Overview
The SIST EN ISO 23753-1:2019/A1:2021 standard, titled Soil quality - Determination of dehydrogenases activity in soils - Part 1: Method using triphenyltetrazolium chloride (TTC) - Amendment 1, is an important European and international standard published by CEN and ISO. This document specifies an amended analytical method for measuring enzymatic activity-specifically, dehydrogenase activity-in soil samples using the TTC assay.
Dehydrogenases are critical soil enzymes involved in biological oxidation processes and serve as indicators of overall microbial activity and soil health. Assessing their activity plays a fundamental role in environmental monitoring, soil quality evaluation, and ecological studies.
Key Topics
Methodology
The amended method quantifies dehydrogenase activity based on the reduction of triphenyltetrazolium chloride (TTC) to triphenyl formazan (TPF), which is measurable spectrophotometrically. The amendment updates the calculation formula for enzymatic activity, accounting for dry soil weight and other key variables.Formula for Activity Calculation
Enzymatic activity (A) is calculated as follows:[ A = \frac{(C_s - C_b) \times V \times 100}{m \times DM \times RT} ]
- C_s: concentration of TPF in sample replicate (nmol/ml)
- C_b: concentration of TPF in control (nmol/ml)
- V: reaction volume in ml (substrate + buffer + ethanol)
- m: mass of soil sample (g)
- DM: dry matter content of soil (%)
- RT: reaction time (minutes)
Standardization and Compliance
The amendment is integrated into the original EN ISO 23753-1:2019 and must be implemented by all CEN member countries to ensure harmonization of soil quality assessment methods across Europe.Technical Committee Involvement
The standard was developed by ISO/TC 190 "Soil quality" and CEN/TC 444 focused on environmental characterization of solid matrices, ensuring international consensus and scientific rigor.
Applications
Soil Quality Monitoring
Regular assessment of dehydrogenase activity provides vital data on soil microbial health, which relates directly to soil fertility and ecosystem functionality.Environmental Impact Assessment
This method is widely used to evaluate the impact of pollutants, agricultural practices, and remediation efforts on soil biology.Agricultural Research
Researchers employ this standardized enzymatic assay to investigate soil biological responses to different land management practices or inputs.Ecotoxicology Studies
Measuring soil enzyme activities enables the detection of toxic substances affecting microbial populations, assisting in risk assessments.Policy and Regulatory Compliance
Governments and environmental agencies adopt SIST EN ISO 23753-1:2019/A1:2021 as part of soil conservation and environmental protection protocols.
