ISO 22634-2:2017 specifies a method for the determination of benzo[a]pyrene (B[a]P) in the total particulate matter of cigarette mainstream smoke using gas chromatography/mass spectrometry (GC/MS) with cyclohexane as extraction solvent. This method was validated using ISO 3308 smoking parameters and is technically compatible with other smoking regimes. ISO 22634-2:2017 provides an alternative method to that specified in ISO 22634-1, with a different clean-up, which is less time consuming and with a reduced analytical run allowing a potential increase of sample throughput in comparison with ISO 22634-1.

Introduction
No machine smoking regime can represent all human smoking behaviours:
— it is recommended that cigarettes also be tested under conditions of a different intensity of machine
smoking than those specified in this document;
— machine smoking testing is useful to characterize cigarette emissions for design and regulatory
purposes, but communication of machine measurements to smokers can result in misunderstandings
about differences in exposure and risk across brand;
— smoke emission data from machine measurements may be used as inputs for product hazard
assessment, but they are not intended to be nor are they valid measures of human exposure or
risks. Communicating differences between products in machine measurements as differences in
exposure or risk is a misuse of testing using ISO standards.

1 Scope
This document specifies a method for the determination of benzo[a]pyrene (B[a]P) in the total
particulate matter of cigarette mainstream smoke using gas chromatography/mass spectrometry
(GC/MS) with cyclohexane as extraction solvent.
This method was validated using ISO 3308 smoking parameters and is technically compatible with
other smoking regimes.
This document provides an alternative method to that specified in ISO 22634-1, with a different clean-
up, which is less time consuming and with a reduced analytical run allowing a potential increase of
sample throughput in comparison with ISO 22634-1.

5 Apparatus
The usual laboratory apparatus and equipment and, in particular, the following.

5 Apparatus
The	usual	laboratory	apparatus	and	equipment	and,	in	particular,	the	following.
5.1 Routine analytical cigarette-smoking machine,	complying	with	the	requirements	of	ISO	3308
and	equipped	for	smoking	in	accordance	with	ISO	4387.
5.2 Gas chromatograph with a mass selective detector,	equipped	with	its	computerized	control
and	data	acquisition	and	processing	system.	This	system	shall	be	able	to	pilot	the	mass	spectrometer	in
order	to	obtain	chromatographic	data	under	single	ion	monitoring	(SIM)	detection	mode.	The	gas
chromatograph	shall	be	configured	to	perform	splitless	injections	on	a	capillary	column.	It	is
recommended	to	equip	the	gas	chromatograph	with	an	autosampler	for	sample	injection.
5.3 Fused silica capillary column,	for	example	a	50	%	phenyl‐,	50	%	methyl‐polysiloxane
stationary	phase	and	a	30	m	length,	0,25	mm	internal	diameter	column	with	a	0,25	μm	film	thickness
are	suitable	for	this	analysis.
NOTE	 Other	columns	can	be	used,	provided	that	appropriate	peak	separation	is	obtained.

Introduction
Aucun régime de fumage mécanique ne peut reproduire parfaitement le comportement des fumeurs:
— il est recommandé que les cigarettes soient aussi soumises à essai sous les conditions d’une différente
intensité de fumage mécanique que celles spécifiées dans le présent document;
— l’essai de fumage mécanique permet de caractériser les émissions des cigarettes pour des besoins
de conception et de réglementation, mais communiquer aux fumeurs les résultats de mesures sur
machines peut susciter des malentendus sur les niveaux d’exposition et de risque selon les marques;
— les données concernant l’émission de fumées obtenues par des mesurages mécaniques peuvent
être utilisées comme des données de base pour l’évaluation des dangers du produit, mais elles ne
constituent pas des mesures valables de l’exposition des individus et des risques et ce n’est pas
leur but. C’est faire un usage impropre des essais réalisés conformément aux Normes ISO que de
communiquer que les différences de mesures mécaniques entre différents produits correspondent
à des différences de niveaux d’exposition ou de risque.

1 Domaine d’application
Le présent document spécifie une méthode de dosage du benzo[a]pyrène (B[a]P) dans la matière
particulaire totale du courant principal de la fumée de cigarette par chromatographie en phase gazeuse
couplée à la spectrométrie de masse (CG/SM) en utilisant du cyclohexane comme solvant d’extraction.
Cette méthode a été validée en utilisant les paramètres de fumage de l’ISO 3308, et elle est techniquement
compatible avec d’autres régimes de fumage.
Le présent document définit une méthode alternative à l’ISO 22634-1, avec un nettoyage différent,
moins chronophage et avec une série d’analyses réduite, qui permet une augmentation potentielle de la
cadence des échantillons par comparaison à l’ISO 22634-1.
