ISO 6488:2004 specifies a method for the determination of water content by the Karl Fischer method. It is applicable to raw tobacco as well as tobacco taken from finished products. The method is suitable for water contents ranging from a mass fraction of at least 2 % to 55 %.

Introduction
During the development of International Standards, inter-laboratory tests were carried out using two different
principles for the determination of water content of raw tobacco and tobacco taken from finished products.
These were
 this Karl Fischer procedure, and
 the gas chromatographic procedure.
These studies show that no differences occur between the results obtained by the two different methods. The
gas chromatographic method is described in ISO 16632.

1 Scope
This International Standard specifies a method for the determination of water content by the Karl Fischer
method. It is applicable to raw tobacco as well as tobacco taken from finished products. The method is
suitable for water contents ranging from a mass fraction of at least 2 % to 55 %.

3 Terms and definitions
For the purposes of this document, the following terms and definitions apply.
3.1
high-moisture tobacco
any tobacco sample containing volatile matter over 20 % as determined by drying at between 100 °C and
105 °C
4 Principle
The water content of a sample of tobacco or a tobacco product is determined by extraction of water by
shaking the sample with dry methanol, followed by injection of an aliquot portion into the titration vessel,
titration with pyridine-free Karl Fischer reagent and calculation of the water content. The method is applicable
to any type of tobacco sample provided that the particle size reduction is less than 4 mm.
NOTE If a size reduction (grinding or cutting) is applied, it can create a decrease in the original water content.
Cryogenic techniques may be used to prevent such moisture losses.

5 Reagents
Use only reagents of recognized analytical grade.
5.1 Karl Fischer reagent, free from pyridine, having a water equivalent per millilitre of reagent of
approximately 2 mg to 5 mg.
5.2 Methanol, with a water content of less than 0,05 g per 100 g.
This is hygroscopic so it is recommended to cap the bottle with an automatic delivery pipette equipped with
drying tube.
WARNING — Methanol is hazardous to human subjects and to the environment. Handle it with care.
5.3 Desiccant, silica gel, freshly activated.
5.4 Water, complying with grade 2 of ISO 3696, or better.
6 Apparatus
Usual laboratory apparatus and, in particular, the following items.
All glassware used in the preparation and in the water determination shall be heated at (105 ± 5) °C for at
least 1 h after visible water has evaporated. The glassware shall then be cooled and stored in a desiccator
over desiccant (5.3) until used.
6.1 Karl Fischer apparatus for automatic titration, consisting of items 6.1.1 to 6.1.5.
6.1.1 Apparatus for the detection of the titration endpoint, according to the biamperiometrical method.
6.1.2 Double electrode, made of platinum.
6.1.3 Magnetic stirrer.
6.1.4 Titration vessel.
6.1.5 Automatic burettes, for methanol and Karl Fischer reagent.

6.2 Mechanical shaker, rotary platform, adjustable to a shaking frequency of 155 min .
6.3 Microsyringe, for the determination of the water equivalent, of capacity 50 µl.
6.4 One-mark pipettes, of capacities 10 ml and 20 ml, complying with Class A of ISO 648.
6.5 Conical flasks, of capacities 250 ml and 500 ml, with a conical ground glass joint.
6.6 Hot-air oven, capable of maintaining a temperature of (105 ± 5) °C.
7 Standardization of the Karl Fischer reagent
Add sufficient methanol (5.2) to the titration vessel (6.1.4) to immerse the tips of the electrodes. Titrate any
residual solution (see Note) in the titration vessel to its endpoint by addition of Karl Fischer reagent (5.1).

Add 50 µl of water to the titration vessel using the microsyringe (6.3). To ensure that the syringe does not
contain air bubbles, fill it to above the 50 µl mark, invert it and tap the air bubbles to the top. Then depress the
plunger to the 50 µl mark and remove excess water quickly from the needle tip with a tissue. As an alternative,

Transfer the 50 µl of water to the titration vessel, taking care to inject the water directly into the solution, not
allowing any to fall onto the neck or walls of the vessel. Where the vessel can be fitted with a rubber
membrane cap, this shall be used and the needle inserted through the cap. If a water droplet remains on the
needle tip, remove it by touching the surface of the solution in the vessel.

réalisés selon deux principes différents pour la détermination de la teneur en eau du tabac brut et du tabac
prélevé dans des produits finis. Il s’agit
 de la présente méthode de Karl Fischer, et
 de la méthode par chromatographie en phase gazeuse.
Ces études ont montré qu’il ne se produit aucune différence entre les résultats obtenus selon les deux
méthodes différentes. La méthode par chromatographie en phase gazeuse est décrite dans l’ISO 16632.

4 Principe
La teneur en eau d’un échantillon de tabac ou d’un produit du tabac est déterminée par extraction de l’eau en
agitant l’échantillon avec du méthanol déshydraté, suivie de l’introduction d’une partie aliquote dans le vase
de titrage, d’un titrage avec le réactif de Karl Fischer exempt de pyridine, et du calcul de la teneur en eau. La
méthode est applicable à n’importe quel type d’échantillon de tabac, à condition que la taille des particules ait
été réduite à moins de 4 mm.
NOTE Si une réduction de la taille (par broyage ou découpage) est appliquée, elle peut entraîner une diminution de
la teneur initiale en eau. Des techniques cryogéniques peuvent être utilisées pour prévenir de telles pertes d’humidité.
5 Réactifs
Utiliser uniquement des réactifs de qualité analytique reconnue.
5.1 Réactif de Karl Fischer, exempt de pyridine, ayant un équivalent en eau d’environ 2 mg à 5 mg par
millilitre de réactif.
5.2 Méthanol, ayant une teneur en eau inférieure à 0,05 g par 100 g.
Le méthanol est hygroscopique, donc il est recommandé de boucher le flacon au moyen d’une pipette
d’alimentation automatique équipée d’un piège à eau.
AVERTISSEMENT — Le méthanol est dangereux pour l’homme et pour l’environnement. Le manipuler
avec précaution.
5.3 Déshydratant, gel de silice, récemment activé.
5.4 Eau, conforme à l’ISO 3696, grade 2 ou mieux.
6 Appareillage
Appareillage courant de laboratoire, et notamment les éléments suivants.
La totalité de la verrerie utilisée pour la préparation et pour le dosage de l’eau doit être chauffée à
(105 ± 5) °C pendant au moins 1 h après l’évaporation de toute trace d’eau visible. La verrerie doit ensuite
être refroidie et entreposée dans un dessiccateur au-dessus d’un déshydratant (5.3) jusqu’à utilisation.
6.1 Appareil de Karl Fischer pour titrage automatique, comprenant les éléments 6.1.1 à 6.1.5.
6.1.1 Appareil pour la détection du point d’équivalence de la réaction, selon la méthode
biampérométrique.
6.1.2 Double électrode, en platine.
6.1.3 Agitateur magnétique.
6.1.4 Vase de titrage.
6.1.5 Burettes automatiques, pour le méthanol et pour le réactif de Karl Fischer.

6.2 Agitateur mécanique, à plate-forme rotative, réglable à une fréquence d’agitation de 155 min .
6.3 Microseringue, pour la détermination de l’équivalent en eau, de 50 µl de capacité.
6.4 Pipettes à un trait, de 10 ml et de 20 ml de capacité, conformes à l’ISO 648, classe A.

6.5 Fioles coniques, de 250 ml et 500 ml de capacité, avec bouchon conique en verre rodé.
6.6 Étuve à air chaud, à même de maintenir une température de (105 ± 5) °C.
7 Étalonnage du réactif de Karl Fischer
Verser une quantité suffisante de méthanol (5.2) dans le vase de titrage (6.1.4) pour immerger les extrémités
des électrodes. Titrer toute solution résiduelle (voir la Note) dans le vase de titrage jusqu’au point
d’équivalence, par ajout de réactif de Karl Fischer (5.1).
Ajouter 50 µl d’eau dans le vase de titrage en utilisant la microseringue (6.3). Pour s’assurer que la seringue
ne contient pas de bulles d’air, la remplir au-delà du trait des 50 µl, la renverser, et tapoter pour faire remonter
les bulles vers l’extrémité supérieure. Appuyer ensuite sur le piston pour l’amener au trait des 50 µl, et retirer
rapidement l’excès d’eau de l’aiguille à l’aide d’un papier absorbant. Une autre solution consiste à remplir la
seringue jusqu’au repère des 50 µl et à peser la seringue. Après le dosage, peser à nouveau la seringue et
noter la masse exacte d’eau.
Transférer les 50 µl d’eau dans le vase de titrage, en prenant soin d’injecter l’eau directement dans la solution,
et en n’en laissant tomber ni sur le col, ni sur la paroi du vase. Quand le vase peut être équipé d’un capuchon
en caoutchouc, celui-ci doit être utilisé et l’aiguille introduite à travers le capuchon. Si une goutte d’eau
