Введение
Содержание поврежденного крахмала является важным показателем качества пшеничной муки,
поскольку непосредственно воздействует на водопоглотительную способность муки и, следовательно,
на ее использование в пищевых продуктах сельского хозяйства.
В прошлом был разработан ряд методов на основе различных принципов для оценки такого
содержания, но сравнение результатов затруднительно из-за различия используемых принципов и
единиц измерения.
Лабораторный прибор предназначен для определения содержания поврежденного крахмала с
использованием амперометрического метода и предлагает выбор единиц измерения в соответствии с
конкретными ссылками.

1 Область применения
Настоящий международный стандарт устанавливает определение количества поврежденного
крахмала, используя амперометрический метод.
Данный метод применим ко всем пробам муки, полученной во время промышленного или
лабораторного помола пшеницы (Triticum aestivum L.).
ПРИМЕЧАНИЕ 1 Пшеница может быть размолота в лаборатории в соответствии с методами, описанными в
ISO 27971[9] или руководящем документе BIPEA BY.102.D.9302.[10]

3 Принцип

4 Реактивы
Используют только реактивы признанного аналитического качества, если не оговорено иначе.
4.1 Вода, осмозированная или деминерализованная или, по меньшей мере, эквивалентного качества.
4.2 Борная кислота или лимонная кислота, порошкообразная, для испытания.
ПРЕДОСТЕРЕЖЕНИЕ — Использование борной кислоты включает опасные операции. В этом
документе не ставится цель решить все проблемы безопасности, связанные с еѐ применением.
Пользователь сам несет ответственность за установление соответствующих мер по технике
безопасности и охране здоровья, а также обеспечение соответствия любым национальным
регламентирующим условиям.
4.3 Йодид калия, порошкообразный, для испытания.
4.4 Тиосульфат натрия, водный раствор концентрацией 0,1 моль/л, приготовленный из готовой к
использованию ампулы, содержащей 0,1 моль тиосульфата натрия, путем разбавления водой (4.1) в
колбе вместимостью 1 л. Также может использоваться порошкообразный тиосульфат натрия, если
концентрация конечного раствора составляет 0,1 моль/л. Защищают раствор от света и используют в
течение 3 месяцев.
5 Оборудование
Используют обычную лабораторную аппаратуру и, в частности, следующую.

6 Отбор проб
Отбор проб не включен в метод, установленный в этом международном стандарте. Рекомендуемый
[8]
метод отбора проб приводится в ISO 24333.
Важно, чтобы лаборатория получила действительно представительную пробу, которая не была
подвергнута порче или изменению во время транспортировки или хранения.
7 Методика
7.1 Взвешивание и растворение реактивов
Взвешивают (5.2), с точностью до 0,5 г, 3,0 г борной кислоты (4.2) или 1,5 г лимонной кислоты (4.2) и
3,0 г (4.3) йодида калия и переносят в чистый и сухой реакционный сосуд (5.1). Добавляют одну каплю
(приблизительно 0,04 мл) раствора тиосульфата натрия (4.4) и отмеряют (5.4) 120 мл
дистиллированной воды (4.1) в реакционный сосуд.
Так как испытание начинается со стадии нагрева и перемешивания, то нет необходимости добиваться
полного растворения реактивов на этой стадии. Для минимизации потерь при переносе добавляют
порошкообразные реактивы непосредственно в реакционный сосуд.
7.2 Взвешивание пробы

Взвешивают (5.3), с точностью до 10 г, 1,000 г ± 0,100 г пробы муки для испытания и помещают ее в
предварительно очищенный держатель пробы (5.1).
7.3 Испытание
Помещают реакционный сосуд в углубление на анализаторе.
Опускают верхнюю часть анализатора и вставляют держатель с пробой муки (7.2) в рабочую камеру.
Начинают испытание. Указывают точную массу пробы для анализа пшеничной муки, взвешенной с
точностью до 0,001 г. Также можно указать содержание влаги и белка в пробе, если необходимо
получить скорректированный на этой основе результат, в противном случае следует оставить
стандартные значения (массовые доли 14 % и 12 % соответственно) для этих двух компонентов.
Подтверждают начало испытания.
Продолжают испытание в течение от 6 мин до 7 мин. В то время как запускают вибратор, необходимо
удостовериться в том, что вся мука опустилась в реакционный сосуд. Используют кончик кисточки или
легкое постукивание, чтобы дать возможность осесть оставшейся на стенках сосуда муке.
Ожидают звуковой сигнал об окончании испытания, на что указывает отображение результата на дисплее.
7.4 Очистка
Поднимают верхнюю часть анализатора и вынимают реакционный сосуд. Ополаскивают, затем
осторожно и тщательно протирают измерительный электрод, нагревательный элемент и
перемешивающее устройство.

Удаляют любой осадок из реакционного сосуда (не выбрасывают в раковину). Тщательно промывают и
вытирают насухо реакционный сосуд, в котором не должно быть никаких следов реактивов, пробы или
влаги и который должен быть готовым для использования в последующем испытании.
7.5 Количество определений
Выполняют два определения на одной и той же пробе для испытания.
8 Выражение результатов
Результат выражают в виде A % (процент поглощения йода), преобразуя в единицы UCD (единицы

Chopin–Dubois). Для расчета эквивалентного значения в других единицах можно использовать
формулы, установленные изготовителем.
За результат следует брать среднеарифметическое двух определений (7.5) если они соответствуют
условиям повторяемости, указанным в 9.2 или Таблице A.5. В противном случае, выполняют два новых
определения.
ПРИМЕЧАНИЕ Может быть полезным проводить расчет количества поврежденного крахмала на основе
постоянного содержания влаги и белка. В этом случае можно определять содержание влаги и белка в пшеничной

муке в соответствии с требованиями ISO 712 относительно содержания влаги и ISO 20483 или

ISO/TS 16634-2 относительно содержания белка.
9 Прецизионность
9.1 Межлабораторные испытания
В ходе проведения двух межлабораторных испытаний были установлены пределы повторяемости и
воспроизводимости данного метода. Статистические результаты исследования приведены в
Приложении A.
Значения, полученные в результате каждого из этих испытаний, применимы к диапазонам
концентрации и сортам муки из пшеницы (Triticum aestivum L.).

Introduction
Damaged starch content is an important parameter in flour quality as it directly impacts the flour water
absorption capacity and therefore its use in the agri-food industry.
In the past, a number of methods based on various principles were developed to estimate such content,
but comparing the results is difficult due to the different principles and units of measurement used.
A laboratory device is dedicated to the determination of damaged starch content using an amperometric
method and which offers a choice of units of measurement according to individual references.

1 Scope
This International Standard specifies the determination of the damage to starch using an
amperometric method.
It is applicable to all flour samples from industrial or laboratory milling of wheat (Triticum aestivum L.).

NOTE 2 In the absence of validity studies, the results on semi-wholemeal or wholemeal flour, although able to
meet the conditions of repeatability given in Clause 9, require careful interpretation.
2 Terms and definitions
For the purposes of this document, the following terms and definitions apply.
2.1
damaged starch
starch granules present in wheat flour mechanically damaged during milling, leading to a greater
capacity to absorb water and increasing susceptibility to amylolytic enzymes
Note 1 to entry: Too high a damaged starch content has a negative effect on quality of flours.
3 Principle
Determination of damaged starch content of a flour sample by measurement of iodine absorption
kinetics in an aqueous medium using an amperometric electrode.
The amperometric method is based on the existing proportionality between iodine absorption capacity
and starch damage content.
4 Reagents
Use only reagents of recognized analytical grade, unless otherwise specified.
4.1 Water, osmosed or demineralized or at least equivalent grade.
4.2 Boric acid or citric acid, powdered, for testing.
WARNING — The use of boric acid involves hazardous operations. This document does not
purport to address all the safety problems, if any, associated with its use. It is the responsibility
of the user to establish appropriate safety and health practices and to ensure complaince with
any national regulatory conditions.
4.3 Potassium iodide, powdered, for testing.
4.4 Sodium thiosulfate, solution in water at 0,1 mol/l prepared from a ready-to-use vial containing
0,1 mol sodium thiosulfate, to be diluted with water (4.1) in a 1 l flask. Powdered sodium thiosulfate can

also be used where the concentration of the final solution is 0,1 mol/l. Protect the solution from light and
use within 3 months.
5 Equipment
Usual laboratory apparatus and, in particular, the following.

6 Sampling
Sampling is not part of the method specified in this International Standard. A recommended sampling

method is given in ISO 24333.
It is important the laboratory receive a truly representative sample which has not been damaged or
changed during transport or storage.
7 Procedure
7.1 Reagent weighing and dissolution
Weigh (5.2), to the nearest 0,5 g, 3,0 g boric acid (4.2) or 1,5 g citric acid (4.2) and 3,0 g (4.3) potassium
iodide and add to a clean and dry reaction vessel (5.1). Add one drop (about 0,04 ml) of sodium thiosulfate
solution (4.4) and dispense (5.4) 120 ml distilled water (4.1) into the vessel.
As the test begins with a heating and stirring phase, it is not necessary to obtain full dissolution of the
reagents at this stage. In order to minimize losses during transfer, add powdered reagents directly to
the reaction vessel.
7.2 Sample weighing

Weigh (5.3), to the nearest 10 g, 1,000 g ± 0,100 g of the test sample of flour and place it in the pre-
cleaned sample holder (5.1).
7.3 Test
Place the reaction vessel in the well on the device.
Lower the head of the device, and insert the sample holder containing the flour (7.2) into the compartment.

Start the test. Indicate the exact mass of the test portion of flour weighed to the nearest 0,001 g. It is also
possible to indicate the water and protein content of the sample if a result corrected on this basis is to
be obtained, otherwise the default values should be left (mass fractions of 14 % and 12 %, respectively)
for the two components. Confirm the start of the test.
The test lasts 6 min to 7 min. Ensure that all of the flour descends into the reaction vessel when the
vibrator starts up. Use the tip of a brush or lightly blow to enable any remaining flour to fall.
Wait for the beep at the end of the test at which point the result is displayed.
7.4 Cleaning
Raise the head of the device and remove the reaction vessel. Rinse, then carefully and gently wipe the
measuring electrode, the heating element and the stirrer.
Remove any residue from the vessel (do not dispose of in the sink). Carefully wash and wipe dry the
reaction vessel, which shall be free of all traces of reagent, sample or moisture, and which shall be left
ready for use in a later test.
7.5 Number of determinations
Perform two determinations on the same test sample.
8 Expression of results
The result is expressed as A % (iodine absorption percentage) converted into UCD (Chopin–Dubois
I
units). Equations provided by the manufacturer can be used to calculate the equivalence in another unit.
The arithmetic mean of the two determinations (7.5) shall be taken as the result if they meet the
conditions of repeatability specified in 9.2 or in Table A.5. Otherwise, perform two new determinations.

Avant-propos
L’ISO (Organisation internationale de normalisation) est une fédération mondiale d’organismes
nationaux de normalisation (comités membres de l’ISO). L’élaboration des Normes internationales est
en général confiée aux comités techniques de l’ISO. Chaque comité membre intéressé par une étude
a le droit de faire partie du comité technique créé à cet effet. Les organisations internationales,
gouvernementales et non gouvernementales, en liaison avec l’ISO participent également aux travaux.
L’ISO collabore étroitement avec la Commission électrotechnique internationale (CEI) en ce qui concerne
la normalisation électrotechnique.
Les Normes internationales sont rédigées conformément aux règles données dans les Directives
ISO/CEI, Partie 2.

Introduction
Le taux d’endommagement de l’amidon est un paramètre important de la qualité d’une farine car il
conditionne le taux d’hydratation de celle-ci et donc son utilisation dans l’industrie agroalimentaire.
Historiquement, de nombreuses méthodes reposant sur des principes différents ont été mises au point
pour estimer ce taux, la comparaison des résultats s’avérant difficile pour des raisons de principe de
mesure et d’unité d’expression.
Un appareil de laboratoire est dédié à la détermination du taux d’endommagement de l’amidon au
moyen d’une méthode ampérométrique. Il propose plusieurs unités de mesure en fonction de références
individuelles.

Farine de blé tendre (Triticum aestivum L.) — Méthode
ampérométrique pour le mesurage de l’endommagement
de l’amidon
1 Domaine d’application
La présente Norme internationale spécifie la détermination de l’endommagement de l’amidon au moyen
d’une méthode ampérométrique.
Elle est applicable à tous les échantillons de farine de blé tendre (Triticum aestivum L.) issus d’une
mouture industrielle ou de laboratoire.

NOTE 2 En l’absence d’études de validité, les résultats obtenus sur des farines semi-complètes ou complètes, bien
que pouvant satisfaire aux conditions de répétabilité données à l’Article 9, nécessitent d’être interprétés avec prudence.
2 Termes et définitions
Pour les besoins du présent document, les termes et définitions suivants s’appliquent.
2.1
endommagement de l’amidon
granules d’amidon présents dans la farine de blé endommagés mécaniquement lors de la mouture, ce qui
accroît la capacité d’absorption d’eau de l’amidon et sa sensibilité aux enzymes amylolytiques

3 Principe
Détermination du taux d’endommagement de l’amidon d’un échantillon de farine par mesure de la
cinétique d’absorption d’iode en milieu aqueux au moyen d’une électrode ampérométrique.
La méthode ampérométrique repose sur la proportionnalité qui existe entre la capacité d’absorption de
l’iode et le taux d’endommagement de l’amidon.
4 Réactifs
Sauf spécification contraire, utiliser uniquement des réactifs de qualité analytique reconnue.
4.1 Eau, osmosée ou déminéralisée ou de qualité au moins équivalente.
4.2 Acide borique ou acide citrique, en poudre, pour analyses.
AVERTISSEMENT — L’utilisation d’acide borique implique des opérations à caractère dangereux.
Le présent document n’a pas pour but d’aborder tous les problèmes de sécurité liés, le cas
échéant, à son utilisation. Il incombe à l’utilisateur d’établir des pratiques d’hygiène et de sécurité
appropriées et de s’assurer de la conformité à la réglementation nationale en vigueur.
4.3 Iodure de potassium, en poudre, pour analyses.

4.4 Thiosulfate de sodium, solution dans l’eau à 0,1 mol/l préparée à partir d’une ampoule prête
à l’emploi contenant 0,1 mol de thiosulfate de sodium, à diluer avec de l’eau (4.1) dans une fiole de 1 l.
L’utilisation de thiosulfate de sodium en poudre est aussi possible à partir du moment où la concentration
de la solution finale est de 0,1 mol/l. Protéger la solution de la lumière et utiliser dans les 3 mois.
5 Matériel
Matériel courant de laboratoire et, en particulier, ce qui suit.

NOTE La présente Norme internationale a été élaborée au moyen du SDmatic Chopin. Elle n’est pas
applicable aux appareils SD4 Chopin et Rapid FT Chopin, qui mesurent également le taux d’endommagement de
l’amidon, mais avec une autre technologie.

6 Échantillonnage
L’échantillonnage ne fait pas partie de la méthode spécifiée dans la présente Norme internationale. Une

méthode d’échantillonnage recommandée est décrite dans l’ISO 24333 .
Il est important que le laboratoire reçoive un échantillon qui soit réellement représentatif et qui n’ait pas
été endommagé ou modifié pendant le transport ou le stockage.
7 Mode opératoire
7.1 Pesée des réactifs et mise en solution
Peser (5.2) à 0,5 g près, 3,0 g d’acide borique (4.2) ou 1,5 g d’acide citrique (4.2) et 3,0 g d’iodure de potassium
(4.3) et les introduire dans un vase à réaction (5.1) propre et sec. Ajouter une goutte (environ 0,04 ml) de
solution de thiosulfate de sodium (5.4) et distribuer (5.4) 120 ml d’eau distillée (4.1) dans le vase.
L’essai commençant par une phase de chauffage et d’agitation, il n’est pas nécessaire d’obtenir, à ce stade,
une dissolution complète des réactifs. Afin de limiter les pertes lors du transvasement, introduire les
réactifs en poudre directement dans le vase à réaction.
7.2 Pesée de l’échantillon

Peser (5.3) à 10 g près, 1,000 g ± 0,100 g de l’échantillon pour essai de farine et le déposer sur le porte-
échantillon (5.1) préalablement nettoyé.

7.3 Essai
Déposer le vase à réaction à l’emplacement prévu à cet effet sur l’appareil.
Abaisser la tête de l’appareil, introduire le porte-échantillon avec la farine (7.2) dans le logement
prévu à cet effet.
Démarrer l’essai. Indiquer la masse exacte de la prise d’essai de farine pesée à 0,001 g près. Il est aussi
possible d’indiquer la teneur en eau et la teneur en protéines de l’échantillon si l’on souhaite obtenir
un résultat corrigé sur cette base; sinon, il convient de laisser les valeurs par défaut (respectivement
fractions massiques de 14 % et 12 %) pour ces deux composantes. Confirmer le lancement de l’essai.
L’essai dure 6 min à 7 min. Veiller à ce que la totalité de la farine descende bien dans le vase à réaction
lorsque le vibreur se déclenche. Au besoin, aider avec la pointe d’un pinceau ou un courant d’air léger.
Attendre la fin de l’essai signalée par un bip sonore et l’affichage du résultat.
7.4 Nettoyage
Relever la tête de l’appareil et extraire le vase à réaction. Rincer, puis essuyer soigneusement et
délicatement l’électrode de mesure, la résistance de chauffage et l’hélice d’agitation.
Évacuer le résidu de réaction contenu dans le vase (ne pas rejeter à l’évier). Laver et essuyer soigneusement
le vase à réaction qui doit être exempt de toute trace de réactif, d’échantillon et d’humidité en vue d’un
prochain essai.
7.5 Nombre de déterminations
Effectuer deux déterminations sur le même échantillon pour essai.
8 Expression des résultats
Le résultat est exprimé en A % (pourcentage d’absorption d’iode) transformé en UCD (unités
I
Chopin-Dubois). Des équations fournies par le constructeur peuvent éventuellement permettre de
calculer l’équivalence dans une autre unité.
Le résultat final doit être la moyenne arithmétique des deux déterminations (7.5) si celles-ci remplissent
les conditions de répétabilité spécifiées en 9.2 ou dans le Tableau A.5. Dans le cas contraire, effectuer
deux nouvelles déterminations.
