ISO 9231|IDF 139:2008 specifies a method for the determination of the benzoic and sorbic acid contents in milk and milk products. The method is applicable to milk, dried milk, yogurt and other fermented milks, and cheese and processed cheese, and is suitable for measuring the contents of both compounds at levels of more than 5 mg/kg.

1 Область применения
Настоящий международный стандарт устанавливает метод определения содержания бензойной и
сорбиновой кислот в молоке и молочных продуктах.
Метод применим к молоку, сухому молоку йогурту и другим кисло-молочным продуктам, а также сыру и
плавленому сыру и пригоден для измерения содержаний обоих соединений при концентрациях свыше
5 мг/кг.

ПРИМЕЧАНИЕ Содержание бензойной и сорбиновой кислот выражают в миллиграммах на килограмм продукта.
4 Принцип
Удаляют жиры и белки из слабо щелочного раствора продукта путем осаждения по Каррезу. После
разбавления результирующего раствора метанолом фильтруют всплывающую жидкость. Разделяют
бензойную и сорбиновую кислоты с помощью высокоэффективной жидкостной хроматографии (HPLC)
на обращено-фазовой колонке C , измеряя оптическую плотность при 227 нм и 250 нм.

5 Реактивы
Если не оговорено иначе, используют только реактивы признанной чистоты, а также
дистиллированную или деминерализованную воду или воду эквивалентной чистоты.
5.1 Метанол (CH OH).

5.2 Реактивы для осаждения, как указано ниже:
5.2.1 Раствор гексацианоферроата(II) калия

в воде в мерной колбе с одной меткой вместимостью 100 мл (6.3). Разбавляют до метки водой и
перемешивают.
В том случае, если дигидрат ацетата цинка полностью не растворяется, нагревают при взбалтывании
колбу вместимостью 100 мл с содержимым в водяной бане (6.2), отрегулированной на температуру
70 °C. После полного растворения дигидрата ацетата цинка охлаждают полученный таким образом
раствор снова до комнатной температуры. Разбавляют до метки водой и опять перемешивают.
ПРИМЕЧАНИЕ 100 мл раствора достаточно для 40 опытов.
5.3 Фосфатный буферный раствор, pH 6,7.
Растворяют 2,5 г дигидроортофосфата калия (KH PO ) и 2,5 г тригидрата гидроортофосфата калия

(K HPO .3H O) в 1 л воды и перемешивают. Фильтруют полученный таким образом через систему

фильтрации растворителя (6.8).
5.4 Подвижная фаза, для HPLC.
Смешивают 10 объемов метанола (5.1) и 90 объемов фосфатного буферного раствора (5.3). Удаляют
любой растворенный газ с помощью небольшого вакуума.
5.5 Раствор гидроксида натрия, c(NaOH) = 0,1 моль/л.
Растворяют 4,0 г гранулированного гидроксида натрия в воде в мерной колбе с одной меткой
вместимостью 1 000 мл (6.3). Разбавляют до метки водой и перемешивают.
5.6 Серная кислота, c(H SO ) = 0,5 моль/л.

Осторожно приливают 15 мл концентрированной серной кислоты с массовой долей, по меньшей мере,
от 95 % до 98 % в 250 мл воды в мерной колбе с одной меткой вместимостью 500 мл (6.3) и оставляют
для охлаждения. Разбавляют до метки водой и перемешивают.
5.7 Стандартные растворы сорбиновой и бензойной кислот, приготовленные следующим

1 Scope
This International Standard specifies a method for the determination of the benzoic and sorbic acid contents in
milk and milk products.
The method is applicable to milk, dried milk, yogurt and other fermented milks, and cheese and processed
cheese, and is suitable for measuring the contents of both compounds at levels of more than 5 mg/kg.

3 Terms and definitions
For the purposes of this document, the following terms and definitions apply.
3.1
benzoic and sorbic acid contents
mass fractions of benzoic acid and sorbic acid determined by the procedure specified in this International
Standard
NOTE The benzoic acid and sorbic acid contents are expressed in milligrams per kilogram of product.
4 Principle
Fats and proteins are removed from a slightly alkaline solution of the product by Carrez precipitation.
Following dilution of the resultant solution with methanol, the supernatant liquid is filtered. The benzoic acid
and sorbic acid are separated by high-performance liquid chromatography (HPLC) on a reversed-phase C

column, measuring the absorbance at 227 nm and 250 nm.
5 Reagents
Unless otherwise specified, use only reagents of recognized analytical grade and distilled water or
demineralized water or water of equivalent purity.
5.1 Methanol (CH OH).

5.2 Precipitating reagents, as follows:
5.2.1 Potassium hexacyanoferrate(II) solution
Dissolve 10,6 g of potassium hexacyanoferrate(II) trihydrate (K [Fe(CN) ].3H O) in water in a 100 ml

one-mark volumetric flask (6.3). Dilute to the mark with water and mix.
NOTE The 100 ml solution is sufficient for 40 runs.
5.2.2 Zinc acetate solution
Dissolve 21,9 g of zinc acetate dihydrate [(CH COO) Zn.2H O] and 32 ml of acetic acid (CH COOH) in water

in a 100 ml one-mark volumetric flask (6.3). Dilute to the mark with water and mix.
If the zinc acetate dihydrate does not dissolve completely, heat the 100 ml flask and its contents in a water
bath (6.2) maintained at 70 °C while swirling. When the zinc acetate dihydrate has dissolved completely, cool
the solution thus obtained back to room temperature. Dilute to the mark with water and mix again.
NOTE The 100 ml solution is sufficient for 40 runs.
5.3 Phosphate buffer solution, pH 6,7.
Dissolve 2,5 g of potassium dihydrogen phosphate (KH PO ) and 2,5 g of potassium hydrogen phosphate

trihydrate (K HPO .3H O) in 1 l of water and mix. Filter the solution thus obtained through the solvent filtration

system (6.8).
5.4 Mobile phase, for HPLC.
Mix 10 volumes of methanol (5.1) with 90 volumes of phosphate buffer solution (5.3). Remove any dissolved
gas by applying a slight vacuum.
5.5 Sodium hydroxide solution, c(NaOH) = 0,1 mol/l.
Dissolve 4,0 g of sodium hydroxide pellets in water in a 1 000 ml one-mark volumetric flask (6.3). Dilute to the
mark with water and mix.
5.6 Sulfuric acid, c(H SO ) = 0,5 mol/l.

Pour cautiously 15 ml of concentrated sulfuric acid, with a mass fraction of at least 95 % to 98 %, into 250 ml
of water in a 500 ml one-mark volumetric flask (6.3) and all

Avant-propos
L'ISO (Organisation internationale de normalisation) est une fédération mondiale d'organismes nationaux
de normalisation (comités membres de l'ISO). L'élaboration des Normes internationales est en général
confiée aux comités techniques de l'ISO. Chaque comité membre intéressé par une étude a le droit de faire
partie du comité technique créé à cet effet. Les organisations internationales, gouvernementales et non
gouvernementales, en liaison avec l'ISO participent également aux travaux. L'ISO collabore étroitement avec
la Commission électrotechnique internationale (CEI) en ce qui concerne la normalisation électrotechnique.
Les Normes internationales sont rédigées conformément aux règles données dans les Directives ISO/CEI,
Partie 2.
La tâche principale des comités techniques est d'élaborer les Normes internationales. Les projets de Normes
internationales adoptés par les comités techniques sont soumis aux comités membres pour vote. Leur
publication comme Normes internationales requiert l'approbation de 75 % au moins des comités membres
votants.

laitier mondial. Les membres de la FIL se composent des Comités nationaux dans chaque pays membre et
des associations laitières régionales avec lesquelles la FIL a signé des accords de coopération. Tout membre
de la FIL a le droit de faire partie des Comités permanents de la FIL auxquels sont confiés les travaux
techniques. La FIL collabore avec l'ISO pour l'élaboration de méthodes normalisées d'analyse et
d'échantillonnage pour le lait et les produits laitiers.

Lait et produits laitiers — Détermination de la teneur en acide
benzoïque et en acide sorbique
1 Domaine d'application
La présente Norme internationale spécifie une méthode de détermination de la teneur en acide benzoïque et
en acide sorbique dans le lait et les produits laitiers.
Cette méthode est applicable au lait liquide, au lait sec, au yaourt et aux autres laits fermentés ainsi qu'aux
fromages et fromages fondus et convient pour la détermination de la teneur des deux composés à des
niveaux de concentration supérieurs à 5 mg/kg.

3 Termes et définitions
Pour les besoins du présent document, les termes et définitions suivants s'appliquent.
3.1

fractions massiques d'acide benzoïque et d'acide sorbique déterminées selon le mode opératoire spécifié
dans la présente Norme internationale
NOTE Les teneurs en acide benzoïque et en acide sorbique sont exprimées en milligrammes par kilogramme de
produit.
4 Principe
Des graisses et des protéines sont éliminées d'une solution légèrement alcaline du produit par la précipitation
de Carrez. Après dilution de la solution résultante avec du méthanol, le surnageant est filtré. L'acide
benzoïque et l'acide sorbique sont séparés par chromatographie liquide haute performance (CLHP) sur une
colonne C à phase inverse, mesurant l'absorbance à 227 nm et à 250 nm.

5 Réactifs
Sauf spécification contraire, utiliser uniquement des réactifs de qualité analytique reconnue et de l'eau distillée
ou déminéralisée ou de l'eau de pureté équivalente.
5.1 Méthanol (CH OH).

5.2 Réactifs de précipitation, comme suit:
5.2.1 Solution d'hexacyanoferrate(II) de potassium
Dissoudre dans de l'eau 10,6 g d'hexacyanoferrate(II) de potassium trihydraté (K [Fe(CN) ],3H O) dans une

fiole jaugée à un trait de 100 ml (6.3). Diluer au trait avec de l'eau et mélanger.
NOTE 100 ml de solution suffisent pour 40 essais.
5.2.2 Solution d'acétate de zinc
Dissoudre 21,9 g d'acétate de zinc dihydraté [(CH COO) Zn,2H O] et 32 ml d'acide acétique (CH COOH)

dans de l'eau, dans une fiole jaugée à un trait de 100 ml (6.3). Diluer au trait avec de l'eau et mélanger.
Si l'acétate de zinc dihydraté ne se dissout pas complètement, chauffer la fiole de 100 ml et son contenu dans
le bain d'eau (6.2) réglé à 70 °C en mélangeant par brassage. Après dissolution complète de l'acétate de zinc
dihydraté, refroidir la solution ainsi obtenue à la température ambiante. Diluer au trait avec de l'eau et
mélanger de nouveau.
NOTE 100 ml de solution suffisent pour 40 essais.
5.3 Solution tampon phosphate, pH 6,7.
Dissoudre, dans 1 l d'eau, 2,5 g de phosphate de potassium dihydrogéné (KH PO ) et 2,5 g de phosphate de

potassium monohydrogéné trihydraté (K HPO ,3H O) et mélanger. Filtrer la solution ainsi obtenue à l'aide du

système de filtration des solvants (6.8).
5.4 Phase mobile, pour CLHP.
Mélanger 10 volumes de méthanol (5.1) avec 90 volumes de solution tampon phosphate (5.3). Éliminer le gaz
dissous en créant un léger vide.
5.5 Solution d'hydroxyde de sodium, c(NaOH) = 0,1 mol/l.
Dissoudre 4,0 g de pastilles d'hydroxyde de sodium dans de l'eau, dans une fiole jaugée à un trait de 1 000 ml
(6.3). Diluer au trait avec de l'eau et mélanger.
5.6 Acide sulfurique, c(H SO ) = 0,5 mol/l.

Verser avec précaution 15 ml d'acide sulfurique concentré, de fraction massique d'au moins 95 % à 98 %,
dans 250 ml d'eau, dans une fiole jaugée à un trait de 500 ml (6.3), puis laisser refroidir. Diluer au trait avec
de l'eau et mélanger.
5.7 Solutions étalon d'acide sorbique et d'acide benzoïque, comme suit:

Dissoudre 50 mg d'acide sorbique et 50 mg d'acide benzoïque dans du méthanol (5.1) dans une fiole jaugée
