release rate of the biocide under the specified
laboratory conditions is then calculated as dichlofluanid.

4 Supplementary information
The items of supplementary information required to be able to use the general extraction procedure, described
in ISO 15181-1, for dichlofluanid and tolylfluanid are given in Annex A.
5 Apparatus
5.1 High-performance liquid chromatograph (HPLC), or other suitable instrument, which has a limit of
quantitation for tolylfluanid, dichlofluanid, or both (as appropriate) in artificial seawater of 7 µg/l or less. The
limit of quantitation shall be determined by the general procedure given in Annex 2 of ASTM D 6442-06
(Determination of the LOQ for Copper in Substitute Ocean Water for the Analytical Method), suitably modified
for tolylfluanid, dichlofluanid, or both (as appropriate). If HPLC is used, the system shall, where possible,
include the components specified in 5.1.1 to 5.1.6.
5.1.1 Gradient-programmed pump, capable of achieving a pressure of 150 bar and a flow-rate of
0,5 ml/min.
5.1.2 Ultraviolet detector, capable of monitoring at 195 nm.
5.1.3 Autosampler, capable of making 200 µl injections.
5.1.4 Chromatography column: a reverse-phase column with an internal diameter of 2,0 mm and a length
of 150 mm, packed with a microparticulate octadecylsilane (C-18, end-capped) stationary phase (mean
particle size 5,0 µm) or equivalent, and equipped with a reverse-phase pre-column with an internal diameter of
4,6 mm and a length of 30 mm, packed with a microparticulate octadecylsilane (C-18, end-capped) stationary
phase (mean particle size 5,0 µm) or equivalent.
5.1.5 Column oven, providing a constant column temperature of 40 °C.
5.1.6 Electronic data-processing system, capable of controlling the HPLC system, acquiring data and
enabling automated integration of peak areas.
5.2 Glass vials, volume 30 ml, screw-topped.
5.3 Dispensers, automatic or repeating, for reagents.
5.4 Pipettes, with disposable tips.

5.5 Volumetric flasks, volume 25 ml and 100 ml.

6 Reagents and materials
Suppliers’ material safety data sheets should be consulted for details of any hazards associated with the
reagents listed below, and the risks associated with their use should be assessed. Appropriate protective
clothing and equipment should be utilized.
Unless otherwise specified, use only reagents of recognized analytical grade.
6.1 Cleaning reagents.
Use one of the following reagents for cleaning all the equipment.
6.1.1 Hydrochloric acid, concentrated aqueous solution, 37 % by mass.
6.1.2 Hydrochloric acid, aqueous solution, 10 % by volume.
6.2 Tolylfluanid, analytical standard with a certified mass fraction of tolylfluanid of at least 99,0 %, for use
in determining the tolylfuanid release rate. See also Clause 9.
6.3 Dichlofluanid, analytical standard with a certified mass fraction of at least 99,0 %, for use in
determining the dichlofluanid release rate. See also Clause 9.
6.4 Dimethyltolylsulfamide (DMST), analytical standard with a certified mass fraction of at least 99,0 %,
for use in preparing the calibration standard for determination of the tolylfluanid release rate. See also
Clause 8.
6.5 Dimethylphenylsulfamide (DMSA), analytical standard with a certified mass fraction of at least 99,0 %
for use in preparing the calibration standard for determination of the dichlofluanid release rate. See also
Clause 8.
6.6 Artificial seawater, as defined in ISO 15181-1.
6.7 Water, conforming to the requirements of grade 2 of ISO 3696.
6.8 Acetonitrile, HPLC grade.
6.9 Formic acid, minimum 98 % by mass.
NOTE Formic acid may be substituted by acetic acid.
6.10 Sodium hydroxide, aqueous solution 0,1 mol/l.
7 Test samples
Use extracts taken from the release rate measuring containers as described in ISO 15181-1.
8 Preparation of calibration standards
8.1 General
Stock solutions of certified reference standards shall be prepared at approximately 1 000 mg/l and 100 mg/l in
acetonitrile, as described in 8.2 and 8.3. These stock solutions shall then be used to prepare calibration
standards by dilution with artificial seawater acidified to pH 2 to 3 with concentrated hydrochloric acid. A
minimum of 5 calibration standards shall be prepared at concentrations appropriate to the samples being
analysed and to define the working range for the determination of DMST and DMSA.
Where tolylfluanid and dichlofluanid release rates are being concurrently determined, both DMST and DMSA
calibration standards shall be prepared.
8.2 Stock solution A
Weigh, to the nearest 0,1 mg, about 100 mg (M) of DMST or DMSA into a 100 ml (V ) volumetric flask, add
50 ml of acetonitrile, and mix to dissolve. Make up to the mark with acetonitrile and mix well to give a
homogenous solution (dilution factor, f = 1).

Pipette 10 ml of stock solution A into a 100 ml volumetric flask, make up to the mark with acetonitrile and mix

8.4 Preparation of calibration standards from stock solutions
Select a minimum of 5 suitable target concentrations for calibration standards, appropriate to the expected
concentrations of analyte in the test samples and in order to define the working range of the method.
Calculate the volume of stock solution A or stock solution B required to achieve each target concentration by
dilution to 100 ml.
EXAMPLE 1 A calibration standard of nominal concentration 10 µg/l may be prepared by dilution of 10 µl of stock
solution B to 100 ml.
EXAMPLE 2 A calibration standard of nominal concentration 300 µg/l may be prepared by dilution of 30 µl of stock
solution A to 100 ml.
For each calibration standard, to a 100 ml (V ) volumetric flask add about 97 ml of seawater acidified to pH 2
to 3 with concentrated hydrochloric acid. Using a microlitre syringe, add the required volume (V ) of stock

solution A or stock solution B by injection below the surface of the acidified seawater, and immediately mix
well. Fill to the mark with artificial seawater acidified to pH 2 to 3 with concentrated hydrochloric acid and mix

9 Recovery and conversion check standards
9.1 General
Stock solutions of certified reference standards shall be prepared at approximately 1 000 mg/l and 100 mg/l in
acetonitrile, as described in 9.2 and 9.3. These stock solutions shall then be diluted with artificial seawater to
prepare conversion check standards that shall then be treated in accordance with 10.2. The conversion check
standards shall have approximate concentrations of 20 µg/l, 50 µg/l, and 200 µg/l dichlofluanid or tolylfluanid.
Additional conversion check standards may be prepared at appropriate concentrations to cover the working
range for the determination of DMST and DMSA.
9.2 Stock solution C
Weigh to the nearest 0,1 mg about 100 mg (M) of tolylfluanid or dichlofluanid into a 100 ml (V ) volumetric
flask, add 50 ml of acetonitrile, and mix to dissolve. Fill to the mark with acetonitrile and mix well to give a
homogenous solution (dilution factor, f = 1).

Introduction
Pour des conditions normales de température, de salinité et de pH et pour de faibles concentrations en
biocides contenues dans un milieu d'eau de mer synthétique, une valeur concordante du taux de lixiviation
dans les conditions de laboratoire spécifiées peut être déterminée au moyen de la méthode indiquée dans la
présente partie de l'ISO 15181, laquelle peut être utilisée pour l'assurance de la qualité et le choix des
matériaux. Le taux réel de lixiviation dans l'environnement des biocides contenus dans des peintures
antisalissures appliquées sur les coques de navires dépendra toutefois de nombreux facteurs, tels que les
conditions opérationnelles de service des navires, la durée de service, les conditions de mouillage, l'état de la
peinture, ainsi que la température, la salinité, le pH, les polluants et la communauté biologique dans une zone
particulière.
Les résultats de cet essai ne reflètent pas les taux de lixiviation de biocides de produits antisalissures dans
l'environnement. Les résultats de la présente méthode d'essai ne s'appliquent pas à une utilisation directe
dans le cadre des procédures d'évaluation des risques liés à l'environnement, d'une estimation de la
contrainte environnementale ou de l'établissement de limites de taux de lixiviation à des fins réglementaires.
Si l'on compare les taux de lixiviation du cuivre et des composés organostanniques obtenus par des mesures
directes ou indirectes du taux de lixiviation du cuivre des coques de navire ou par des mesures effectuées sur
des panneaux exposés dans des ports, toutes les données disponibles indiquent que les résultats de la
présente méthode générique surestiment de manière significative le taux de lixiviation de biocides dans les
conditions de service. Les résultats publiés démontrent que les résultats de la présente méthode donnent
généralement des valeurs supérieures d'un facteur d'environ 10, ou plus, aux mesures directes effectuées sur
site pour les taux de lixiviation du cuivre et des composés organostanniques des coques de navire en service,
[1, 2]
pour plusieurs revêtements antisalissures du commerce . Un rapport similaire est escompté pour d'autres
biocides. Des estimations réalistes des taux de lixiviation de biocides d'une coque de navire dans des
conditions de service ne peuvent être obtenues avec la présente méthode d'essai que lorsque cette différence
est prise en compte.
Lorsque les résultats de la présente méthode d'essai sont utilisés dans le cadre des procédures d'évaluation
des risques liés à l'environnement, d'une estimation de la contrainte environnementale ou à des fins
réglementaires, il est vivement recommandé de tenir compte du rapport existant entre les taux de lixiviation de
laboratoire et les quantités réelles relarguées dans l'environnement, pour obtenir une approximation plus
précise du taux de lixiviation de biocides des revêtements antisalissures, dans des conditions réelles. Ceci

peut être réalisé en appliquant les facteurs de correction appropriés .

1 Domaine d'application
La présente partie de l'ISO 15181 spécifie l'appareillage et la méthode d'analyse permettant de déterminer,
conformément au mode opératoire de l'ISO 15181-1, la quantité de tolylfluanide et de dichlofluanide libérée
par une peinture antisalissures dans une eau de mer synthétique.
Le tolylfluanide et le dichlofluanide sont instables en milieu marin; ils se dégradent pour former,
respectivement, du diméthyl-tolylsulfamide (DMST) et du diméthyl-phénylsulfamide (DMSA). La présente
partie de l'ISO 15181 spécifie une méthode permettant de convertir les espèces relarguées en ces produits de
dégradation et de quantifier leur concentration dans des échantillons d'eau de mer synthétique traités. Elle
donne le mode de calcul final du taux de lixiviation du tolylfluanide et du dichlofluanide dans des conditions
spécifiées de laboratoire.
La présente partie de l'ISO 15181 est conçue pour permettre la détermination simultanée du tolylfluanide, du
dichlofluanide et d'autres biocides pouvant être relargués par une peinture antisalissures donnée (le cuivre,
par exemple), par l'analyse de sous-échantillons séparés d'un extrait d'eau de mer synthétique confectionné
conformément à l'ISO 15181-1.
Conjointement à l'ISO 15181-1, les limites pratiques de quantification des taux de lixiviation à l'aide de la
−2 −1 −2 −1
présente méthode sont comprises entre 1,3 µg⋅cm ⋅d et 500 µg⋅cm ⋅d . La quantification de taux de
lixiviation inférieurs à cette plage nécessite l'emploi d'une méthode d'analyse dont la limite de quantification
du tolylfluanide, du dichlofluanide ou des deux (selon le cas) dans l'eau de mer synthétique, est inférieure à
celle indiquée dans l'Article 3 et en 5.1.

3 Principe
La quantité de tolylfluanide relargué par un cylindre d'essai dans de l'eau de mer synthétique selon la
méthode décrite dans l'ISO 15181-1 est déterminée en favorisant la dégradation du tolylfluanide dans le
lixiviat dans des conditions contrôlées, puis en quantifiant la concentration du produit de dégradation, en
l'occurrence le diméthyl-tolylsulfamide (DMST), par chromatographie liquide haute performance (CLHP) ou
par toute autre méthode analytique présentant une limite de quantification du tolylfluanide dans l'eau de mer
synthétique, inférieure ou égale à 7 µg/l. Le taux de lixiviation du biocide dans les conditions spécifiées de
laboratoire est, ensuite, exprimé sous forme de tolylfluanide.
La quantité de dichlofluanide relargué par un cylindre d'essai dans de l'eau de mer synthétique selon la
méthode indiquée dans l'ISO 15181-1 est déterminée en favorisant la dégradation contrôlée du dichlofluanide
dans le lixiviat, puis en mesurant la concentration du produit de dégradation, en l'occurrence le diméthyl-
phénylsulfamide (DMSA), par une méthode de CLHP similaire ou par toute autre méthode analytique, à
condition que celle-ci présente une limite de quantification du dichlofluanide dans l'eau de mer synthétique,
inférieure ou égale à 7 µg/l. Le taux de lixiviation du biocide dans les conditions spécifiées de laboratoire est,
ensuite, exprimé sous forme de dichlofluanide.
Des informations supplémentaires relatives au tolylfluanide, au dichlofluanide, au DMST et au DMSA sont
données dans l'Annexe B.
4 Informations supplémentaires
Les informations supplémentaires exigées pour permettre l'application de la méthode générale d'extraction
décrite dans l'ISO 15181-1, pour le dichlofluanide et le tolylfluanide, sont données dans l'Annexe A.
5 Appareillage
5.1 Chromatographe liquide haute performance (CLHP), ou tout autre instrument approprié,
présentant une limite de quantification du tolylfluanide, du dichlofluanide ou des deux (selon le cas) inférieure
ou égale à 7 µg/l dans de l'eau de mer synthétique. La limite de quantification doit être déterminée selon le
mode opératoire général donné dans l'Annexe 2 de l'ASTM D 6442-06 (Determination of the LOQ for Copper
in Substitute Ocean Water for the Analytical Method), en y apportant les adaptations nécessaires pour le
tolylfluanide, le dichlofluanide ou les deux (selon le cas). En cas d'utilisation de la méthode CLHP, le système
doit, dans la mesure du possible, comprendre les éléments spécifiés de 5.1.1 à 5.1.6.
5.1.1 Pompe à gradient programmée, pouvant atteindre une pression de 150 bar et un débit de
0,5 ml/min.
5.1.2 Détecteur ultraviolet, réglable à une longueur d'onde de 195 nm.
5.1.3 Échantillonneur automatique, capable d'effectuer des injections de 200 µl.
5.1.4 Colonne de chromatographie, colonne à phase inversée ayant un diamètre intérieur de 2,0 mm et
une longueur de 150 mm, remplie d'une phase stationnaire à base de microparticules d'octadécylsilane (taille
moyenne des particules: 5,0 µm) (C-18, avec masquage des hydroxyles résiduels) ou d'une substance
équivalente. La colonne est munie d'une précolonne à phase inversée, ayant un diamètre intérieur de 4,6 mm
et une longueur de 30 mm, remplie d'une phase stationnaire à base de microparticules d'octadécylsilane
(taille moyenne des particules: 5,0 µm) (C-18, avec masquage des hydroxyles résiduels) ou d'une substance
équivalente.
5.1.5 Four intégré à la colonne, permettant d'obtenir une température de colonne constante de 40 °C.
5.1.6 Système électronique d'acquisition de données, capable de gérer le système CLHP, de saisir des
données et de permettre l'intégration automatique des surfaces de pics.

5.2 Flacons en verre, d'un volume de 30 ml, munis de bouchons à vis.
5.3 Distributeurs, automatiques ou à répétition, pour les réactifs.
5.4 Pipettes, munies d'embouts jetables.
5.5 Fioles jaugées, d'un volume de 25 ml et 100 ml.
5.6 Seringue à fractionnement en microlitres, permettant de distribuer avec exactitude des volumes
compris entre 1 µl et 100 µl.
5.7 Seringue, en verre ou jetable, permettant de distribuer 2 ml.
6 Réactifs et matériaux
Il convient de consulter les fiches techniques de sécurité des fournisseurs pour obtenir des détails afférents
aux dangers associés aux réactifs répertoriés ci-dessous et d'évaluer les risques associés à leur utilisation. Il
est recommandé d'utiliser des vêtements de protection adaptés et des équipements de protection appropriés.
Sauf spécification contraire, n'utiliser que des réactifs de qualité analytique reconnue.
6.1 Réactifs de nettoyage.
Utiliser l'un des réactifs suivants pour nettoyer tous les équipements.
6.1.1 Acide chlorhydrique, solution aqueuse concentrée, 37 % en masse.
6.1.2 Acide chlorhydrique, solution aqueuse, 10 % en volume.
6.2 Tolylfluanide, étalon de qualité analytique, de fraction massique certifiée en tolylfluanide d'au moins
99,0 %, destiné à la détermination du taux de lixiviation du tolylfluanide. Voir également l'Article 9.
6.3 Dichlofluanide, étalon de qualité analytique, de fraction massique certifiée d'au moins 99,0 %, destiné
à la détermination du taux de lixiviation du dichlofluanide. Voir également l'Article 9.
6.4 Diméthyl-tolylsulfamide (DMST), étalon de qualité analytique, de fraction massique certifiée d'au
moins 99,0 %, utilisé pour préparer l'étalon pour la détermination du taux de lixiviation du tolylfluanide. Voir
également l'Article 8.
6.5 Diméthyl-phénylsulfamide (DMSA), étalon de qualité analytique, avec une fraction massique certifiée
d'au moins 99,0 %, utilisé pour préparer l'étalon pour la détermination du taux de lixiviation du dichlofluanide.

6.6 Eau de mer synthétique, telle que spécifiée dans l'ISO 15181-1.
6.7 Eau, conforme aux exigences relatives à l'eau de qualité 2, spécifiées dans l'ISO 3696.
6.8 Acétonitrile, de qualité CLHP.
6.9 Acide formique, au moins 98 % en masse.
NOTE Il est possible de remplacer l'acide formique par de l'acide acétique.
6.10 Hydroxyde de sodium, solution aqueuse à 0,1 mol/l.
7 Échantillons pour essai
Extraits prélevés dans les récipients de mesure du taux de lixiviation spécifiés dans l'ISO 15181-1.
8 Préparation des étalons
8.1 Généralités
Les solutions mères d'étalons de référence certifiés doivent être préparées à environ 1 000 mg/l et 100 mg/l
dans de l'acétonitrile, comme spécifié en 8.2 et 8.3. Ces solutions mères doivent ensuite être utilisées pour la
préparation des étalons, par dilution dans de l'eau de mer synthétique acidifiée (pH 2 à pH 3) à l'aide d'acide
chlorhydrique concentré. Pour analyser les échantillons et définir le domaine de travail pour la détermination
du DMST et du DMSA, préparer au moins cinq étalons aux concentrations appropriées.
Lorsque les taux de lixiviation du tolylfluanide et du dichlofluanide sont déterminés simultanément, il faut
préparer les étalons de DMST et de DMSA.
8.2 Solution mère A
Peser (à 0,1 mg près) environ 100 mg (M) de DMST ou de DMSA dans une fiole jaugée de 100 ml (V ),
ajouter 50 ml d'acétonitrile et homogénéiser pour dissoudre le tout. Compléter au trait avec de l'acétonitrile et
bien mélanger pour obtenir une solution homogène (facteur de dilution f = 1).

8.3 Solution mère B
Au moyen d'une pipette, introduire 10 ml de solution mère A dans une fiole jaugée de 100 ml, compléter au
trait avec de l'acétonitrile et bien mélanger pour obtenir une solution homogène (facteur de dilution f = 0,1).

8.4 Préparation des étalons à partir des solutions mères
Choisir au moins cinq concentrations cibles convenables pour les étalons, correspondant aux concentrations
attendues de l'analyte dans les échantillons pour essai et à partir desquels le domaine de travail de la
méthode pourra être défini. Calculer le volume de la solution mère A ou B nécessaire pour atteindre chaque
concentration cible en diluant à 100 ml.
EXEMPLE 1 Il est possible de préparer un étalon ayant une concentration nominale de 10 µg/l en diluant 10 µl de la
solution mère B dans 100 ml.
EXEMPLE 2 Il est possible de préparer un étalon ayant une concentration nominale de 300 µg/l en diluant 30 µl de la
solution mère A dans 100 ml.
Pour chaque étalon, ajouter, dans une fiole jaugée de 100 ml (V ), environ 97 ml d'eau de mer acidifiée (pH 2
à 3) avec de l'acide chlorhydrique concentré. À l'aide d'une seringue à fractionnement en microlitres, ajouter le
volume (V) requis de solution mère A ou B par injection sous la surface de l'eau de mer acidifiée, et

immédiatement mélanger avec soin. Compléter au trait avec de l'eau de mer synthétique acidifiée à pH 2 ou 3
avec de l'acide chlorhydrique concentré et bien mélanger pour obtenir des solutions d'étalons homogènes.
Calculer les concentrations réelles en étalon, C , en µg/l, dans chaque étalon de référence à partir de la

9 Étalons de contrôle des phases de récupération et de conversion
9.1 Généralités
