Soil Quality - Determination of dehydrogenase activity in soils - Part 2: Method using iodotetrazolium chloride (INT) (ISO 23753-2:2005)

ISO 23753-2:2005 specifies a method for determining soil dehydrogenase activity using 2-(4-iodophenyl)-3-(4-nitrophenyl)-5-phenyltetrazolium chloride (INT). As the INT reduction is less sensitive to O2, the method is more reproducible than the TTC-method described in ISO 23753-1. It is not applicable for determining the dehydrogenase activity in upper layers (L, F, H horizons) of forest humus forms with low microbial activity (e.g. mor), or in soils showing reducing properties (e.g. waterlogged soils).

Diese Internationale Norm legt ein Verfahren zur Bestimmung der Dehydrogenaseaktivität von Böden mit
2-(4-Iodophenyl-)-3(4-nitrophenyl-)-5-phenyltetrazoliumchlorid (INT) fest. Da die INT-Reduktion gegenüber O2
weniger empfindlich ist, ist das Verfahren reproduzierbarer als das in ISO 23753-1 beschriebene TTCVerfahren.
Diese Norm gilt nicht für die Bestimmung der Dehydrogenaseaktivität in Auflagehorizonten (L, F, H) von
Waldhumusformen mit geringer mikrobieller Aktivität (z. B. Moor) oder in Böden mit reduzierenden
Eigenschaften (z. B. staunasse Böden).

ISO 23753-2:2005 opredeljuje metodo za določevanje aktivnosti dehidrogenaze v tleh z uporabo 2-(4-jodofenil)-3-(4-nitrofenil)-5-feniltetrazolijevega klorida (INT). Ker je redukcija z INT manj občutljiva za O2, je metoda bolj ponovljiva kot metoda s TTC, opisana v ISO 23753-1. Ne velja za določevanje aktivnosti dehidrogenaze v zgornjih plasteh (horizontih L, F, H) gozdnih oblik humusa z nizko mikrobno aktivnostjo (npr. trhlini) ali v tleh z redukcijskimi lastnostmi (tleh, prepojenih z vodo).

The text of ISO 23753-2:2005 has been prepared by Technical Committee ISO/TC 190 “Soil quality” of the

International Organization for Standardization (ISO) and has been taken over as EN ISO 23753-2:2011 by

Technical Committee CEN/TC 345 “Characterization of soils” the secretariat of which is held by NEN.

This European Standard shall be given the status of a national standard, either by publication of an identical

text or by endorsement, at the latest by January 2012, and conflicting national standards shall be withdrawn at

the latest by January 2012.

Attention is drawn to the possibility that some of the elements of this document may be the subject of patent

rights. CEN [and/or CENELEC] shall not be held responsible for identifying any or all such patent rights.

Foreword

ISO (the International Organization for Standardization) is a worldwide federation of national standards bodies
