This International Standard specifies a procedure which can be used to determine the genotoxicity1) of water and waste water using the umu-test. This assay is based on the detection of genotoxicity of a test sample which increases the expression of the SOSrepair system2) associated with the umuC-gene3).

Introduction
The genetically engineered bacterium Salmonella typhimurium TA1535/pSK1002 serves as a test organism.
The bacteria are exposed under controlled conditions to different concentrations of the samples to be tested. The
test is based on the capability of genotoxic agents to induce the umuC-gene in the Salmonella strain in response to
genotoxic lesions in the DNA.
Due to its capability to respond to different types of genotoxic lesions, only one single strain is necessary to detect
different kinds of genotoxic substances.
The induction of the umuC-gene is thus a measure for the genotoxic potential of the sample. Since the umuC-gene
is fused with the lacZ-gene for �-galactosidase, the induction of the umuC-gene can be easily assessed by
determination of the�-galactosidase activity.

licensing may restrict the use of these organisms.
Test conducted according to this International Standard should be carried out by qualified experts or by a
qualified testing laboratory.
When applying this International Standard it is necessary in each case, depending on the range to be
tested, to determine if and to which extent additional criteria should be established.

3 Terms and definitions
For the purposes of this International Standard, the following terms and definitions apply. Other related terms and
definitions have been included in annex A for information.

stock culture
culture of a bacterial strain to preserve the original test strain and to prepare the inoculation material for the
overnight culture or the pre-culture
1) Toxicity which specifically affects the genome (genetic material).
2) SOS repair occurs when cells are overwhelmed by genotoxins allowing the cell to survive at the cost of mutagenesis.
3) umuC-gene is the acronym for UV mutagenesis gene C. The induction of the umuC-gene is part of the specific response of
the bacterial cell to DNA-damage.

�metabolic activation system� 9 000 g centrifugation supernatant prepared from the livers of male rats pretreated
with enzyme-inducing agents
NOTE Bacteria are exposed to the test sample both with and without an appropriate metabolic activation system.
4Principle
The test organisms are exposed to the test sample with and without metabolic activation system using microplates.
After 4 h of incubation, the genotoxin-dependent induction of the umuC-gene is compared to the spontaneous
activation of the untreated, control culture.
5 Test organism and reagents
5.1 Test organism and stock culture
5.1.1 Test organism
Salmonella typhimurium is a gram-negative, facultative, anaerobic bacterium from the Enterobacteriaceae family.
Salmonella typhimurium TA1535 is the original strain. The test organism carries the plasmid pSK1002 with the
umuC-lacZ gene and a gene for ampicillin resistance. The designation of this Salmonella strain is "TA1535/
pSK1002" (see annex B). This bacterial strain can be easily selected due to its ampicillin resistance.
5.1.2 Stock culture preparation and preservation
Preserve Salmonella typhimurium TA1535/pSK1002 in 150 μl culture medium with 10 % dimethyl sulfoxide (DMSO)
or 20 % glycerol in 2 ml ampoules at a temperature not above � 80 °C. For the preparation of an overnight culture
only one ampoule is used.
5.2 Reagents
Chemicals shall be of analytical grade. Prepare all solutions with purified deionized water or water of equivalent
purity.
5.2.1 Hydrochloric acid, c(HCl) = 1mol/l.
5.2.2 Sodium hydroxide solution, c(NaOH) = 1 mol/l.
5.2.3 Dimethyl sulfoxide (DMSO),C H SO .

WARNING — DMSO forms mutagenic products over a period of time.
5.2.4 TGA-culture medium, consisting of tryptone, glucose and ampicillin, prepared as follows.
Dissolve 10 g of tryptone, 5 g of sodium chloride (NaCl) and 11,9 g of 4-(2-hydroxyethyl)-I-
piperazineethanesulphonic acid (HEPES) in water, adjust the pH-value to 7,0� 0,2, dilute to 980 ml and autoclave
for 20 min at 121 °C. Dissolve 2 g of D(+)-glucose (anhydrous) in 20 ml distilled water and autoclave separately.
After autoclaving, mix the two solutions in equal proportions and add 50 mg of ampicillin to 1 000 ml of cooled TGA
medium under sterile conditions. The solution can be stored in portions at � 20 °C for up to 4 weeks.
5.2.5 Concentrated 10�����TGA-culture medium, consisting of a tenfold concentrated TGA (5.2.4) solution, which
canbestoredfor 14days at4°C.

5.2.5.1 For incubation without S9, prepared as follows.
Dissolve 10 g of tryptone, 5 g of sodium chloride (NaCl) and 11,9 g of HEPES in 80 ml water. Adjust the pH-value
to 7,0� 0,2. Dissolve 2 g of D(+)-glucose (anhydrous) in 20 ml of water. Autoclave the solutions separately for
20 min at 121 °C, mix the solutions and add 50 mg of ampicillin to 100 ml of the mixed solution under sterile
conditions.
5.2.5.2 For incubation with S9, prepared as follows.
Dissolve 10 g of tryptone, 5 g of sodium chloride (NaCl), 2,46 g of potassium chloride (KCl), 1,63 g of magnesium
chloride hexahydrate (MgCl �6H O), and 11,9 g of HEPES in 80 ml of water. Adjust the pH to 7,0� 0,2. Dissolve

2gof D(+)-glucose (anhydrous) in 20 ml of distilled water. Autoclave both solutions separately for 20 min at 121 °C,
mix the solutions and add 50 mg of ampicillin to 100 ml mixed solution under sterile conditions.

5.2.6 B-buffer , consisting of a cell-lysis and reaction buffer, prepared as follows.
Dissolve 20,18 g of disodium hydrogenphosphate dihydrate (Na HPO �2H 0), 5,5 g of sodium

dihydrogenphosphate monohydrate (NaH PO � H O), 0,75 g of potassium chloride (KCl), 0,25 g of magnesium

sulfate heptahydrate (MgSO �7H 0) in 900 ml water. Adjust the pH to 7,0� 0,2. Then add 1,0 g of

sodiumdodecylsulfate (SDS) and dilute to 1 000 ml. Before use, add 0,27 ml of 2-mercaptoethanol to 100 ml of
B-buffer and mix.

5.2.7 Phosphate buffer pH (7,0���� 0,2) .
Dissolve 1,086 g disodium hydrogenphosphate dihydrate (Na HPO �2H 0) and 0,538 g sodium

dihydrogenphosphate monohydrate (NaH PO � H O) in 100 ml water.

If necessary adjust pH value to 7,0� 0,2. Autoclave the solution at 121 °C for 20 min.

5.2.8 Stop reagent .
Dissolve 105,99 g of sodium carbonate (Na CO ) in 900 ml water and dilute to 1 000 ml.

5.2.9 o-Nitrophenol-����-D-galactopyranoside (ONPG) solution.
Dissolve 45 mg ONPG in 10 ml phosphate buffer (5.2.7).
Due to the poor solubility of ONPG, prepare this solution in advance and stir at room temperature in the dark until
completely dissolved (approximately 2 h). Keep the solution in the dark.
5.2.10 S9 fraction, the required quantity being taken from the freezer.
Immediately after thawing, the S9 fraction is to be cooled on ice until use. Shake briefly before adding to the pre-
culture.
NOTE The S9 fraction is available commercially.
5.2.11 Cofactor solution, consisting of a freshly prepared solution, kept on ice during the test and prepared as
follows.
Dissolve 148 mg NADP (sodium salt) and 76 mg glucose-6-phosphate (disodium salt) in 5 ml 10� TGA (5.2.5).
5.2.12 Positive-control substances in dimethysulfoxide (DMSO), prepared as follows.
Warning — 2-AA can be rapidly photooxidized. Avoid prolonged exposure to light.

Dissolve 5 mg of 4-nitroquinoline-N-oxide (4-NQO) in 5 ml of DMSO. This stock solution can be stored in aliquots at
� 20 °C.
Prior to the test dilute 2 000-fold using a 30 % by volume DMSO solution (DMSO/distilled water: 3/7).
Dissolve 5 mg of aminoanthracene (2-AA) in 5 ml of DMSO. This stock solution can be stored in aliquots at
� 20 °C.
Prior to the test, dilute 500-fold using a 30 % by volume DMSO solution (DMSO/distilled water: 3/7).
6 Apparatus
The apparatus shall consist of the following:
� storage bottles: 250 ml and 500 ml;
� pipettes with rated volume: 1 ml, 10 ml, 25 ml;
� multi-channel pipettes (8 channel) having volumes: 5 μl to 50 μl, 50 μl to 200 μl, 50 μl to 300 μl;
� reservoirs for charging the multi-channel pipettes having volumes: 17 ml and 34 ml;
� measuring cylinders: 500 ml;
� culture vessels, conical flasks: 100 ml;
� sterile preservation ampoules: 2 ml (cryotubes);
� thermometer capable of measuring in the range 25 °C to 30 °C (�1°C);
� temperature- and time-controlled water bath;
� microplate-incubator for (28� 1) °C and (37� 1) °C with shaker (frequency of 125 r/min to 150 r/min);
� autoclave;
� centrifuge;
� 96-well microplates with lids and flat transparent bottoms (well capacity 380 μl);
� pH-meter;
� UV/Vis photometer (1 cm cuvettes);
� photometer for the microplates.
7 Sample preparation and preservation
Test water and waste water samples as soon as possible after sampling.
If immediate testing is not possible, store the sample at 4 °C. In this case, analyse the sample within 48 h after
collection. Otherwise, keep the sample at a temperature below� 18 °C in accordance with ISO 5667-16.

3 Terms and definitions
For the purposes of this International Standard, the following terms and definitions apply. Other related terms and
definitions have been included in annex A for information.

culture of a bacterial strain to preserve the original test strain and to prepare the inoculation material for the
overnight culture or the pre-culture
1) Toxicity which specifically affects the genome (genetic material).
2) SOS repair occurs when cells are overwhelmed by genotoxins allowing the cell to survive at the cost of mutagenesis.
3) umuC-gene is the acronym for UV mutagenesis gene C. The induction of the umuC-gene is part of the specific response of
the bacterial cell to DNA-damage.

�metabolic activation system� 9 000 g centrifugation supernatant prepared from the livers of male rats pretreated
with enzyme-inducing agents
NOTE Bacteria are exposed to the test sample both with and without an appropriate metabolic activation system.
4Principle
The test organisms are exposed to the test sample with and without metabolic activation system using microplates.
After 4 h of incubation, the genotoxin-dependent induction of the umuC-gene is compared to the spontaneous
activation of the untreated, control culture.
5 Test organism and reagents
5.1 Test organism and stock culture
5.1.1 Test organism
Salmonella typhimurium is a gram-negative, facultative, anaerobic bacterium from the Enterobacteriaceae family.
Salmonella typhimurium TA1535 is the original strain. The test organism carries the plasmid pSK1002 with the
umuC-lacZ gene and a gene for ampicillin resistance. The designation of this Salmonella strain is "TA1535/
pSK1002" (see annex B). This bacterial strain can be easily selected due to its ampicillin resistance.
5.1.2 Stock culture preparation and preservation
Preserve Salmonella typhimurium TA1535/pSK1002 in 150 μl culture medium with 10 % dimethyl sulfoxide (DMSO)
or 20 % glycerol in 2 ml ampoules at a temperature not above � 80 °C. For the preparation of an overnight culture
only one ampoule is used.
5.2 Reagents
Chemicals shall be of analytical grade. Prepare all solutions with purified deionized water or water of equivalent
purity.
5.2.1 Hydrochloric acid, c(HCl) = 1mol/l.
5.2.2 Sodium hydroxide solution, c(NaOH) = 1 mol/l.
5.2.3 Dimethyl sulfoxide (DMSO),C H SO .

WARNING — DMSO forms mutagenic products over a period of time.
5.2.4 TGA-culture medium, consisting of tryptone, glucose and ampicillin, prepared as follows.
Dissolve 10 g of tryptone, 5 g of sodium chloride (NaCl) and 11,9 g of 4-(2-hydroxyethyl)-I-
piperazineethanesulphonic acid (HEPES) in water, adjust the pH-value to 7,0� 0,2, dilute to 980 ml and autoclave
for 20 min at 121 °C. Dissolve 2 g of D(+)-glucose (anhydrous) in 20 ml distilled water and autoclave separately.
After autoclaving, mix the two solutions in equal proportions and add 50 mg of ampicillin to 1 000 ml of cooled TGA
medium under sterile conditions. The solution can be stored in portions at � 20 °C for up to 4 weeks.
5.2.5 Concentrated 10�����TGA-culture medium, consisting of a tenfold concentrated TGA (5.2.4) solution, which
canbestoredfor 14days at4°C.

5.2.5.1 For incubation without S9, prepared as follows.
Dissolve 10 g of tryptone, 5 g of sodium chloride (NaCl) and 11,9 g of HEPES in 80 ml water. Adjust the pH-value
to 7,0� 0,2. Dissolve 2 g of D(+)-glucose (anhydrous) in 20 ml of water. Autoclave the solutions separately for
20 min at 121 °C, mix the solutions and add 50 mg of ampicillin to 100 ml of the mixed solution under sterile
conditions.
5.2.5.2 For incubation with S9, prepared as follows.
Dissolve 10 g of tryptone, 5 g of sodium chloride (NaCl), 2,46 g of potassium chloride (KCl), 1,63 g of magnesium
chloride hexahydrate (MgCl �6H O), and 11,9 g of HEPES in 80 ml of water. Adjust the pH to 7,0� 0,2. Dissolve

2gof D(+)-glucose (anhydrous) in 20 ml of distilled water. Autoclave both solutions separately for 20 min at 121 °C,
mix the solutions and add 50 mg of ampicillin to 100 ml mixed solution under sterile conditions.

5.2.6 B-buffer , consisting of a cell-lysis and reaction buffer, prepared as follows.
Dissolve 20,18 g of disodium hydrogenphosphate dihydrate (Na HPO �2H 0), 5,5 g of sodium

dihydrogenphosphate monohydrate (NaH PO � H O), 0,75 g of potassium chloride (KCl), 0,25 g of magnesium

sulfate heptahydrate (MgSO �7H 0) in 900 ml water. Adjust the pH to 7,0� 0,2. Then add 1,0 g of

sodiumdodecylsulfate (SDS) and dilute to 1 000 ml. Before use, add 0,27 ml of 2-mercaptoethanol to 100 ml of
B-buffer and mix.

5.2.7 Phosphate buffer pH (7,0���� 0,2) .
Dissolve 1,086 g disodium hydrogenphosphate dihydrate (Na HPO �2H 0) and 0,538 g sodium

dihydrogenphosphate monohydrate (NaH PO � H O) in 100 ml water.

If necessary adjust pH value to 7,0� 0,2. Autoclave the solution at 121 °C for 20 min.

5.2.8 Stop reagent .
Dissolve 105,99 g of sodium carbonate (Na CO ) in 900 ml water and dilute to 1 000 ml.

5.2.9 o-Nitrophenol-����-D-galactopyranoside (ONPG) solution.
Dissolve 45 mg ONPG in 10 ml phosphate buffer (5.2.7).
Due to the poor solubility of ONPG, prepare this solution in advance and stir at room temperature in the dark until
completely dissolved (approximately 2 h). Keep the solution in the dark.
5.2.10 S9 fraction, the required quantity being taken from the freezer.
Immediately after thawing, the S9 fraction is to be cooled on ice until use. Shake briefly before adding to the pre-
culture.
NOTE The S9 fraction is available commercially.
5.2.11 Cofactor solution, consisting of a freshly prepared solution, kept on ice during the test and prepared as
follows.
Dissolve 148 mg NADP (sodium salt) and 76 mg glucose-6-phosphate (disodium salt) in 5 ml 10� TGA (5.2.5).
5.2.12 Positive-control substances in dimethysulfoxide (DMSO), prepared as follows.
Warning — 2-AA can be rapidly photooxidized. Avoid prolonged exposure to light.

Dissolve 5 mg of 4-nitroquinoline-N-oxide (4-NQO) in 5 ml of DMSO. This stock solution can be stored in aliquots at
� 20 °C.
Prior to the test dilute 2 000-fold using a 30 % by volume DMSO solution (DMSO/distilled water: 3/7).
Dissolve 5 mg of aminoanthracene (2-AA) in 5 ml of DMSO. This stock solution can be stored in aliquots at
� 20 °C.
Prior to the test, dilute 500-fold using a 30 % by volume DMSO solution (DMSO/distilled water: 3/7).
6 Apparatus
The apparatus shall consist of the following:
� storage bottles: 250 ml and 500 ml;
� pipettes with rated volume: 1 ml, 10 ml, 25 ml;
� multi-channel pipettes (8 channel) having volumes: 5 μl to 50 μl, 50 μl to 200 μl, 50 μl to 300 μl;
� reservoirs for charging the multi-channel pipettes having volumes: 17 ml and 34 ml;
� measuring cylinders: 500 ml;
� culture vessels, conical flasks: 100 ml;
� sterile preservation ampoules: 2 ml (cryotubes);
� thermometer capable of measuring in the range 25 °C to 30 °C (�1°C);
� temperature- and time-controlled water bath;
� microplate-incubator for (28� 1) °C and (37� 1) °C with shaker (frequency of 125 r/min to 150 r/min);
� autoclave;
� centrifuge;

Introduction
L’organisme d’essai est la bactérie génétiquement modifiée Salmonella typhimurium TA1535/pSK1002.
Les bactéries sont exposées, dans des conditions controlées, à différentes concentrations des échantillons à
expérimenter. L’essai est basé sur la capacité des agents génotoxiques à induire le gène umuC dans la souche
Salmonella en réponse aux liaisons génotoxiques de l’ADN.
Du fait de sa capacité à réagir à différents types de liaisons génotoxiques, une seule souche suffit à déceler
différents types de substances génotoxiques.
L’induction du gène umuC représente ainsi une mesure du potentiel génotoxique de l’échantillon. Dans la mesure
où le gène umuC s’identifie au gène lacZ de la �-galactosidase, l’induction du gène umuC peut être facilement
évaluée par détermination de l’activité de la�-galactosidase.

AVERTISSEMENT — Le présent essai implique l’utilisation d’organismes génétiquement modifiés.
L’utilisation de tels organismes peut être soumise à des autorisations nationales ou internationales.
Il convient que les essais réalisés conformément à la présente Norme internationale soient effectués par
des experts qualifiés ou par un laboratoire d’essai qualifié.
L’application de la présente Norme internationale implique, dans tous les cas, de déterminer, en fonction
du domaine de travail étudié, la nécessité de prendre en compte d’éventuels critères supplémentaires.

3 Termes et définitions
Pour les besoins de la présente Norme internationale, les termes et définitions suivants s'appliquent. D’autres

2) Le réparateur de secours se déclenche quand les cellules sont ensevelies dans les génotoxines qui permettent à la cellule

4Principe
Les organismes d’essai sont exposés, à l'aide de microplaques, à l’échantillon d’essai avec et sans système
d’activation métabolique. Après 4 h d’incubation, une comparaison est effectuée entre l’induction du gène umuC,
produite par les agents génotoxiques, et l’activation spontanée de la culture témoin, non traitée.
5 Organisme d’essai et réactifs
5.1 Organisme d’essai et culture mère
5.1.1 Organisme d’essai
Salmonella typhimurium est une bactérie anaérobie facultative, Gram négatif, de la famille des entérobactériacées.
La souche d'origine est Salmonella typhimurium TA1535. L'organisme d'essai véhicule le plasmide pSK1002 avec
le gène umuC-lacZ et un gène résistant à l'ampicilline. Cette souche de Salmonella est désignée
«TA1535/pSK1002» (voir annexe B). Du fait de sa résistance à l'ampicilline, cette souche bactérienne peut être
aisément choisie.
5.1.2 Préparation et conservation de la culture mère
Conserver Salmonella typhimurium TA1535/pSK1002 dans des ampoules de 2 ml, contenant 150 μl de milieu de
culture, avec du diméthylsulfoxyde (DMSO) à 10 % ou du glycérol à 20 %, maintenues à une température
maximale de � 80 °C. Utiliser une seule ampoule pour la préparation d'une culture de bactéries de toute une nuit.
5.2 Réactifs
Les produits chimiques doivent être de qualité analytique. Préparer toutes les solutions dans de l'eau déionisée
purifiée, ou dans de l’eau d'un degré de pureté équivalent.
5.2.1 Acide chlorhydrique, c(HCl) = 1 mol/l.
5.2.2 Solution d'hydroxyde de sodium, c(NaOH) = 1 mol/l.
5.2.3 Diméthylsulfoxyde (DMSO),C H SO .

AVERTISSEMENT — Le DMSO engendre des produits mutagènes au bout d'un certain temps.
5.2.4 Milieu de culture TGA, composé de tryptone, glucose et ampicilline, préparé comme indiqué ci- après.
Dissoudre 10 g de tryptone, 5 g de chlorure de sodium (NaCl) et 11,9 g de 4-(hydroxy-2 éthyle)-l-acide
pipérazineéthanesulfonique (HEPES) dans de l'eau, ajuster le pH à 7,0� 0,2, diluer à 980 ml et autoclaver à
121 °C pendant 20 min. Dissoudre 2 g de D(+)-glucose (anhydre) dans 20 ml d'eau distillée et autoclaver
séparément. Après l'autoclavage, mélanger les deux solutions en proportions égales puis ajouter 50 mg

d'ampicilline aux 1 000 ml de milieu TGA refroidi dans des conditions stériles. La solution peut être divisée en
portions et conservée à�20 °C pendant au maximum 4 semaines.
5.2.5 Milieu de culture TGA concentré 10����, composé de TGA concentré 10 fois, qui peut être conservé 14
jours à 4 °C.
5.2.5.1 Pour incubation sans S9, préparé comme suit.
Dissoudre 10 g de tryptone, 5 g de chlorure de sodium (NaCl) et 11,9 g de HEPES dans 80 ml d'eau. Ajuster le pH
à7,0� 0,2. Dissoudre 2 g de D(+)-glucose (anhydre) dans 20 ml d'eau. Autoclaver les solutions séparément à
121 °C pendant 20 min, mélanger ensuite les solutions puis ajouter 50 mg d'ampicilline à 100 ml du mélange dans
des conditions stériles.
5.2.5.2 Pour incubation avec S9, préparé comme suit.
Dissoudre 10 g de tryptone, 5 g de chlorure de sodium (NaCl), 2,46 g de chlorure de potassium (KCl), 1,63 g de
chlorure de magnésium hexahydraté (MgCl ,6H O) et 11,9 g de HEPES dans 80 ml d'eau. Ajuster le pH à

7,0� 0,2. Dissoudre 2 g de D(+)-glucose (anhydre) dans 20 ml d'eau distillée. Autoclaver les solutions séparément
à 121 °C pendant 20 min, mélanger ensuite les solutions puis ajouter 50 mg d'ampicilline aux 100 ml de solution
dans des conditions stériles.

5.2.8 Réactif d'arrêt .
Dissoudre 105,99 g de carbonate de sodium (Na CO ) dans 900 ml d’eau et diluer à 1 000 ml.

5.2.9 Solution de o-nitrophénol-����-D-galactopyranoside (ONPG).
Dissoudre 45 mg de ONPG dans 10 ml de tampon phosphaté (5.2.7).
En raison de la faible solubilité de l'ONPG, cette solution doit être préparée à l'avance, entièrement dissoute par
agitation à l'obscurité, et à température ambiante (environ 2 h). Cette solution doit être conservée à l’obscurité.
5.2.10 Fraction S9, la quantité nécessaire étant sortie du congélateur.
Une fois décongelée, la fraction S9 doit être conservée dans de la glace jusqu’à son utilisation. Agiter le flacon,
avant de l'ajouter à la préculture.
NOTE Des fractions S9 sont disponibles dans le commerce.

5.2.11 Solution cofactorielle, fraîchement préparée, conservée dans de la glace pendant la durée de l’essai et
réalisée comme suit.
Dissoudre 148 mg de NADP (sel de sodium) et 76 mg de glucose-6-phosphate (sel disodique) dans 5 ml de milieu
de culture TGA concentré 10� (5.2.5).
5.2.12 Témoins positifs dans le diméthylsulfoxyde (DMSO), préparés comme suit.
AVERTISSEMENT — Le 2-AA peut se photo-oxyder rapidement. Éviter toute exposition prolongée à la
lumière.
Dissoudre 5 mg de N-oxyde de 4-nitroquinoléine (4-NQO) dans 5 ml de DMSO. Cette solution mère peut être
divisée en parties aliquotes et conservée à�20 °C.
Avant l’essai, diluer 2 000 fois en utilisant une solution DMSO à 30 % en fraction volumique (DMSO/eau
déminéralisée: 3/7).
Dissoudre 5 mg d'aminoanthracène (2-AA) dans 5 ml de DMSO. Cette solution mère peut être divisée en parties
aliquotes et conservée à�20 °C.
Avant l’essai, diluer 500 fois en utilisant une solution DMSO à 30 % en fraction volumique (DMSO/eau
déminéralisée: 3/7).
6 Appareillage
L'appareillage doit comprendre les éléments suivants:
� flacons de conservation de 250 ml et 500 ml;
� pipettes de capacité de 1 ml, 10 ml et 25 ml;
� pipettes multicanaux (8 canaux) de volume de 5 μl à 50 μl, 50 μl à 200 μl, 50 μl à 300 μl;
� réservoirs de charge de pipettes multicanaux, ayant des volumes de 17 ml et 34 ml;
� éprouvettes graduées de 500 ml;
� récipients de culture, fioles coniques, de 100 ml;
� ampoules de conservation stériles de 2 ml (cryotubes);
� thermomètre gradué entre 25 °C et 30 °C (�1°C);
� bain-marie à température et durée régulables;
� microplaques-incubateur pour (28� 1) °C et (37� 1) °C avec agitateur (vitesse de 125 tr/min à 150 tr/min);
� autoclave;
� centrifugeuse;
� microplaques 96 puits à couvercles, et fonds plats transparents (capacité du puits de 380 μl);
� pH-mètre;
� photomètre UV/visible (cuves de 1 cm);
� photomètre pour les microplaques.

7 Préparation et conservation d’échantillon
Il convient que les échantillons d'eaux et d'eaux résiduaires soient soumis à l'essai le plus rapidement possible
après le prélèvement.
Si l'essai ne peut être réalisé immédiatement, l'échantillon doit être conservé à 4 °C. Dans ce cas, l'échantillon doit
être analysé dans les 48 h qui suivent le prélèvement. Si ce n’est pas le cas, l’échantillon doit être conservé à une
température de�18 °C conformément à l’ISO 5667-16.
Dans certains cas exceptionnels, il est possible de procéder à d'autres mesures préparatoires comme, par
exemple, la centrifugation et la filtration. Ceci peut conduire à l'élimination de la substance génotoxique
